Progress on reducing nutrient loss from annual croplands has been hampered by perceived conflicts between short‐term profitability and long‐term stewardship, but these may be overcome through strategic integration of perennial crops. Perennial biomass crops like switchgrass can mitigate nitrate‐nitrogen (NO3‐N) leaching, address bioenergy feedstock targets, and – as a lower‐cost management alternative to annual crops (i.e., corn, soybeans) – may also improve farm profitability. We analyzed publicly available environmental, agronomic, and economic data with two integrated models: a subfield agroecosystem management model, Landscape Environmental Assessment Framework (LEAF), and a process‐based biogeochemical model, DeNitrification‐DeComposition (DNDC). We constructed a factorial combination of profitability and NO3‐N leaching thresholds and simulated targeted switchgrass integration into corn/soybean cropland in the agricultural state of Iowa, USA. For each combination, we modeled (i) area converted to switchgrass, (ii) switchgrass biomass production, and (iii) NO3‐N leaching reduction. We spatially analyzed two scenarios: converting to switchgrass corn/soybean cropland losing >US$ 100 ha?1 and leaching >50 kg ha?1 (‘conservative’ scenario) or losing >US$ 0 ha?1 and leaching >20 kg ha?1 (‘nutrient reduction’ scenario). Compared to baseline, the ‘conservative’ scenario resulted in 12% of cropland converted to switchgrass, which produced 11 million Mg of biomass and reduced leached NO3‐N 18% statewide. The ‘nutrient reduction’ scenario converted 37% of cropland to switchgrass, producing 34 million Mg biomass and reducing leached NO3‐N 38% statewide. The opportunity to meet joint goals was greatest within watersheds with undulating topography and lower corn/soybean productivity. Our approach bridges the scales at which NO3‐N loss and profitability are usually considered, and is informed by both mechanistic and empirical understanding. Though approximated, our analysis supports development of farm‐level tools that can identify locations where both farm profitability and water quality improvement can be achieved through the strategic integration of perennial vegetation.  相似文献

Accelerometry is growing in popularity for remotely measuring fish swimming metrics, but appropriate sampling frequencies for accurately measuring these metrics are not well studied. This research examined the influence of sampling frequency (1–25 Hz) with tri‐axial accelerometer biologgers on estimates of overall dynamic body acceleration (ODBA), tail‐beat frequency, swimming speed and metabolic rate of bonefish Albula vulpes in a swim‐tunnel respirometer and free‐swimming in a wetland mesocosm. In the swim tunnel, sampling frequencies of ≥ 5 Hz were sufficient to establish strong relationships between ODBA, swimming speed and metabolic rate. However, in free‐swimming bonefish, estimates of metabolic rate were more variable below 10 Hz. Sampling frequencies should be at least twice the maximum tail‐beat frequency to estimate this metric effectively, which is generally higher than those required to estimate ODBA, swimming speed and metabolic rate. While optimal sampling frequency probably varies among species due to tail‐beat frequency and swimming style, this study provides a reference point with a medium body‐sized sub‐carangiform teleost fish, enabling researchers to measure these metrics effectively and maximize study duration.  相似文献

Re‐introduction is an important tool for recovering endangered species; however, the magnitude of genetic consequences for re‐introduced populations remains largely unknown, in particular the relative impacts of historical population bottlenecks compared to those induced by conservation management. We characterize 14 microsatellite loci developed for the Seychelles paradise flycatcher and use them to quantify temporal and spatial measures of genetic variation across a 134‐year time frame encompassing a historical bottleneck that reduced the species to ~28 individuals in the 1960s, through the initial stages of recovery and across a second contemporary conservation‐introduction‐induced bottleneck. We then evaluate the relative impacts of the two bottlenecks, and finally apply our findings to inform broader re‐introduction strategy. We find a temporal trend of significant decrease in standard measures of genetic diversity across the historical bottleneck, but only a nonsignificant downward trend in number of alleles across the contemporary bottleneck. However, accounting for the different timescales of the two bottlenecks (~40 historical generations versus <1 contemporary generation), the loss of genetic diversity per generation is greater across the contemporary bottleneck. Historically, the flycatcher population was genetically structured; however, extinction on four of five islands has resulted in a homogeneous contemporary population. We conclude that severe historical bottlenecks can leave a large footprint in terms of sheer quantity of genetic diversity lost. However, severely depleted genetic diversity does not render a species immune to further genetic erosion upon re‐introduction. In some cases, the loss of genetic diversity per generation can, initially at least, be greater across re‐introduction‐induced bottlenecks.  相似文献

Last-century climate change has led to variable increases of the intrinsic water-use efficiency (Wi; the ratio of net CO2 assimilation to stomatal conductance for water vapor) of trees and C3 grassland ecosystems, but the causes of the variability are not well understood. Here, we address putative drivers underlying variable Wi responses in a wide range of grassland communities. Wi was estimated from carbon isotope discrimination in archived herbage samples from 16 contrasting fertilizer treatments in the Park Grass Experiment, Rothamsted, England, for the 1915 to 1929 and 1995 to 2009 periods. Changes in Wi were analyzed in relation to nitrogen input, soil pH, species richness, and functional group composition. Treatments included liming as well as phosphorus and potassium additions with or without ammonium or nitrate fertilizer applications at three levels. Wi increased between 11% and 25% (P < 0.001) in the different treatments between the two periods. None of the fertilizers had a direct effect on the change of Wi (ΔWi). However, soil pH (P < 0.05), species richness (P < 0.01), and percentage grass content (P < 0.01) were significantly related to ΔWi. Grass-dominated, species-poor plots on acidic soils showed the largest ΔWi (+14.7 μmol mol−1). The ΔWi response of these acidic plots was probably related to drought effects resulting from aluminum toxicity on root growth. Our results from the Park Grass Experiment show that Wi in grassland communities consistently increased over a wide range of nutrient inputs, soil pH, and plant community compositions during the last century.The intrinsic water-use efficiency (Wi) of plants is controlled by photosynthetic carbon assimilation and stomatal conductance via the leaf-level coupling of CO2 and water fluxes. A general, but variable, increase of Wi under rising atmospheric CO2 has been observed in long-term studies (Peñuelas et al., 2011; Franks et al., 2013; Saurer et al., 2014), but little is known about other environmental or ecosystem factors, which may interact with the effect of increasing CO2 on Wi. An improved understanding of putative interactive mechanisms is important because changes in Wi may have significant effects on the global terrestrial carbon and water cycles (Gedney et al., 2006; Betts et al., 2007). This study explores the interactive effects of the increase in atmospheric CO2 (observed over the last century), nutrient loading, and soil pH together with other related effects on plant species richness and functional group composition on the coupling of plant CO2 and water fluxes in a seminatural grassland in southeastern England.Wi is a leaf-level efficiency that has also been termed potential water-use efficiency or physiological water-use efficiency, as it excludes the direct influence of vapor pressure deficit (VPD), a parameter determined by environmental conditions, on leaf-level water-use efficiency (Farquhar et al., 1989; Franks et al., 2013). Wi reports the relationship between net CO2 assimilation rate (An) and stomatal conductance for water vapor (gH2O):(1)According to the first law of Fick, An can be given as the product of the stomatal conductance for CO2 (gCO2) and the concentration gradient between the atmosphere (ca) and the leaf internal gas space (ci): An= gCO2 (caci). Using gCO2 (caci) instead of An in Equation 1, replacement of gH2O/gCO2 by the numerical value of gH2O/gCO2 (1.6) and rearrangement yields the following alternative expression of Wi:(2)Equation 2 reveals that past changes of Wi must have been controlled by two parameters: the change of ca and the concurrent change of 1 – ci/ca, the relative gradient for CO2 diffusion into the leaf (Franks et al., 2013). A change in the relative gradient is determined by the changes in An relative to gH2O, as leaves respond to changing ca and other environmental factors. In particular, Equation 2 shows that any variation in the climate change response of Wi is determined by the ci/ca response, if the comparison is made for vegetation at the same location and in the same period of time.Studies with C3 vegetation, including trees/forests and C3 grasslands, have revealed a general increase of Wi in the last century (Bert et al., 1997; Duquesnay et al., 1998; Feng, 1999; Arneth et al., 2002; Saurer et al., 2004; Barbosa et al., 2010; Köhler et al., 2010; Andreu-Hayles et al., 2011). In many cases, ci/ca, estimated by 13C discrimination (Farquhar et al., 1989), varied relatively little. Indeed, it has been suggested, based on theoretical grounds and empirical evidence from studies over geological/evolutionary to short time scales, that adaptive feedback responses will tend to maintain ci/ca approximately constant (Ehleringer and Cerling, 1995; Franks et al., 2013), as plants optimize carbon gain with respect to water loss (Cowan and Farquhar, 1977). Yet, ci/ca-dependent variation in the Wi response to climate change has also been noted (Peñuelas et al., 2011; Köhler et al., 2012) over the last century, indicating that additional factors, perhaps including other global change drivers, can modify the Wi response over this time scale, at least transiently. A meta-analysis by Peñuelas et al. (2011) reports ci/ca-dependent increases of Wi for different forests between 6% and 36% from the early 1960s to 2000s. A recent study by Saurer et al. (2014) on European forest trees found increases in Wi ranging from 1% to 53% during the last century. The strongest increase of Wi was recorded in regions where summer soil-water availability decreased in the last century. For different grassland communities, the ci/ca-dependent increases of Wi varied between 13% and 28% at one site (Köhler et al., 2012) from 1915 to 2009. Evidently, such variation can have important repercussions for the coupling of terrestrial CO2 and water fluxes. Yet, little is known about the mechanism(s) underlying the variation.At the Park Grass Experiment (PGE) at Rothamsted, England, Köhler et al. (2012) observed a nitrogen supply-dependent enhancement of the Wi response on plots receiving nitrate fertilizer and maintained at a near-neutral soil pH by liming. However, the actual relationship between nitrogen supply and Wi response did not hold when the unlimed control (soil pH approximately 5.2) was included in the comparison. Remarkably, however, there was a significant positive relationship between the grass content of the community and the Wi response of the experimental plots in the investigation. These results suggested that the effect of nutrient supply on the Wi response of the grassland communities was indirect, perhaps working via effects on soil pH and/or vegetation composition (plant species richness or functional group composition).The PGE provides a unique opportunity to study century-scale variation in the ci/ca-dependent variation of Wi for a wide range of diverse grassland communities. Much of the extant ecosystem-scale variability of plant species richness and soil pH in temperate grasslands of Europe (Ceulemans et al., 2014) is included in the range of plot-scale plant species richness and soil pH at the PGE (which is reported in this investigation). The different long-term applications of fertilizer and lime over the past century have resulted in substantial changes in soil pH, species richness, and grass content on the experimental plots, but in most cases, within-plot changes over the study period considered here (1915–2009) were comparatively small (Crawley et al., 2005; Silvertown et al., 2006). All experimental plots are located at the same site and are exposed to the same weather conditions. Consequently, trends in climate as a direct driver for differences in Wi between plots can be ruled out.Here, we explore putative mechanisms underlying eventual ci/ca-dependent variation of Wi during the last century at the PGE by, first, quantifying the sustained effect of a wide range of contrasting fertilizer treatments (n = 16) on the change of Wi during the last century and, second, analyzing the relationships between the observed Wi response of treatments and the respective nutrient status, soil pH, plant species richness, and plant functional group composition of the grassland communities.  相似文献

Summary: Intuitively, it may seem that from the perspective of an individual bacterium the ocean is a vast, dilute, and largely homogeneous environment. Microbial oceanographers have typically considered the ocean from this point of view. In reality, marine bacteria inhabit a chemical seascape that is highly heterogeneous down to the microscale, owing to ubiquitous nutrient patches, plumes, and gradients. Exudation and excretion of dissolved matter by larger organisms, lysis events, particles, animal surfaces, and fluxes from the sediment-water interface all contribute to create strong and pervasive heterogeneity, where chemotaxis may provide a significant fitness advantage to bacteria. The dynamic nature of the ocean imposes strong selective pressures on bacterial foraging strategies, and many marine bacteria indeed display adaptations that characterize their chemotactic motility as “high performance” compared to that of enteric model organisms. Fast swimming speeds, strongly directional responses, and effective turning and steering strategies ensure that marine bacteria can successfully use chemotaxis to very rapidly respond to chemical gradients in the ocean. These fast responses are advantageous in a broad range of ecological processes, including attaching to particles, exploiting particle plumes, retaining position close to phytoplankton cells, colonizing host animals, and hovering at a preferred height above the sediment-water interface. At larger scales, these responses can impact ocean biogeochemistry by increasing the rates of chemical transformation, influencing the flux of sinking material, and potentially altering the balance of biomass incorporation versus respiration. This review highlights the physical and ecological processes underpinning bacterial motility and chemotaxis in the ocean, describes the current state of knowledge of chemotaxis in marine bacteria, and summarizes our understanding of how these microscale dynamics scale up to affect ecosystem-scale processes in the sea.  相似文献

DNA binding of Klenow polymerase has been characterized with respect to temperature to delineate the thermodynamic driving forces involved in the interaction of this polymerase with primed-template DNA. The temperature dependence of the binding affinity exhibits distinct curvature, with tightest binding at 25-30 degrees C. Nonlinear temperature dependence indicates Klenow binds different primed-template constructs with large heat capacity (DeltaCp) values (-870 to -1220 cal/mole K) and thus exhibits large temperature dependent changes in enthalpy and entropy. Binding is entropy driven at lower temperatures and enthalpy driven at physiological temperatures. Large negative DeltaCp values have been proposed to be a 'signature' of site-specific DNA binding, but type I DNA polymerases do not exhibit significant DNA sequence specificity. We suggest that the binding of Klenow to a specific DNA structure, the primed-template junction, results in a correlated thermodynamic profile that mirrors what is commonly seen for DNA sequence-specific binding proteins. Klenow joins a small number of other DNA-sequence independent DNA binding proteins which exhibit unexpectedly large negative DeltaCp values. Spectroscopic measurements show small conformational rearrangements of both the DNA and Klenow upon binding, and small angle x-ray scattering shows a global induced fit conformational compaction of the protein upon binding. Calculations from both crystal structure and solution structural data indicate that Klenow DNA binding is an exception to the often observed correlation between DeltaCp and changes in accessible surface area. In the case of Klenow, surface area burial can account for only about half of the DeltaCp of binding.  相似文献

Two outlines for mixed model based approaches to quantitative trait locus (QTL) mapping in existing maize hybrid selection programs are presented: a restricted maximum likelihood (REML) and a Bayesian Markov Chain Monte Carlo (MCMC) approach. The methods use the in-silico-mapping procedure developed by Parisseaux and Bernardo (2004) as a starting point. The original single-point approach is extended to a multi-point approach that facilitates interval mapping procedures. For computational and conceptual reasons, we partition the full set of relationships from founders to parents of hybrids into two types of relations by defining so-called intermediate founders. QTL effects are defined in terms of those intermediate founders. Marker based identity by descent relationships between intermediate founders define structuring matrices for the QTL effects that change along the genome. The dimension of the vector of QTL effects is reduced by the fact that there are fewer intermediate founders than parents. Furthermore, additional reduction in the number of QTL effects follows from the identification of founder groups by various algorithms. As a result, we obtain a powerful mixed model based statistical framework to identify QTLs in genetic backgrounds relevant to the elite germplasm of a commercial breeding program. The identification of such QTLs will provide the foundation for effective marker assisted and genome wide selection strategies. Analyses of an example data set show that QTLs are primarily identified in different heterotic groups and point to complementation of additive QTL effects as an important factor in hybrid performance.  相似文献

Various types of post-translational modifications of the histone tails have been revealed, but a few modifications have been found within the histone core sequences. Histone core post-translational modifications have the potential to modulate nucleosome structure and DNA accessibility. Here, we studied the histone H2B core domain and found that phosphorylation of H2B serine 32 occurs in normal cycling and mitogen-stimulated cells. Notably, this phosphorylation is elevated in skin cancer cell lines and tissues compared with normal counterparts. The JB6 Cl41 mouse skin epidermal cell line is a well established model for tumor promoter-induced cell transformation and was used to study the function of H2B during EGF-induced carcinogenesis. Remarkably, cells overexpressing a nonphosphorylatable H2BS32A mutant exhibited suppressed growth and EGF-induced cell transformation, possibly because of decreased activation of activator protein-1, compared with control cells overexpressing wild type H2B. We identified ribosomal S6 kinase 2 (RSK2) as the kinase responsible for H2BS32 phosphorylation. Serum-starved JB6 cells contain very little endogenous H2BS32 phosphorylation, and EGF treatment induced this phosphorylation. The phosphorylation was attenuated in RSK2 knock-out MEFs and RSK2 knockdown JB6 cells. Taken together, our results demonstrate a novel role for H2B phosphorylation in cell transformation and show that H2BS32 phosphorylation is critical for controlling activator protein-1 activity, which is a major driver in cell transformation.  相似文献

Obesity is associated with the higher risk of breast cancer in postmenopausal women. The leptin signaling pathway is recognized to primarily regulate energy balance and associated with breast cancer. Furthermore, the estrogen signaling pathway plays a critical role in breast carcinogenesis. Due to its inherent asymmetry, competition for light is thought to cause loss of diversity from eutrophied systems. However, most of the work on this topic in grasslands has been phenomenological and has not measured light directly. We present the results of one of the few mechanistic experiments investigating the outcome of short-term competition using measurements of light interception from monocultures of five perennial grass species grown under fertilized and irrigated conditions. We found that the level of incident light intercepted by each species in monoculture, a direct measure of resource-reduction ability, was an excellent predictor of the relative competitive effect in pairwise mixtures. Competition for light was asymmetric in relation to differences in light intercepting ability. Our results are consistent with the idea that when light is a limiting resource, competition between species for this resource can be asymmetric, contributing to high dominance and low diversity.  相似文献
