Prolyl endopeptidases (PEPs) have been found in numerous species. Inhibitors of human enzyme could correct cognitive deficits in Alzheimer patients while inhibition of Trypanosoma cruzi PEP could prevent invasion phase in Chagas disease. A structure-activity relationship study carried out in a tetrahydroisoquinoline series allowed to obtain potent competitive inhibitors superior to SUAM-1221. Besides, inhibitors expected to act according to an irreversible mechanism revealed to be superior to JPT-4819, for applications linked to human enzyme inhibition. 相似文献

Structural characterization of membrane proteins is hampered by the instability of the isolated proteins in detergent solutions. Here, we describe a new class of phospholipid-like surfactants that stabilize the G protein-coupled receptor, BLT1. These compounds, called C(13)U(9), C(13)U(19), C(15)U(25) and C(17)U(16), were synthesized by radical polymerization of Tris(hydroxymethyl) acrylamidomethane in the presence of thioglycerol, first endowed with two hydrocarbon chains with variable lengths (13-17 carbon atoms), as transfer reagent. C(13)U(19), C(17)U(16) or C(15)U(25) significantly enhanced the stability of BLT1 in solution compared to what was obtained with common detergents. These molecules therefore represent a promising step towards the structural characterization of BLT1 and possibly other membrane proteins. 相似文献

The fitness consequence of many behaviours of the small digger wasp, Cerceris rubida (Hymenoptera: Crabronidae), the only European species of its genus in which females share nests, are still unknown. Here, I present novel data on the nesting patterns and nest parasites of an Italian population of this wasp, with emphasis on which behavioural strategies may have evolved to reduce brood parasitism. Nests were established mainly in horizontal surfaces with scarce vegetation and hard soil, resulting in spatially clumped nests; the extent of nest aggregation increased over a 6-year period. Wasp brood cells are attacked by the miltogrammine fly Pterella grisea (Diptera: Sarcophagidae), which waits for nest-returning wasps on perching sites and then follows them in flight (satellite flight), eventually landing on the nest entrance and larvipositing without entering further in the tunnel. This technique seems to be adaptive for the parasitic flies, which would be rejected from nests by the guarding wasps if attempting to enter. The daily activity of the flies closely matched the host wasp’s provisioning activity, but C. rubida females were able to partially confound the tracking flies by performing evasive manoeuvres while returning to the nest. Patches with higher nest density and nests with more resident females did suffer more fly landings on nest entrances (a prerequisite for larviposition). These trends, however, disappeared on a per nest basis and on a per wasp provisioning flight basis, respectively. Across two years, only 6% of brood cells were parasitized, a picture similar to what observed for miltogrammine flies at nest aggregations of other Cerceris spp., and new data are necessary to test if there is a benefit of either nest density or nest sharing against P. grisea parasitism. 相似文献

The chemical composition of trunk bark oil from Cleistopholis patens (Benth .) Engl . & Diels , growing wild in Côte d'Ivoire, has been investigated by GC (FID) in combination with retention indices, GC/MS and 13C‐NMR. Moreover, one oil sample has been subjected to CC and all the fractions analyzed by GC (RI) and 13C‐NMR. In total, 61 components have been identified, including various sesquiterpene esters scarcely found in essential oils. 13C‐NMR was particularly efficient for the identification of a component not eluted on GC and for the quantification of heat‐sensitive compounds. Then, 36 oil samples, isolated from trunk bark harvested in six Ivoirian forests have been analyzed. The content of the main components varied drastically from sample to sample: (E)‐β‐caryophyllene (0.4 – 69.1%), β‐pinene (0 – 57%), α‐phellandrene (0 – 33.2%), α‐pinene (0.1 – 30.6%), β‐elemol (0.1 – 29.9%), germacrene D (0 – 25.4%), juvenile hormone III (0 – 22.9%), germacrene B (0 – 20.6%) and sabinene (tr‐20.3%). Statistical analysis, hierarchical clustering and principal components analysis, carried out on the 36 compositions evidenced a fair chemical variability of the stem bark oil of this species. Indeed, three clusters have been distinguished: the composition of group I (ten samples) was dominated by β‐pinene and α‐pinene, group II (nine samples) was represented by α‐phellandrene and p‐cymene and group III (16 samples) by β‐elemol. The selection of productive varieties of modern Criollo cocoa, showing fine aromatic qualities in their beans, is of major
interest for some producing countries, such as Venezuela. Cultivated populations of Modern Criollo or Trinitario varieties
may be suitable for admixture mapping analysis, as large blocks of alleles derived from two identified divergent ancestors,
recently admixed, are still preserved, after a few generations of recombination, similar to experimental mapping progenies.
Two hundred and fifty-seven individuals from a cultivated population of Modern Criollo were selected and analysed with 92
microsatellite markers distributed along the genome. This population exhibited a wide range of variability for yield factors
and morphological features. Population structure analysis identified two main subgroups corresponding to the admixture from
the two ancestors Criollo and Forastero. Several significant associations between markers and phenotypic data (yield factors
and morphological traits) were identified by a least squares general linear model (GLM) taking into account the population
structure and the percentage of admixture of each individual. Results were compared with classical QTL analyses previously
reported for other cacao populations. Most markers associated to quantitative traits were very close to QTLs detected formerly
for the same traits. Associations were also identified between markers and several qualitative traits including the red pigmentation

Rhodococcus equi is a facultative intracellular bacterium that can cause bronchopneumonia in foals and AIDS patients. Here, we have analyzed R. equi-containing vacuoles (RCVs) in murine macrophages by confocal laser scanning microscopy, by transmission electron microscopy and by immunochemistry upon purification. We show that RCVs progress normally through the early stages of phagosome maturation acquiring PI3P, early endosome antigen-1, and Rab5, and loosing all or much of them within minutes. Although mature RCVs possess the normally late endocytic markers, lysosome-associated membrane proteins, lysobisphosphatidic acid and Rab7, they lack other hallmark features of late endocytic organelles such as possession of cathepsin D, acid beta-glucuronidase, proton-pumping ATPase and the ability to fuse with prelabeled lysosomes. Bacterial strains possessing a virulence-associated plasmid maintain a nonacidified compartment for 48 h, whereas isogenic strains lacking such plasmids acidify progressively. In summary, RCVs represent a novel phagosome maturation stage positioned after completion of the early endosome stage and before reaching a fully mature late endosome compartment. 1. The effect of alkaline earth cations on the dissociation of the extracellular hemoglobin of Lumbricus terrestris and the effect of ionic strength on the dissociation of the hemoglobins of L. terrestris and Tubifex tubifex at concentrations of ca 2.5 mg/ml, over the pH range 9.0-10.5 was investigated using ultracentrifugation to separate the dissociated from the undissociated molecules. 2. Mg(II), Ca(II) and Sr(II) at concentrations of up to 0.2 M, decreased the dissociation of Lumbricus oxyhemoglobin from 70% at pH 9.0 and 100% at pH 9.5 and higher, to 20-30% at 0.05 M. The three cations were equally effective in decreasing the extent of dissociation of L. terrestris oxyhemoglobin over the pH range 9.0-10.5, with a K1/2 of ca 10 mM. 3. The dissociation of L. terrestris oxyhemoglobin over the pH range 9.0-10.5 was decreased only to 50-60% in the presence of up to 0.5 M NaCl or KCl; there was no further decrease in dissociation at concentrations of the two salts up to 1.5 M. 4. The dissociation of T. tubifex oxyhemoglobin over the pH range 9.0-10.0 was decreased from 100% to ca 40-50% in the presence of 0.5 M NaCl or KCl with little or no change at higher concentrations. At pH 10.5 and 11.0 the decrease in dissociation was more gradual, reaching ca 50% at 1.5 M NaCl. 相似文献
