In vitro differentiation of rabbit blastocyst cells

Summary Six- and seven-day post-coitus (p.c.) rabbit embryos have been cultured in an attempt to establish a trophectodermal cell line. Results indicate that cells with epithelial characteristics (i.e. positive staining for cytokeratin) will survive in culture until Passage 3. At that time a fibroblastlike cell becomes predominant. In addition, we have found that the presence of the inner cell mass is required for embryo explants often results in the development of cells that spontaneously contract. These cells stain positively for myosin, which indicates that they may be precardiac cells. Maximum diastolic potential was −59±1.2 mV and the threshold potential was −53±2.3 mV. Spontaneously contracting cells did not respond to atropine, acetylcholine, epinephrine, isoproterenol, or propranolol. Action potential seems to be a result of an inward calcium current, because the beating rate is decreased in a dose-related manner with the calcium channel blocker verapamil, whereas the voltage-sensitive sodium channel blocker tetrodotoxin was without effect. Dissimilarity for partially ranked data and its application to cover-abundance data

Cover-abundance estimates are commonly employed in phytosociological investigations to record the performance of species. Because the coded values are on an ordinal scale of measure, various authors have suggested that some transformation is necessary before such values can be used for classification and ordination. However, it is not clear that transformation is a sufficient treatment, and it would seem preferable to use ordinal data directly. In this paper we examine such direct use of partial rankings and show that several dissimilarity measures can be defined for this case without invoking any transformations. They include dissimilarity measures associated with various rank correlation measures and with distances between strings; all the measure are variant forms of Hausdorf's interset distance. Certain other kinds of data, such as those employing dominant and subdominant species and the dry-weight-rank estimation of biomass, are also on an ordinal scale and could be analysed using similar techniques.To illustrate the approach, a string dissimilarity measure is used to analyse a set of data from Slovakian grasslands which appear to reflect a simple gradient. The original data were recorded with 10 classes of performance and are analysed using hierarchical and nondeterministic, overlapping, classifications.     

Somaclonal variation inLolium multiflorum L. andL. temulentum L.

Tiller-derived callus ofLolium multiflorum L. gave rise to 65 regenerants displaying variation in height, flowering time, chromosome number and alteration to the banding pattern of the isoenzyme superoxide dismutase. Plants were also regenerated from callus cultures initiated from immature embryos of the related inbreeding speciesLolium temulentum L. Polymorphic repetitive DNA sequences in Mycobacterium tuberculosis detected with a gene probe from a Mycobacterium fortuitum plasmid

Clinical isolates of Mycobacterium tuberculosis were shown by Southern blotting to contain DNA sequences hybridizing to a probe derived from a Mycobacterium fortuitum plasmid. Two such M. tuberculosis DNA fragments, isolated from a gene library, were used as probes to show restriction fragment length polymorphism in M. tuberculosis strains by detecting a repetitive sequence apparently located at different points on the chromosome. This could indicate the presence of a transposable element in M. tuberculosis which is partly homologous to a region of the M. fortuitum plasmid. The probes described can be used to fingerprint M. tuberculosis isolates, and in addition are capable of distinguishing M. tuberculosis from Mycobacterium bovis and BCG.     

The effects of phosphate supply on growth of plants from the Brasilian Cerrado: experiments with seedlings of the annual weed,Bidens gardneri Baker (Compositeae) and the tree,Qualea grandiflora (Mart.) (Vochysiaceae)

Summary Plants of the cerrado tree species Qualea grandiflora and the annual herb Bidens gardneri were grown from seed in controlled environment rooms at 30/20° C and 12 hour photoperiod. Seedlings were grown in pots or small tubes containing sand and provided with various amounts of mineral solutions based on the formulation of Hoagland and Arnon but with the phosphate content modified in some cases. In a long-term experiment lasting 213 days, plants supplied with full strength Hoagland's solution all died but plants of Qualea given 1/10 strength solution survived, although they grew very slowly. Low relative growth rates (0.008–0.036 d^–1) were also a feature of other experiments with Qualea and calculated rates of net assimilation rate gave values of 3–7 mg CO₂ dm^–2 h^–1. Expansion of the photosynthetic surface proceeded slowly and the cotyledons were the main site of photosynthesis for more than 40 days. The low rates of growth occurred despite significant uptake of phosphorus by young plants and in shortterm experiments growth was independent of the amount of phosphate supplied and accumulated. In contrast, the values of R found for plants of Bidens reached 0.24 d^–1. Growth of young plants was dependent on the external supply of phosphorus, being reduced when this was low and also when it was very high. Growth of the photosynthetic surface was also much more rapid than for Qualea and also varied with supply of phosphorus. The results are discussed in the context of the occurrence of these species in the Cerrado.     

Sex differences in the behaviour of immature captive lowland gorillas

Studies of the behaviour of 26 (12 males and 14 females) captive infant and juvenile lowland gorillas showed clear sex differences. Females showed greater interest in young infants and were more active in nest building as well as in solitary and social grooming. Males were more active in locomotive, dominance, and aggressive behaviour and in social play. Hand-rearing further increased aggression. Males were more aggressive when they lived with only one partner, and they rose in rank even above older females, a pattern that has not been observed in naturally reared gorillas.
