Globally, soil CO2 efflux rates (Fs) have been linked to changes in soil water content (SWC), rainfall and temperature and/or productivity. However, within an ecosystem, Fs can vary based on site structure and function, which can be affected by a combination of abiotic and biotic factors. This becomes particularly important when an ecosystem is faced with disturbances, such as drought or fire. Site-specific compensatory responses to disturbances may therefore alter C mineralization, as well as root respiration. Hence, single location Fs estimates may not be a representative for ecosystems across their distributional ranges. We conducted a 6-year study along an edaphic moisture gradient of longleaf pine ecosystems that were maintained with prescribed fire, using eddy covariance and soil respiration measurements to address how Fs varies with changes in ecosystem structure and function, as well as disturbances. Lower air temperatures (Tair) decreased Fs at all sites, but that response was also affected by productivity and SWC. Productivity significantly altered Fs rates at all sites, especially when we accounted for changes in temperature and SWC. Plant regrowth post-fire temporarily increased Fs (10–40%), whereas drought reduced Fs at all sites. Our results show that site productivity, Fs and the degree to which ecosystems adapt to climate variations and disturbance can be site specific. Hence, model forecasting of carbon dynamics would strongly benefit from multi-location measurements of Fs across the distributional range of an ecosystem. 相似文献

AbstractThe reproductive cycle and growth of the pen shell, Atrina maura, which was cultured in the Ensenada Pabellones lagoon system, Gulf of California, from March 2008 to March 2009, is described in this article. Histological techniques and the condition index were used to determine its reproductive condition. The sex ratio was 0.57 females:1.72 males within the population studied. There were no differences (χ2, p < 0.05) in shell length (SL) between males and females. The mean length of the sampled specimens ranged between 50.99 ± 4.86 mm and 218.16 ± 8.87 mm. The histological results confirmed that A. maura is a gonochoristic organism that presents synchronic development of the gonads. The maturity and spawning phases were observed throughout the study period, with the exception of March and May 2008. The frequency of the gonad development stages obtained per month suggests that this species reproduced two times annually, with one important reproductive period from June to September, a minor reproductive period from November to February, and two resting periods as follows: July to August and January to February 2009. 相似文献

A malaria vaccine is a public health priority. In order to produce an effective vaccine, a multistage approach targeting both the blood and the liver stage infection is desirable. The vaccine candidates also need to induce balanced immune responses including antibodies, CD4+ and CD8+ T cells. Protein-based subunit vaccines like RTS,S are able to induce strong antibody response but poor cellular reactivity. Adenoviral vectors have been effective inducing protective CD8+ T cell responses in several models including malaria; nonetheless this vaccine platform exhibits a limited induction of humoral immune responses. Two approaches have been used to improve the humoral immunogenicity of recombinant adenovirus vectors, the use of heterologous prime-boost regimens with recombinant proteins or the genetic modification of the hypervariable regions (HVR) of the capsid protein hexon to express B cell epitopes of interest. In this study, we describe the development of capsid modified Ad5 vectors that express a promiscuous Plasmodium yoelii T helper epitope denominated PyT53 within the hexon HVR2 region. Several regimens were tested in mice to determine the relevance of the hexon modification in enhancing protective immune responses induced by the previously described protein-based multi-stage experimental vaccine PyCMP. A heterologous prime-boost immunization regime that combines a hexon modified vector with transgenic expression of PyCMP followed by protein immunizations resulted in the induction of robust antibody and cellular immune responses in comparison to a similar regimen that includes a vector with unmodified hexon. These differences in immunogenicity translated into a better protective efficacy against both the hepatic and red blood cell stages of P. yoelii. To our knowledge, this is the first time that a hexon modification is used to deliver a promiscuous T cell epitope. Cold-water coral ecosystems are characterised by a high diversity and population density. Living and dead foraminiferal assemblages
from 20 surface sediment samples from Galway and Propeller Mounds were analysed to describe the distribution patterns of benthic
foraminifera on coral mounds in relation to different sedimentary facies. Hard substrates were examined to assess the foraminiferal
microhabitats and diversities in the coral framework. We recognised 131 different species, of which 27 prefer an attached
lifestyle. Epibenthic species are the main constituents of the living and dead foraminiferal assemblages. The frequent species
Discanomalina coronata was associated with coral rubble, Cibicides refulgens showed preference to the off-mound sand veneer, and Uvigerina mediterranea displayed abundance maxima in the main depositional area on the southern flank of Galway Mound, and in the muds around Propeller
Mound. The distribution of these species is rather governed by their specific ecological demands and microhabitat availability
than by the sedimentary facies. Benthic foraminiferal assemblages from coral mounds fit well into basin-wide-scale distribution
patterns of species along the western European continental margin. The diversity of the foraminiferal faunas is not higher
on the carbonate mounds as in their vicinity. The living assemblages show a broad mid-slope diversity maximum between 500
and 1,300 m water depth, which is the depth interval of coral mound formation at the Celtic and Amorican Margin. The foraminiferal
diversity maximum is about 700 m shallower than comparable maxima of nematodes and bivalves. This suggests that different

Interest in airborne smallpox transmission has been renewed because of concerns regarding the potential use of smallpox virus as a biothreat agent. Air disinfection via upper-room 254-nm germicidal UV (UVC) light in public buildings may reduce the impact of primary agent releases, prevent secondary airborne transmission, and be effective prior to the time when public health authorities are aware of a smallpox outbreak. We characterized the susceptibility of vaccinia virus aerosols, as a surrogate for smallpox, to UVC light by using a benchtop, one-pass aerosol chamber. We evaluated virus susceptibility to UVC doses ranging from 0.1 to 3.2 J/m2, three relative humidity (RH) levels (20%, 60%, and 80%), and suspensions of virus in either water or synthetic respiratory fluid. Dose-response plots show that vaccinia virus susceptibility increased with decreasing RH. These plots also show a significant nonlinear component and a poor fit when using a first-order decay model but show a reasonable fit when we assume that virus susceptibility follows a log-normal distribution. The overall effects of RH (P < 0.0001) and the suspending medium (P = 0.014) were statistically significant. When controlling for the suspending medium, the RH remained a significant factor (P < 0.0001) and the effect of the suspending medium was significant overall (P < 0.0001) after controlling for RH. Virus susceptibility did not appear to be a function of virus particle size. This work provides an essential scientific basis for the design of effective upper-room UVC installations for the prevention of airborne infection transmission of smallpox virus by characterizing the susceptibility of an important orthopoxvirus to UVC exposure. 相似文献

Two experiments were done to develop an effective superovulatory treatment protocol in wood bison for the purpose of embryo collection and transfer. In experiment 1, donor bison were assigned randomly to four treatment groups (N = 5 per group) to examine the effects of method of synchronization (follicular ablation vs. estradiol-progesterone treatment) and ovarian follicular superstimulation (single slow-release vs. split dose of FSH). Recipient bison were synchronized with donor bison by either follicular ablation (N = 8) or estradiol-progesterone treatment (N = 9). In experiment 2, bison were assigned randomly to four treatment groups (N = 5 per group) to examine the ovarian response to two versus four doses of FSH, and the effect of progesterone (ovarian superstimulation with or without an intravaginal progesterone-releasing device). Donor bison were inseminated with fresh chilled wood bison semen 12 and 24 hours after treatment with GnRH (experiment 1) or LH (experiment 2). The ovarian response was assessed using ultrasonography. In experiment 1, the number of large follicles (≥7 mm) increased in response to both FSH treatments, but the diameter of the largest follicle detected 4 and 5 days after the start of ovarian superstimulation was greater in bison treated with a single dose of FSH than in those treated with two doses (P < 0.05). A total of 10 ova and/or embryos were collected. One blastocyst was transferred to each of five recipient bison resulting in the birth of two live wood bison calves. In experiment 2, two doses of FSH resulted in a greater number of large follicles (≥9 mm) on Days 4, 5, and 6 (P < 0.05) after beginning of superstimulation (Day 0), and more ovulations than four doses of FSH (11.2 ± 2.4 vs. 6.4 ± 0.8; P < 0.05). Embryo collection was performed on only five donors, and a total of 19 ova and/or embryos were recovered. In summary, fewer FSH treatments were as good or better than multiple treatments, consistent with the notion that minimizing handling stress improves the superovulatory response in bison. Follicular ablation and estradiol plus progesterone treatment were effective for inducing ovarian synchronization in embryo donor and recipient bison, and an intravaginal progesterone-releasing device during superstimulatory treatment did not influence the superovulatory response or embryo collection. Delaying ovulation-inducing treatment (GnRH or LH) to 5 days after superstimulatory treatment resulted in a greater number of ovulations and improved embryo collection efficiency (experiment 2). Methanol limited fed-batch cultivation was applied for production of a plant derived beta-glucosidase by Pichia pastoris. The beta-glucosidase was recovered by expanded bed adsorption chromatography applied to the whole culture broth. The new Streamline Direct HST1 adsorbent was compared with Streamline SP. Higher bead density made it possible to operate at two times higher feedstock concentration and at two times higher flow velocity. The higher binding capacity in the conductivity range 0-48 mS cm(-1) of Streamline Direct HST1 might be caused by the more complex interaction of multi-modal ligand in Streamline Direct HST1 compared to the single sulphonyl group in Streamline SP. Harsher elution condition had to be applied for dissociation of beta-glucosidase from Streamline Direct HST1 due to stronger binding interaction. The 5% dynamic binding capacity was 160 times higher for Streamline Direct HST1 compared to Streamline SP. The yield of beta-glucosidase on Streamline Direct HST1 (74%) was significantly higher than on Streamline SP (48%). Furthermore, beta-glucosidase was purified with a factor of 4.1 and concentrated with a factor of 17 on Streamline Direct HST1 while corresponding parameters were half of these values for Streamline SP. Thus, for all investigated parameters Streamline Direct HST1 was a more suitable adsorbent for recovery of recombinant beta-glucosidase from unclarified P. pastoris high-cell-density cultivation broth. 相似文献
