油菜菌核病菌生理生态研究

通过试验分析,结果得出,油菜菌核病菌生长温度范围在0～34℃之间,23℃以下菌丝生长速度与温度呈正相关,25℃以上则呈负相关,23～25℃最适宜,菌丝和菌核致死温度分别为45℃和75℃．该菌对酸碱度适应范围很广,PH1～14范围内只在PH_1条件下不能生长,其最适PH值为5～6。病菌利用铵态氮源能力最强,硝态氮源次之,不能利用亚硝态氮;对氨基酸利用能力以精氨酸最强,其次是酪氨酸、天门冬氨酸和谷氨酸,赖氨酸最差。植株内精氨酸和赖氨酸比值大的白菜型油菜“黄鳝籽”病情扩展最快。

Productive infection of a cervical epithelial cell line with human immunodeficiency virus: implications for sexual transmission.

The human cervix-derived epithelial cell line (ME180) used in this study displays a characteristics epithelial morphology, including numerous desmosomes, tonofilaments, and epidermal filaments. When T-cell lines infected with human immunodeficiency virus (HIV) are added to epithelial cultures, they rapidly adhere to the epithelial monolayer. Within a few minutes, the T cells shed numerous virions into narrow spaces formed between the epithelial cell and the adherent T cells. Virions subsequently enter the ME180 cells via large vesicles. A few days after infection, cytopathic effects and syncytium formation were observed. Infected clones of ME180 cells have remained infected for 8 months. p24 enzyme-linked immunosorbent assay and infectivity assays show that one subclone of the cell line produces virus titers equivalent to those of high-secreting HIV-infected T-cell lines. Electron microscopy reveals numerous virions budding from both the basal and apical surfaces of the epithelium. These observations suggest that cervical epithelium has the potential to serve as a site of HIV infection.     

Degradation and debittering of a tryptic digest from beta-casein by aminopeptidase N from Lactococcus lactis subsp. cremoris Wg2.

The mode of action of purified aminopeptidase N from Lactococcus lactis subsp. cremoris Wg2 on a complex peptide mixture of a tryptic digest from bovine beta-casein was analyzed. The oligopeptides produced in the tryptic digest before and after aminopeptidase N treatment were identified by analysis of the N- and C-terminal amino acid sequences and amino acid compositions of the isolated peptides and by on-line liquid chromatography-mass spectrometry. Incubation of purified peptides with aminopeptidase N resulted in complete hydrolysis of many peptides, while others were only partially hydrolyzed or not hydrolyzed. The tryptic digest of beta-casein exhibits a strong bitter taste, which corresponds to the strong hydrophobicity of several peptides in the tryptic digest of beta-casein. The degradation of the "bitter" tryptic digest by aminopeptidase N resulted in a decrease of hydrophobic peptides and a drastic decrease of bitterness of the reaction mixture.     

Genetic studies of water buffalo blood markers. I. Red cell acid phosphatase,albumin, catalase,red cell α-esterase-3, group-specific component,and protease inhibitor

We have developed the methodologies for typing and family studies to establish the modes of inheritance of water buffalo red cell acid phosphatase (Acp), protease inhibitor (Pi), and group-specific component (Gc) on isoelectric focusing and albumin (Alb), red cell -esterase-3 (Est-3), and catalase (Cat) on polyacrylamide gel electrophoresis. Family studies showed that Pi, Gc, Alb, and Cat are coded by autosomal genes with two codominant alleles, while Est-3 is autosomal with two codominant alleles and a recessive null allele and Acp exhibits three codominant alleles.This project was funded by the Australian Centre for International Agricultural Research through Grant PN 8364 and the Malaysian programme for Intensification of Research in Priority Areas through Grant IRPA 1-07-05-057.     

激光辐照选育水稻新品系的过程及酯酶同工酶的分析

激光辐照选育的435与1002早籼新晶系比湘早籼7号早热,高产,米质好。醋酶同工酶多2—3条带,酶活性增强,增产潜力大。     

G418对IL—2基因包装细胞的筛选

本文用高浓度的G418(800μg/ml)和低浓度的G418(200μg/ml)对包装细胞PLXSN/IL-2/PA317细胞进行40天的选择筛选培养,使其细胞呈稳定状态生长时,收集上清液转染NIH/3T3细胞,进行病毒滴度测定。试图在高选择标记的情况下筛选出高表达目的基因的包装细胞。实验结果表明:高、低浓度的G418对PLXSN/IL-2/PA317包装细胞的选择作用相同,即包装细胞的病毒滴度同选择标记物浓度无关。提示可用低浓度的G418来维持包装细胞的生命。
