In total 73 species of Euglena were ordered according to their saprobic index (S) and saprobic valency starting with species tolerating a heavy organic pollution and ending with those from most clean water. Adiaspiromycosis was identified in 6 animals in the examination of the lungs of 90 large carnivores. Emmonsia crescens (Chrysosporium parvum var. crescens) was demonstrated as the causative agent in 5 cases of disease — in the badger (Meles meles), the otter (Lutra lutra) and the fox (Vulpes vulpes). E. parva was demonstrated in the remaining case of disease in a fox. The badger is a new, up to the present unknown host of E. crescens. The sporadic occurrence of adiaspiromycosis in the fox and the otter classifies this disease among rare diseases of these animals.  相似文献

The effect of mersalyl, an inhibitor of phosphate transport across the inner mitochondrial membrane, was investigated on the uncoupled respiration of pig kidney mitochondria in the presence of glutamine as substrate and on the activity of the phosphate-dependent glutaminase in the intact organelles. In addition, the submitochondrial location of the enzyme was reinvestigated.

1. (1) It was found that mersalyl completely inhibits uncoupled respiration of the mitochondria in the presence of glutamine as substrate, whereas respiration with glutamate was not affected. The same amount of mersalyl which inhibits coupled oxidation of glutamine also inhibits coupled oxidation of glutamate and some other substrates.

2. (2) Mersalyl strongly inhibited the activation of glutaminase in intact mitochondria only in the presence of inhibitors of electron transport or of an uncoupler. The addition of a detergent prevented or fully released the inhibition. The effect of mersalyl was observed even when the mitochondria were pre-incubated with phosphate or incubated in the phosphate-free medium. If mersalyl and carbonyl cyanide m-chlorophenylhydrazone (CCCP) were added 3 min after pre-incubation with phosphate the same intramitochondrial concentration of the anion as in control experiments was found, whereas the activity of glutaminase was severely inhibited. These findings suggest that the activation of the enzyme by phosphate in intact nonenergized mitochondria occurs only if the activator moves across the inner mitochondrial membrane.

3. (3) Mersalyl (plus CCCP) markedly decreased [14C]glutamine- and [32P]-phosphate-permeable mitochondrial spaces. A close correlation between the decrease of phosphate and glutamine permeable spaces and the inhibition of glutaminase activity was found.

(4) If the activation energy of the enzyme was determined with frozen mitochondrial preparations, a discontinuity or break in the Arrhenius plot was observed, whereas the presence of a detergent completely abolished the break. Digitonin or ultrasonic treatment of the mitochondria followed by separation of the membrane and the soluble fraction revealed that glutaminase is a membrane-bound enzyme.

On the basis of these findings it is concluded that there is an association between the transport of phosphate on one side and the transport of glutamine and glutaminase activity on the other. It is possible that the movement of phosphate across the membrane activates the enzyme which facilitates diffusion of glutamine down a concentration gradient. However, the existence of a specific glutamine-phosphate carrier is not ruled out.  相似文献

The sensitization of chlorococcal algae by 5-BdU for the purpose of UV-light mutagenesis was studied. The results obtained were compared with our earlier findings on the sensitization of the same algal strains by 5-BU. No shielding effect of the 5-BdU molecules against UV-light was observed. Probably, the uptake of them from the liquid medium did not result in such excess as compared with the treatment by 5-BU, even if the cells were long enough (24 h) exposed to the concentration of 5-BdU. Likewise, neither stimulating nor inhibiting growth effects on the growing cell colonies were observed after treatment with 5-BdU. The sensitization of the algal cells for UV-light effects was effective in all the experiments. An increased damage of the algal cells by UV-light after sensitization was proved in all the parameters recorded. The frequencies of permanent changes of the cells or their colonies were also increased, but their spectrum did not change significantly. A suitable combination of the 5-BdU sensitization of the cells before their influencing by UV-light and the induction of their repair mechanisms by visible light may decrease the frequencies of the lethal or sublethal damage and increase the frequencies of the useful permanent changes in the characteristics of the chlorococcal algae. The results obtained are discussed from the viewpoint of the regulated mutation process in the breeding of algae.  相似文献

The mangrove formation on the coasts of S. Atlantic is very poor in species (only 6) compared with mangrove in S. and SE. Asia or E. Africa (more than 30). This fact cannot be explained by ecology. Ultrathin sectioning of submerged mycelium of Claviceps purpurea Tul. producing clavine alkaloids revealed yeast-like budding resulting in asexual sporesblastospores. These deciduous spores were born by extended hyphal cells and retained the same ultrastructure of cell organelles. Both the extended hyphae and the blastospores resembled the cells of ergot sclerotial tissue. A surface culture of C. purpurea Tul. producing no alkaloids was used as a reference.  相似文献

Results obtained when studying conjugation in mycobacteria by means of different methods are summarized. The method of conjugation on surface of a solid complete medium was tested with different auxotrophic mutants of different strains ofMycobacterium smegmatis. It was not possible to obtain positive results even by means of the above method. This was probably due to unsuitability of the chosen strains ofMycobacterium smegmatis. Preparation of the donor strain by transfer of the F factor fromEscherichia coli F’ORF 1ade + lac+ pro+ toMycobacterium phlei PA adeStm r by means of sexduction is described. Frequency of the phenotype PAade + Stmr increased in the average by two and a half orders of magnitude with respect to the control, however, a further transfer from cultures of the cellsade + Stmr to cells ade could not be demonstrated. Experiments aimed at transferring the R factor from strainsEscherichia coli K-12 toMycobacterium phlei were unsuccessful.  相似文献
