In this preliminary study defense behaviour patterns (fear responses) are described in two closely related, behaviourally different inbred labyrinth fish subspecies and in their F1 generation. The subspecies M. opercularis (characterized briefly by “active escape”) and M. opercularis concolor (characterized by “passive escape”) showed specific differences in the manifestation of certain defense behaviour patterns. In the F1 hybrid generation dominance and overdominance of M. opercularis was found in most defense behaviour patterns. Analysing the frequencies and sequences of movement patterns it could be shown that defensive behaviour is not a random or entirely “plastic” process but that there is sequential linkage between the patterns and they form characteristic clusters. Our results suggest that manifestations of different patterns are under genetic control and presumably, genetic determination of certain patterns is not very complex. Attempts were made to determine whole brain noradrenaline, serotonine and dopamine levels of the two subspecies and a significant difference was found in the noradrenaline content.  相似文献

The light and dark forms of phosphoenolpyruvate (PEP) carboxylase(PEPC) from the dicot plant Amaranthus viridis L. were purifiedand their kinetic properties were studied in water-based orbinary alcohol-water solvents. At pH 7.3, the specific activityof the purified light form was about 2.7-fold higher than thatpresented by the dark form of PEPC under optimal conditions,while Km remained virtually unchanged in both forms. The enzyme'slight form was better activated by glucose 6-phosphate and lessinhibited by L-malate than the dark PEPC. From the organic solventsstudied, methanol showed the most important effect, enhancingPEPC activity by two-fold at 20% (v/v). Ethanol, ethylene glycol,tert-butanol and 2-propanol were also activators to a lesserdegree, but at high concentrations (typically greater than 20%,v/v) the effect was reduced or turned to inhibition. Km (PEP)was reduced by an order of magnitude in the presence of 20%(v/v) methanol (i.e. from 0.32 to 0.022 mM for the light formof the enzyme). The inhibitory effect of malate at low PEP waslessened by methanol for both forms (i.e. I50 0.25 mM in aqueousmedium to 0.48 mM in binary mixture for the dark form), whileglucose-6-P activation of PEPC was not affected by methanol.The results suggest that the kinetics of PEPC in a medium thatmimics more closely in vivo conditions are different from thoseobserved by standard procedures consisting of aqueous media,and provide a new insight on the properties of PEPC as relatedto its regulation in vivo. 1. Chemical modification by o-iodosobenzoate of soluble chloroplast coupling factor 1 (CF1) during heat activation resulted in inhibition of its Ca-ATPase activity and in the formation of two new intrapeptide disulfide bridges as suggested by: (a) the disappearance of three out of four accessible thiol groups, two from gamma and one from a beta subunit as a consequence of CF1 modification by o-iodosobenzoate; (b) the total free sulphydryl groups of CF1 were reduced from 8 to 4 after modification of CF1 by o-iodosobenzoate. Two groups disappeared from beta and two from gamma subunits; (c) a second heating step of CF1 in the presence of 10 mM dithioerythritol reversed the inhibition of the ATPase and reduced both the newly formed disulfide bridges and those present in native CF1. 2. Modification of chloroplasts in the light with o-iodosobenzoate resulted in the inhibition of photophosphorylation and ATPase. CF1 isolated and purified from these chloroplasts had its Ca-ATPase activity inhibited and two new disulfide bridges. The total number of free sulphydryl groups was reduced from 8 to 4 and three accessible groups disappeared from beta and gamma subunits.  相似文献

Phosphoenolpyruvate carboxylase from Amaranthus viridis leaves was activated by inorganic orthophosphate in a concentration- and pH-dependent manner. Maximal activation at pH 7.0 was achieved at phosphate concentrations above 20 mM, and a positive cooperativity was observed for the binding of the anion at this pH. At pH 8.0 the maximum of activity was achieved at 10 mM phosphate; higher concentrations reduced the activation. KM for phosphoenolpyruvate-Mg at pH 7.0 was lowered by phosphate in all concentrations tested up to 30 mM. While at pH 8.0 the KM values were lower than that of the control up to 10 mM phosphate; higher anion concentrations raised the minimum value of KM at this pH. VMAX increased at pH 7.0, and remained unchanged at pH 8.0. A KA value of 0.41 mM was calculated for phosphate at the alkaline pH. The phosphate analogue arsenate also behaved as an activating agent, while other anions (e.g. nitrate, nitrite, sulfate, tetraborate) were ineffective. The phosphate-activated enzyme was shown to be insensitive to glucose-6-phosphate, but was inhibited by l -malate to the same extent as the control.  相似文献

The water quality of the Siluko River, Edo State, Nigeria was investigated from March to August 2015 to determine its suitability for drinking and usage for domestic purposes. Water samples collected from three stations were tested for thirteen physico-chemical parameters using standard analytical procedures. Temperature, phosphate and chloride were significantly different across the three stations. All other parameters, with the exception of turbidity, dissolved oxygen and phosphate, were within the permissible limits recommended by the Nigerian Standard for Drinking Water Quality (NSDWQ) and World Health Organization (WHO). Water Quality Index (WQI) values ranged from 11.24 to 16.15, indicating excellent water quality. While the quality of the water from the Siluko River is suitable for drinking and domestic usage, to prevent future deterioration of the water, it is recommended that the regulating authorities monitor effluents discharged into the river from human activities.  相似文献
