We studied the role of phytohormones: zeatin, kinetin, and abscisic acid, in the regulation of development of the conidial inoculum of Erysiphe cichoracearumDC. f. phlogisJacz. and E. graminisDC. f. hordeiMarchal. When the pathogen conidia were in direct contact with phytohormones, the intensity of their germination significantly increased. In the presence of cytokinins, the amount of normal appressoria decreased and that of abnormal growth tubes increased. On the phlox leaves treated with cytokinins, the intensity of germination of the conidia increased, as compared to the control, while abscisic acid exerted the opposite effect. The treatment of barley leaves with cytokinins did not affect markedly the development of conidial inoculum, as compared to the control, while abscisic acid significantly decreased the intensity of germination of the conidia. On the leaves of different Phloxspecies, the degree of germination of the conidial negative correlated with their resistance against the powdery mildew. The role of cytokinins in pathogenesis of biotrophic fungi is discussed. 相似文献

Passive transport of ions and metabolites across the peribacteroid membrane (PBM) was investigated on symbiosome preparations isolated from the broad bean (Vicia faba L.) root nodules and suspended in a potassium-free medium. Optical density of the symbiosome suspension at 546 nm was monitored as an indicator of light-scattering changes. Depolarization of the PBM with tetraphenylphosphonium cation (TPP+) caused an increase in light scattering of symbiosome suspension. This effect was enhanced after adding a K+ ionophore valinomycin to the incubation medium. A similar effect was observed after supplementing the symbiosome suspension with nigericin, a K+/H+ antiporter. Similar experiments on bacteroid suspensions prepared from isolated symbiosomes did not reveal any appreciable changes in light scattering in the presence of the same membrane-active substances. The light scattering by symbiosome suspensions decreased after adding malate or succinate, while the subsequent addition of centimolar concentrations of K+ substantially accelerated this process. Light scattering by the symbiosome suspension was insensitive to the addition of glutamate, a substance normally impermeant through the PBM of legume root nodules. These results suggest that the changes in light scattering by symbiosomes reflect the osmotically induced changes of symbiosome volume. These volume changes were assigned to alteration of the peribacteroid space (PBS). The incubation of symbiosomes in a potassium-free medium acidified their the PBS; this acidification was accelerated by valinomycin, carbonyl cyanide p-(trifluoromethoxy)phenylhydrazone (FCCP), and nigericin, and it was abolished in the presence of comparatively high concentrations of K+ in the incubation medium. Heterotrimeric translation initiation factor (IF) a/eIF2 (archaeal/eukaryotic IF 2) is present in both Eukarya and Archaea. Despite strong structural similarity between a/eIF2 orthologs from the two domains of life, their functional relationship is obscure. Here, we show that aIF2 from Sulfolobus solfataricus can substitute for its mammalian counterpart in the reconstitution of eukaryotic 48S initiation complexes from purified components. aIF2 is able to correctly place the initiator Met-tRNAi into the P-site of the 40S ribosomal subunit and accompany the entire set of eukaryotic translation IFs in the process of cap-dependent scanning and AUG codon selection. However, it seems to be unable to participate in the following step of ribosomal subunit joining. In accordance with this, aIF2 inhibits rather than stimulates protein synthesis in mammalian cell-free system. The ability of recombinant aIF2 protein to direct ribosomal scanning suggests that some archaeal mRNAs may utilize this mechanism during translation initiation. 相似文献

The spatial configuration of a relativistic plasma bunch generated under the gyromagnetic autoresonance and confined in a magnetic mirror trap has been studied experimentally and numerically. The characteristics of bremsstrahlung generated by the plasma bunch from the gas and chamber walls were investigated using X-ray spectroscopy and radiometry, which made it possible to determine the localization of the bunch and analyze the dynamics of its confinement. 相似文献

The parameters of the injector of an axial plasma beam injected into a plasma accelerator operating on the basis of gyroresonance acceleration of electrons in the reverse magnetic field are determined. The trapping of the beam electrons into the regime of gyroresonance acceleration is numerically simulated by the particle- in-cell method. The optimal time of axial injection of the beam into a magnetic mirror trap is determined. A novel compound was identified in the acidic extract of Thymus armeniacus collected in the Lake Sevan region of Armenia. This compound, named “sevanol,” to our knowledge is the first low molecular weight natural molecule that has a reversible inhibition effect on both the transient and the sustained current of human ASIC3 channels expressed in Xenopus laevis oocytes. Sevanol completely blocked the transient component (IC50 353 ± 23 μm) and partially (∼45%) inhibited the amplitude of the sustained component (IC50 of 234 ± 53 μm). Other types of acid-sensing ion channel (ASIC) channels were intact to sevanol application, except ASIC1a, which showed more than six times less affinity to it as compared with the inhibitory action on the ASIC3 channel. To elucidate the structure of sevanol, the set of NMR spectra in two solvents (d6-DMSO and D2O) was collected, and the complete chemical structure was confirmed by liquid chromatography-mass spectrometry with electrospray ionization (LC-ESI+-MS) fragmentation. This compound is a new lignan built up of epiphyllic acid and two isocitryl esters in positions 9 and 10. In vivo administration of sevanol (1–10 mg/kg) significantly reversed thermal hyperalgesia induced by complete Freund''s adjuvant injection and reduced response to acid in a writhing test. Thus, we assume the probable considerable role of sevanol in known analgesic and anti-inflammatory properties of thyme. 相似文献

The physical properties of lipid bilayers, such as curvature and fluidity, can affect the interactions of polypeptides with membranes, influencing biological events. Additionally, given the growing interest in peptide-based therapeutics, understanding the influence of membrane properties on membrane-associated peptides has potential utility. pH low insertion peptides (pHLIPs) are a family of water-soluble peptides that can insert across cell membranes in a pH-dependent manner, enabling the use of pH to follow peptide-lipid interactions. Here we study pHLIP interactions with liposomes varying in size and composition, to determine the influence of several key membrane physical properties. We find that pHLIP binding to bilayer surfaces at neutral pH is governed by the ease of access to the membrane’s hydrophobic core, which can be facilitated by membrane curvature, thickness, and the cholesterol content of the membrane. After surface binding, if the pH is lowered, the kinetics of pHLIP folding to form a helix and subsequent insertion across the membrane depends on the fluidity and energetic dynamics of the membrane. We showed that pHLIP is capable of forming a helix across lipid bilayers of different thicknesses at low pH. However, the kinetics of the slow phase of insertion corresponding to the translocation of C-terminal end of the peptide across lipid bilayer, vary approximately twofold, and correlate with bilayer thickness and fluidity. Although these influences are not large, local curvature variations in membranes of different fluidity could selectively influence surface binding in mixed cell populations. 相似文献

The energy characteristics of an electron bunch accelerated by a wakefield are largely determined by the initial bunch dimensions. Present-day injectors are still incapable of ensuring the initial spatial parameters of the bunches required for their acceleration without increasing the energy spread of the bunch electrons. In connection with this, the possibility is studied of improving the energy characteristics of an accelerated bunch by precompressing it in the longitudinal direction in the stage of trapping by a wakefield. Analytic formulas are derived that describe the one-dimensional dynamics of the spatial and energy characteristics of a short (much shorter than the wakefield wavelength) electron bunch in both the trapping and acceleration stages. The analytical results obtained are shown to agree fairly well with the results from one-dimensional and three-dimensional simulations, provided that the electrons are injected into the region that is optimum for acceleration. The possibility is discussed of forming compressed bunches so as to ensure the high quality of the bunch in the course of its acceleration to high energies. 相似文献
