Effects of free long-chain fatty acids on thermophilic anaerobic digestion   总被引:3,自引:0,他引:3  
Summary Low concentrations of the long-chain fatty acids oleate and stearate inhibited all steps of the anaerobic thermophilic biogas process during digestion of cattle manure. The lag phase increased when the concentrations of oleate and stearate were 0.2 g/l and 0.5 g/l, respectively, and no growth was found at concentrations of 0.5 g/l for oleate and 1.0 g/l for stearate. The toxic effect of these acids was permanent as growth did not occur when inhibited cultures were diluted to a non-inhibitory concentration. No adaptation to the fatty acids toxicity was observed by pre-exposing the cultures to non-inhibitory concentrations and the inhibitory response was the same as for cultures not pre-exposed to the fatty acids. Oleate was less inhibitory when added as a neutral oil in the form of the glycerol ester. This indicates that it is the free fatty acid that influences the bacterial activity. Correspondence to: B. The influence of environmental parameters on the diversity of methanogenic communities in 15 full-scale biogas plants operating under different conditions with either manure or sludge as feedstock was studied. Fluorescence in situ hybridization was used to identify dominant methanogenic members of the Archaea in the reactor samples; enriched and pure cultures were used to support the in situ identification. Dominance could be identified by a positive response by more than 90% of the total members of the Archaea to a specific group- or order-level probe. There was a clear dichotomy between the manure digesters and the sludge digesters. The manure digesters contained high levels of ammonia and of volatile fatty acids (VFA) and were dominated by members of the Methanosarcinaceae, while the sludge digesters contained low levels of ammonia and of VFA and were dominated by members of the Methanosaetaceae. The methanogenic diversity was greater in reactors operating under mesophilic temperatures. The impact of the original inoculum used for the reactor start-up was also investigated by assessment of the present population in the reactor. The inoculum population appeared to have no influence on the eventual population.  相似文献

Biohydrogen could efficiently be produced in glucose-fed biofilm reactors filled with plastic carriers and operated at 70 degrees C. Batch experiments were, in addition, conducted to enrich and cultivate glucose-fed extreme-thermophilic hydrogen producing microorganisms from a biohydrogen CSTR reactor fed with household solid waste. Kinetic analysis of the biohydrogen enrichment cultures show that substrate (glucose) likely inhibited hydrogen production when its concentration was higher than 1 g/L. Different start up strategies were applied for biohydrogen production in biofilm reactors operated at 70 degrees C, and fed with synthetic medium with glucose as the only carbon and energy source. A biofilm reactor, started up with plastic carriers, that were previously inoculated with the enrichment cultures, resulted in higher hydrogen yield (2.21 mol H(2)/mol glucose consumed) but required longer start up time (1 month), while a biofilm reactor directly inoculated with the enrichment cultures reached stable state much faster (8 days) but with very low hydrogen yield (0.69 mol H(2)/mol glucose consumed). These results indicate that hydraulic pressure is necessary for successful immobilization of bacteria on carriers, while there is the risk of washing out specific high yielding bacteria.  相似文献

The newly isolated extreme thermophile Thermoanaerobacter pentosaceus was used for ethanol production from alkaline‐peroxide pretreated rapeseed straw (PRS). Both the liquid and solid fractions of PRS were used. T. pentosaceus was able to metabolize the typical process inhibitors present in lignocellulosic hydrolysate, namely 5‐hydroxymethyl furfural (HMF) and furfural, up to concentrations of 1 and 0.5 g L?1, respectively. Above these levels, xylose consumption was inhibited up to 70% (at 3.4 g‐furfural L?1) and 75% (at 3.4 g‐HMF L?1). T. pentosaceus was able to grow and produce ethanol directly from the liquid fraction of PRS, without any dilution or need for additives. However, when the hydrolysate was used undiluted the ethanol yield was only 37% compared to yield of the control, in which pure sugars in synthetic medium were used. The decrease of ethanol yield was attributed to the high amounts of salts resulting from the alkaline‐peroxide pretreatment. Finally, a two‐stage ethanol production process from PRS using Saccharomyces cerevisiae (utilization of hexoses in the first step) and T. pentosaceus (utilization of pentoses in the second step) was developed. Results showed that the two strains together could achieve up to 85% of the theoretical ethanol yield based on the sugar composition of the rapeseed straw, which was 14% and 50% higher compared to the yield with the yeast or the bacteria alone, respectively. The potential of wheat straw hydrolysate for biogas production was investigated in continuous stirred tank reactor (CSTR) and up-flow anaerobic sludge bed (UASB) reactors. The hydrolysate originated as a side stream from a pilot plant pretreating wheat straw hydrothermally (195 °C for 10–12 min) for producing 2nd generation bioethanol [Kaparaju, P., Serrano, M., Thomsen, A.B., Kongjan, P., Angelidaki, I., 2009. Bioethanol, biohydrogen and biogas production from wheat straw in a biorefinery concept. Bioresource Technology 100 (9), 2562–2568]. Results from batch assays showed that hydrolysate had a methane potential of 384 ml/g-volatile solids (VS)added. Process performance in CTSR and UASB reactors was investigated by varying hydrolysate concentration and/or organic loading rate (OLR). In CSTR, methane yields increased with increase in hydrolysate concentration and maximum yield of 297 ml/g-COD was obtained at an OLR of 1.9 g-COD/l d and 100% (v/v) hydrolysate. On the other hand, process performance and methane yields in UASB were affected by OLR and/or substrate concentration. Maximum methane yields of 267 ml/g-COD (COD removal of 72%) was obtained in UASB reactor when operated at an OLR of 2.8 g-COD/l d but with only 10% (v/v) hydrolysate. However, co-digestion of hydrolysate with pig manure (1:3 v/v ratio) improved the process performance and resulted in methane yield of 219 ml/g-COD (COD removal of 72%). Thus, anaerobic digestion of hydrolysate for biogas production was feasible in both CSTR and UASB reactor types. However, biogas process was affected by the reactor type and operating conditions.  相似文献

The energy efficiency of microwave-assisted dilute sulfuric acid pretreatment of rape straw for the production of ethanol was investigated. Different microwave energy inputs and solid loadings were tested to find economic pretreatment conditions. The lowest energy consumption was observed when solid loading and energy input were fixed at 50% (w/w) and 54 kJ (900 W for 1 min), respectively, and amounted to 5.5 and 10.9 kJ to produce 1 g of glucose after enzymatic hydrolysis and 1 g ethanol after fermentation, respectively. In general, 1 g ethanol can produce about 30 kJ of energy, and therefore, the energy input for the pretreatment was only 35% of the energy output. The approach developed in this study resulted in 92.9% higher energy savings for producing 1 g ethanol when compared with the results of microwave pretreatments previously reported.  相似文献

A submersible microbial fuel cell (SBMFC) was developed as a biosensor for in situ and real time monitoring of dissolved oxygen (DO) in environmental waters. Domestic wastewater was utilized as a sole fuel for powering the sensor. The sensor performance was firstly examined with tap water at varying DO levels. With an external resistance of 1000?, the current density produced by the sensor (5.6±0.5-462.2±0.5mA/m(2)) increased linearly with DO level up to 8.8±0.3mg/L (regression coefficient, R(2)=0.9912), while the maximum response time for each measurement was less than 4min. The current density showed different response to DO levels when different external resistances were applied, but a linear relationship was always observed. Investigation of the sensor performance at different substrate concentrations indicates that the organic matter contained in the domestic wastewater was sufficient to power the sensing activities. The sensor ability was further explored under different environmental conditions (e.g. pH, temperature, conductivity, and alternative electron acceptor), and the results indicated that a calibration would be required before field application. Lastly, the sensor was tested with different environmental waters and the results showed no significant difference (p>0.05) with that measured by DO meter. The simple, compact SBMFC sensor showed promising potential for direct, inexpensive and rapid DO monitoring in various environmental waters.  相似文献
