Lactic acid bacteria (LAB) are widely used in the agro-food industry. Some of the LAB also participate in the natural flora in humans and animals. We review here proteomic studies concerning LAB. Two methods of research can be distinguished. In the first one, a systematic mapping of proteins is attempted, which will be useful for taxonomy and to function assignment of proteins. The second one focuses particularly on proteins whose synthesis is induced by various environmental situations or stresses. However, both approaches are complementary and will give new insights for the use of bacteria in industry, in human health and in the struggle against bacterial pathogens. Interest in LAB is growing, showing thus an increasing concern of their rational use and one can foresee in the near future an increasing use of proteomics as well as genomics. 相似文献

Bile salts are natural detergents that facilitate the digestion and absorption of the hydrophobic components of the diet. However, their amphiphilic nature makes them very inhibitory for bacteria and strongly influences bacterial survival in the gastrointestinal tract. Adaptation to and tolerance of bile stress is therefore crucial for the persistence of bacteria in the human colonic niche. Bifidobacterium animalis subsp. lactis, a probiotic bacterium with documented health benefits, is applied largely in fermented dairy products. In this study, the effect of bile salts on proteomes of B. animalis subsp. lactis IPLA 4549 and its bile-resistant derivative B. animalis subsp. lactis 4549dOx was analyzed, leading to the identification of proteins which may represent the targets of bile salt response and adaptation in B. animalis subsp. lactis. The comparison of the wild-type and the bile-resistant strain responses allowed us to hypothesize about the resistance mechanisms acquired by the derivative resistant strain and about the bile salt response in B. animalis subsp. lactis. In addition, significant differences in the levels of metabolic end products of the bifid shunt and in the redox status of the cells were also detected, which correlate with some differences observed between the proteomes. These results indicate that adaptation and response to bile in B. animalis subsp. lactis involve several physiological mechanisms that are jointly dedicated to reduce the deleterious impact of bile on the cell's physiology. 相似文献

This study gathers the first data concerning the arthropod assemblages associated with Erica arborea L. and E. scoparia L. It demonstrates that it is necessary to sample at least ten Erica scoparia L. to give a representative image of its arthropod assemblage. This study reveals that neither plant size nor architecture nor floristic environment can explain the different patterns of colonisation of both species of Erica by the arthropod assemblages. If our attention is focused on the three main species collected on both species of Erica, it appears that the nearly exclusive presence of Sitona lineatus L. on E. arborea L. might be correlated with highly different concentrations in tannins between the two species of Erica. In contrast, the different patterns of colonisation and the number of individuals of Colaspidea oblonga Blanchard collected on both species of Erica might be linked to the life-history of this Chrysomelideae. Finally, the presence of Psallus crotchi Scott to the same extent on both Erica species can be connected to the destruction of its natural host-plant and to the similar concentrations of nitrogen, a limiting factor for sap-sucker's colonisation, in both Erica species. 相似文献

Extracellular proteins of Bifidobacterium longum may mediate important interactions with the host. Here, we report on a comprehensive analysis of such proteins by using protein-free culture conditions and two-dimensional gel electrophoresis followed by mass spectrometry for protein identification. Seventeen proteins were detected in the culture supernatant, and 14 of them could be identified. Among these were 3 hypothetical solute-binding proteins of ABC transporters, an invasion-associated protein homolog, putative enzymes catalyzing cell wall turnover, several polypeptides with similarity to bacterial conjugation proteins, and 3 proteins of unknown function. Surprisingly, aldolase, usually considered as a cytoplasmic protein, was found in the culture supernatant. All proteins, excluding aldolase, were predicted to contain a signal peptide and a signal peptide cleavage site in their immature form. Some of the excreted proteins are interesting targets for further genetic and physiological studies. 相似文献

Despite the large number of studies devoted to the pituitary of teleosts, the origin of the direct pituitary innervation is still largely unknown. Although such a model is ideal for applying retrograde transport techniques, these methods involve the difficult in vivo injection of tracers into the pituitary and have never been applied. Recently, a lipophilic fluorescent dye (1-1-dioctadecyl-3,3,3,3-tetramethylindocarbocyanin; DiI) has been introduced and shown to have the capacity of being transported by the membranes of paraformaldehydefixed tissues. Microcrystals of DiI were implanted via a ventral approach into the pituitary of goldfish previously fixed by intracardiac perfusion of paraformaldehyde. The goldfish heads were kept immersed in paraformaldehyde for various periods of time (2–6 weeks). The brains were then dissected and cut transversally using a Vibratome. The results demonstrate that hypophysiotrophic areas are essentially restricted to the preoptic region, the mediobasal hypothalamus and the nucleus dorsolateralis thalami. In addition, cell bodies probably containing gonadotrophin releasing-hormone were also retrogradely stained along a pathway that can be traced up to the olfactory bulbs. The results also confirm that cell bodies, located around the ventral aspect of the preoptic recess and probably corresponding to dopaminergic neurons, project to the pituitary. Large neurons have also been observed in the rostral dorsal midbrain tegmentum just caudal to the posterior commissure. Most neurons of the so-called paraventricular organ remain unstained. Finally, a fiber tract originating from an undetermined territory of the posterior brain has been observed. The results are discussed in relation to the possible chemical nature of the hypophysiotropic neurons. 相似文献

In this paper, I have made a theoretical calculation of the Cave bear's bite force (BF) following the “dry skull method” and I present for the first time BF data that can be of interest to elucidate the mechanisms underlying the dietary choice of the Cave bears. In the skulls studied, males show higher BF than females in absolute terms, but more similar with regard to their body mass, which partly compensates for the smaller size of the females. The whole sample studied shows lower BF in the upper carnassial than those of large cats, similar to the one calculated for the Giant panda and higher than that of Polar bear. 相似文献

Summary Immunohistochemical localization of substance P (SP), CGRP, VIP, neuropeptide Y (NPY), and somatostatin (SOM) in the carotid labyrinth were compared in some species of amphibians using the peroxidase-antiperoxidase method. Immunoreactivity of SP, CGRP, VIP, and NPY was found in the nerve fibers distributed in the intervascular stroma of the carotid labyrinth. SP, CGRP, and VIP immunoreactive varicose fibers were densely distributed in the peripheral portion of the carotid labyrinth. Some SP-immunoreactive fibers were distributed similarly to CGRP-immunoreactive fibers. The density of NPY and SOM immunoreactive varicose fibers was low. No immunoreactivity of enkephalins was observed in the labyrinth. The intensities of these peptides were varied from species to species. No glomus cells showed immunoreactivity for any of the 7 peptides studied. These results suggest that the vascular regulatory function, which is one of the possible functions of the carotid labyrinth, is controlled by the peptidergic mechanisms in addition to regulation through intimate apposition of glomus and smooth muscle cells (g-s connection). 相似文献

Lactobacillus sakei is a food-borne bacterium naturally found in meat and fish products. A study was performed to examine the intraspecies diversity among 73 isolates sourced from laboratory collections in several different countries. Pulsed-field gel electrophoresis analysis demonstrated a 25% variation in genome size between isolates, ranging from 1,815 kb to 2,310 kb. The relatedness between isolates was then determined using a PCR-based method that detects the possession of 60 chromosomal genes belonging to the flexible gene pool. Ten different strain clusters were identified that had noticeable differences in their average genome size reflecting the natural population structure. The results show that many different genotypes may be isolated from similar types of meat products, suggesting a complex ecological habitat in which intraspecies diversity may be required for successful adaptation. Finally, proteomic analysis revealed a slight difference between the migration patterns of highly abundant GapA isoforms of the two prevailing L. sakei subspecies (sakei and carnosus). This analysis was used to affiliate the genotypic clusters with the corresponding subspecies. These findings reveal for the first time the extent of intraspecies genomic diversity in L. sakei. Consequently, identification of molecular subtypes may in the future prove valuable for a better understanding of microbial ecosystems in food products. 相似文献

A high intensity direct current was applied for more than 10 min onto a bacterial suspension of Pseudomonas oleovorans ATCC 29347 suspended in silicone oil. The application of a gradually increased electric field from 0 to 2500 V x cm(-1) resulted in a decrease of the optical density of the bacterial suspension and the occurrence of a peak current of several hundred microA for living cells instead of a linear increase (few microA) for killed or lyophilised cells. This procedure is not only a rapid way of investigating the living state of cell cultures but also an efficient experimental tool to study the cellular effects of a controlled electrical stress. 相似文献
