The temporal parameters of the perception of radially moving sound sources partly masked with broadband internalized noise at an intensity of 40, 46, or 52 dB above the hearing threshold have been studied. The threshold of sound duration necessary for identifying the direction of movement of the sound source (75% correct answers) increases from 135 ms in silence to 285 ms at all intensities of continuous noise studied. The minimum duration of the stimulus beginning with which a subsequent increase in duration does not increase the number of correct responses is the same (385 ms) under all conditions of stimulus presentation. Broadband noise of any intensity increases the time of response to stimuli in the range of durations studied. At a noise of 52 dB, which is close to the threshold of full masking, the reaction time is not increased significantly compared to its estimation at a noise of 46 dB. The minimum duration of the stimulus has proved to be the stablest temporal parameter of the perception of movement of a sound source. The present study analyzed effects of different cAMP-elevators on cell morphology in primary culture of human intimal and medial cells from grossly normal and atherosclerotic areas. In primary culture of human aortic cells adenylate cyclase activator forskolin and other cAMP elevators induced arborization of cells, i.e. they reversibly changed the shape of cells. This resulted in the formation of thin branching processes and in the concentration of cytoplasm around the nucleus. In the culture, the shape of the arborized cells resembled that of stellate ones detected in the aortic intima in situ. The arborization of cells was accompanied by destruction of myofilaments. Due to cAMP elevators' effect, most of the arborized cells were exhibited in the cultures isolated from the elastic-hyperplastic layer of the intima. The number of arborized cells was significantly less in the cultures isolated from the musculo-elastic layer and still lesser in those isolated from media. We failed to reveal any significant difference in the number of arborized cells cultured from fatty streaks, atherosclerotic plaques and grossly normal aortic areas. Obtained results suggest that the previously revealed polymorphism of human aortic intimal cells may be accounted for by the cell shape transformations underlined by the mechanism similar to that of arborization in vitro. 相似文献
We discovered that simple proline-containing peptides Gly-Pro, Pro-Gly, Pro-Gly-Pro, and semax had an antistress protective
effect on the organism appearing as anticoagulation system activation. Repeated intranasal injection of each of these peptides
to rats prior to acute immobilization stress prevented a hypercoagulation response to prolonged stress lasting 60 min. At
the same time there was increase of anti-thrombotic, anticoagulant, and fibrin depolymerization activity and recovery of enzymatic
fibrinolytic activity. Dipeptides were found to have the greatest antistress effect. Our results showed that semax had a protective
effect against enhanced blood coagulability resulting from repeated immobilization stress. 相似文献
Rhodopsin kinase (GRK1) is a member of G protein-coupled receptor kinase family and a key enzyme in the quenching of photolysed rhodopsin activity and desensitisation of the rod photoreceptor neurons. Like some other rod proteins involved in phototransduction, GRK1 is posttranslationally modified at the C terminus by isoprenylation (farnesylation), endoproteolysis and α-carboxymethylation. In this study, we examined the potential mechanisms of regulation of GRK1 methylation status, which have remained unexplored so far. We found that considerable fraction of GRK1 is endogenously methylated. In isolated rod outer segments, its methylation is inhibited and demethylation stimulated by low-affinity nucleotide binding. This effect is not specific for ATP and was observed in the presence of a non-hydrolysable ATP analogue AMP-PNP, GTP and other nucleotides, and thus may involve a site distinct from the active site of the kinase. GRK1 demethylation is inhibited in the presence of Ca(2+) by recoverin. This inhibition requires recoverin myristoylation and the presence of the membranes, and may be due to changes in GRK1 availability for processing enzymes upon its redistribution to the membranes induced by recoverin/Ca(2+). We hypothesise that increased GRK1 methylation in dark-adapted rods due to elevated cytoplasmic Ca(2+) levels would further increase its association with the membranes and recoverin, providing a positive feedback to efficiently suppress spurious phosphorylation of non-activated rhodopsin molecules and thus maximise senstivity of the photoreceptor. This study provides the first evidence for dynamic regulation of GRK1 α-carboxymethylation, which might play a role in the regulation of light sensitivity and adaptation in the rod photoreceptors. 相似文献
