Rhodococcus equi is a facultative intracellular bacterium that can cause bronchopneumonia in foals and AIDS patients. Here, we have analyzed R. equi-containing vacuoles (RCVs) in murine macrophages by confocal laser scanning microscopy, by transmission electron microscopy and by immunochemistry upon purification. We show that RCVs progress normally through the early stages of phagosome maturation acquiring PI3P, early endosome antigen-1, and Rab5, and loosing all or much of them within minutes. Although mature RCVs possess the normally late endocytic markers, lysosome-associated membrane proteins, lysobisphosphatidic acid and Rab7, they lack other hallmark features of late endocytic organelles such as possession of cathepsin D, acid beta-glucuronidase, proton-pumping ATPase and the ability to fuse with prelabeled lysosomes. Bacterial strains possessing a virulence-associated plasmid maintain a nonacidified compartment for 48 h, whereas isogenic strains lacking such plasmids acidify progressively. In summary, RCVs represent a novel phagosome maturation stage positioned after completion of the early endosome stage and before reaching a fully mature late endosome compartment. In addition, vacuole biogenesis can be influenced by bacterial plasmids.  相似文献

Ethanol ingestion, like food ingestion, stimulates release of the signaling molecule cholecystokinin (CCK) from the small intestine. Here, we investigated the possibility that ethanol-induced CCK release might be a negative-feedback control of ethanol ingestion, similar to its function as part of the mechanism by which ingested food produces meal-ending satiation. We used Sardinian alcohol-preferring (sP) and Marchesian Sardinian (msP) alcohol-preferring rats, two apparently identical substrains that spontaneously ingest pharmacologically relevant amounts of ethanol, as well as their background strain, Wistar (W) rats. We demonstrated that: (1) intraperitoneal (IP), but not intracerebroventricular, injections of 0.5-4 microg/kg CCK-8 produced transient, dose-related reductions in 10% ethanol ingestion; (2) this inhibitory effect of CCK-8 on ethanol intake appeared behaviorally similar to its inhibitory action on ingestion of sucrose solutions; (3) the inhibitory effect of IP CCK-8 on ethanol ingestion occurred without evidence of tolerance when tests were repeated on consecutive days; (4) IP CCK-8 reduced ethanol intake despite simultaneously reducing blood ethanol levels (BALs); and (5) antagonism of CCK1 receptors with devazepide increased ethanol intake, indicating that endogenous CCK normally limits the size of bouts of ethanol ingestion. These results implicate peripheral CCK in the control of ethanol ingestion in sP and msP alcohol-preferring rats.  相似文献

The larval–pupal endoparasitoid Diachasmimorpha longicaudata (Hymenoptera: Braconidae) is currently the most commonly employed biological control agent against Tephritid fruit flies in the Americas. However, this parasitoid remains largely ignored and is not used in many regions, including the Mediterranean Basin. In this study, the potential of D. longicaudata as a biocontrol agent against the Mediterranean fruit fly (medfly) Ceratitis capitata (Diptera: Tephritidae) was addressed in an area of eastern Spain (the Valencian community). The parasitic activity of parasitoids and the effects of climatic conditions were evaluated throughout a 1‐year period in field‐cage experiments in which parasitoids were confined with apples artificially infested with medfly larvae. The following parameters were calculated and related statistically to several environmental conditions: the parasitism rate, the induced mortality and progeny sex ratio. The results show that D. longicaudata is able to parasitize medfly larvae throughout the year under semi‐natural conditions. Important fluctuations in the parasitism rate (from almost zero to 42%) and the induced mortality (from 6% to 80%) were partially influenced by climatic conditions. The parasitism rate increased with mean temperature and decreased with mean relative humidity, while the induced mortality decreased with minimum relative humidity. The optimal climatic conditions for the activity of the parasitoid were a mean temperature of 16–24°C combined with a relative humidity of 45%–60%. Overall, these results suggest that reduction in the medfly population due to D. longicaudata activity is feasible and provide information about the optimal time period for parasitoid release in the field. In conclusion, D. longicaudata has a significant potential to control C. capitata in the Mediterranean region.  相似文献

Niche width and inter‐individual diet variation in predator populations are known to be affected by intrinsic factors such as body size, age, cognitive constraints, and by intra‐ and interspecific competition. By contrast, how variation in prey biological traits may affect niche width and partitioning is still a poorly explored topic. One of these candidate traits is prey mobility, which can affect the predators’ niche because acting on the rate of encounter and, assuming mobility as a proxy for escape capability, on the success of predator attacks. Here we analysed 20 wasp populations and their prey as individual‐based food‐webs to test if prey mobility may explain niche width (here defined by the Shannon entropy‐based index ) and patterns of inter‐individual diet variation (here defined by the interaction‐exclusiveness index H2 and the interaction evenness index E2). Niche width was very variable among populations and overall network specialization (H2) was always higher than the expected by null models. In case of high‐speed flying prey (e.g. flies, bees), wasps showed wider niches () and lower specialization (H2) than in case of non‐flying or slow‐flying prey (e.g. spiders, beetles). Evenness (E2), on the other side, did not vary with prey mobility. Altogether, these results suggest that highly elusive prey may lead to wider predators’ niche but somehow limit their individual niche specialization.  相似文献

Objectives. To determine the antioxidant activities of nonsteroidal anti-inflammatory drugs (NSAIDS), we examined by chemiluminescence (CL) and electron spin resonance (ESR) their scavenging properties towards lipid peroxides, hypochlorous acid and peroxynitrite.

Methods. The antioxidant properties of nimesulide (NIM), 4-hydroxynimesulide (4-HONIM), aceclofenac (ACLO), 4-hydroxyaceclofenac (4-HOA-CLO), diclofenac (DICLO) and indomethacin (INDO) were tested on four different reactive oxygen species (ROS) generating systems: (I) phorbol-myristate acetate (PMA)-activated neutrophils, (II) Fe2+/ascorbate-induced lipid peroxidation, (III) HOCl-induced light emission, (IV) the kinetics of ONOO- decomposition followed by spectrophotometry. ROS production was monitored by luminol-enhanced CL or by ESR using two different spin traps.

Results. At 10 μM, ACLO, NIM, 4-HONIM, 4-HOA-CLO, and DICLO decreased luminol-enhanced CL generated by PMA-activated neutrophils. Inversely, INDO increased the luminol enhanced CL. Interestingly, hydroxylated metabolites were more potent antioxidants than the parent drugs. Furthermore, all drugs tested, excepted ACLO, lowered lipid peroxidation induced by Fe2+/ascorbate system. ACLO and DICLO, even at the highest concentration tested (100 μM), did not significantly lower HOCl induced CL, whereas the other drugs were potent scavengers. Finally, all the NSAIDS accelerated decomposition of ONOO-, suggesting a potential capacity of the molecules to scavenge peroxynitrite.

Conclusion. The NSAIDs possess variable degrees of antioxidant activities, linked to their ability to react with HOCl, lipid peroxides or ONOO-. These antioxidant activities could offer interesting targeted side-effects in the treatment of joint inflammatory diseases.  相似文献

2,5,7,8-tetramethyl-2-(2'-carboxyethyl)-6-hydroxychroman ( &#102 -CEHC) has been identified as a major water-soluble metabolite of vitamin E, which circulates in the blood and is excreted with the urine. The aim of this study was to assess the antioxidant activity of &#102 -CEHC using several methods with different prooxidant challenges. In the Oxygen Radical Absorbance Capacity assay, a fluorescent protein acts as a marker for oxidative damage induced by peroxyl radicals. In the Trolox Equivalent Antioxidant Capacity (TEAC) assay, a stable free radical, 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS &#148 + ) is reduced directly by antioxidants. Scavenging properties vs. reactive nitrogen species were studied measuring the effects on tyrosine nitration after reaction with peroxynitrite. Trolox, &#102 -tocopherol, ascorbic acid, and ( &#109 )-epicatechin were simultaneously tested in order to compare their antioxidant activities. In all mentioned systems, &#102 -CEHC exhibited antioxidant properties similar to those of Trolox. We conclude that &#102 -CEHC is a molecule with good antioxidant activity, having the advantage over Trolox of being a naturally occurring compound. These properties might be useful for research or industrial purposes.  相似文献

The importance of the spatial organisation of individuals in explaining species coexistence within a community is widely recognised. However, few analyses of spatial structure have been performed on tropical agroforests.The main objective of this study was to highlight the links between spatial organisation of shade trees on the one hand, and shade tree species richness and cacao yield on the other, using data from 29 cacao agroforests in Costa Rica.A method of spatial statistics, Ripley's K-function, was used to analyse the spatial organisation of shade and cacao trees in the study plots. For each stand, the X and Y coordinates of ≥2.5-m-tall trees were recorded. In each plot we also assessed shade tree species richness and cacao yield (with total number of pods = number of pods damaged by frosty pod rot + number of healthy pods).Three types of stands were identified: the first was characterised by significant clustering of shade trees, the highest shade tree species richness (S = 6), and the highest number of damaged pods (139 pods ha?1 year?1). The second type was characterised by random spatial organisation of shade trees. The third type showed a trend towards regular organisation. Species richness of shade trees did not differ significantly between the last two types (S = 4 for both), nor did the number of damaged pods (56 pods ha?1 year?1 and 67 pods ha?1 year?1 respectively).Although the trends were not statistically significant for all the variables in our data set, the clustered spatial structure appears to favour a synergy between environmental (tree species richness), and provisioning (cacao production) services.  相似文献

The present work aims at identifying new ion channel modulators able to target mitochondrial ATP-sensitive potassium channels (mitoKATP channels). An innovative approach should consist in fixing a cationic and hydrophobic triphenylphosphonium fragment on the structure of known KATP channel openers. Such phosphonium salts are expected to cross the biological membranes and to accumulate into mitochondria.Previous works revealed that the presence of an (R)-1-hydroxy-2-propylamino chain at the 3-position of 4H-1,2,4-benzothiadiazine 1,1-dioxides KATP channel openers increased, in most cases, the selectivity towards the pancreatic-type (SUR1/Kir6.2) KATP channel. In order to target cardiac mitoKATP channels, we decided to introduce a triphenylphosphonium group through an ester link on the SUR1-selective (R)-7-chloro-3-(1-hydroxy-2-propyl)amino-4H-1,2,4-benzothiadiazine 1,1-dioxide. The new compounds were found to preserve an inhibitory activity on insulin secretion (SUR1-type KATP channel openers) while no clear demonstration of an impact on mitochondria from cardiomyocytes (measurement of oxygen consumption, respiratory parameters and ATP production on H9C2 cells) was observed. However, the most active (inhibition of insulin release) compound 17 was found to penetrate the cardiac cells and to reach mitochondria.  相似文献

There is an impressing body of evidence supporting the beneficial role of balanced nutrition in lowering the risk of dementia and its commonest form, Alzheimer’s disease. Nevertheless, and despite worldwide dementia epidemic, there is much unfounded skepticism and lack of information among physicians. As a result, the diagnosis of cognitive impairment occurs still far too late, at best symptomatic drugs keep being prescribed and patients and caregivers are left with little concrete support in the hands of the natural history of the disease. This review summarizes knowledge about the impact of nutrition as part of a healthy lifestyle and of micronutrients in particular on delaying and avoiding dementia onset.  相似文献
