Background
The pro-inflammatory status of the elderly triggers most of the age-related diseases such as cancer and atherosclerosis. Atherosclerosis, the leading cause world wide of morbidity and death, is an inflammatory disease influenced by life-style and genetic host factors. Stimuli such as oxLDL or microbial ligands have been proposed to trigger inflammation leading to atherosclerosis. It has recently been shown that oxLDL activates immune cells via the Toll-like receptor (TLR) 4/6 complex. Several common single nucleotide polymorphisms (SNPs) of the TLR system have been associated with atherosclerosis. To investigate the role of TLR-6 we analyzed the association of the TLR-6 SNP Pro249Ser with atherogenesis.Results
Genotyping of two independent groups with CAD, as well as of healthy controls revealed a significant association of the homozygous genotype with a reduced risk for atherosclerosis (odds ratio: 0.69, 95% CI 0.51-0.95, P?=?0.02). In addition, we found a trend towards an association with the risk of restenosis after transluminal coronary angioplasty (odds ratio: 0.53, 95% CI 0.24-1.16, P?=?0.12). In addition, first evidence is presented that the frequency of this protective genotype increases in a healthy population with age. Taken together, our results define a role for TLR-6 and its genetic variations in modulating the inflammatory response leading to atherosclerosis.Conclusions
These results may lead to a better risk stratification, and potentially to an improved prophylactic treatment of high-risk populations. Furthermore, the protective effect of this polymorphism may lead to an increase of this genotype in the healthy elderly and may therefore be a novel genetic marker for the well-being during aging.83.

Progressive supranuclear palsy (PSP), multiple system atrophy (MSA) and idiopathic Parkinson’s disease (IPD) can be clinically indistinguishable, especially in the early stages, despite distinct patterns of molecular pathology. Structural neuroimaging holds promise for providing objective biomarkers for discriminating these diseases at the single subject level but all studies to date have reported incomplete separation of disease groups. In this study, we employed multi-class pattern recognition to assess the value of anatomical patterns derived from a widely available structural neuroimaging sequence for automated classification of these disorders. To achieve this, 17 patients with PSP, 14 with IPD and 19 with MSA were scanned using structural MRI along with 19 healthy controls (HCs). An advanced probabilistic pattern recognition approach was employed to evaluate the diagnostic value of several pre-defined anatomical patterns for discriminating the disorders, including: (i) a subcortical motor network; (ii) each of its component regions and (iii) the whole brain. All disease groups could be discriminated simultaneously with high accuracy using the subcortical motor network. The region providing the most accurate predictions overall was the midbrain/brainstem, which discriminated all disease groups from one another and from HCs. The subcortical network also produced more accurate predictions than the whole brain and all of its constituent regions. PSP was accurately predicted from the midbrain/brainstem, cerebellum and all basal ganglia compartments; MSA from the midbrain/brainstem and cerebellum and IPD from the midbrain/brainstem only. This study demonstrates that automated analysis of structural MRI can accurately predict diagnosis in individual patients with Parkinsonian disorders, and identifies distinct patterns of regional atrophy particularly useful for this process. 相似文献

Objective
The use of pictorial warning labels on cigarette packages is one of the provisions included in the first ever global health treaty by the World Health Organization against the tobacco epidemic. There is substantial evidence demonstrating the effectiveness of graphic health warning labels on intention to quit, thoughts about health risks and engaging in cessation behaviors. However, studies that address the implicit emotional drives evoked by such warnings are still underexplored. Here, we provide experimental data for the use of pictorial health warnings as a reliable strategy for tobacco control.Methods
Experiment 1 pre-tested nineteen prototypes of pictorial warnings to screen for their emotional impact. Participants (n = 338) were young adults balanced in gender, smoking status and education. Experiment 2 (n = 63) tested pictorial warnings (ten) that were stamped on packs. We employed an innovative set-up to investigate the impact of the warnings on the ordinary attitude of packs’ manipulation, and quantified judgments of warnings’ emotional strength and efficacy against smoking.Findings
Experiment 1 revealed that women judged the warning prototypes as more aversive than men, and smokers judged them more aversive than non-smokers. Participants with lower education judged the prototypes more aversive than participants with higher education. Experiment 2 showed that stamped warnings antagonized the appeal of the brands by imposing a cost to manipulate the cigarette packs, especially for smokers. Additionally, participants’ judgments revealed that the more aversive a warning, the more it is perceived as effective against smoking.Conclusions
Health warning labels are one of the key components of the integrated approach to control the global tobacco epidemic. The evidence presented in this study adds to the understanding of how implicit responses to pictorial warnings may contribute to behavioral change. 相似文献85.

Objective:
Consuming smaller, more frequent meals is often advocated as a means of controlling body weight, but studies demonstrating a mechanistic effect of this practice on factors associated with body weight regulation are lacking. The purpose of this study was to compare the effect of consuming three (3M) vs. six meals (6M) per day on 24‐h fat oxidation and subjective ratings of hunger.Design and Methods:
Lean (body mass index <25 kg/m2) subjects (7M, 8F) were studied in a whole‐room calorimeter on two occasions in a randomized cross‐over design. Subjects were provided isoenergetic, energy balanced diets with a 1‐ to 2‐week washout between conditions. Hunger, fullness, and “desire to eat” ratings were assessed throughout the day using visual analog scales and quantified as area under the curve (AUC).Results:
There were no differences (P < 0.05) in 24‐h energy expenditure (8.7 ± 0.3 vs. 8.6 ± 0.3 mj d?1), 24‐h respiratory quotient (0.85 ± 0.01 vs. 0.85 ± 0.01), or 24‐h fat oxidation (82 ± 6 vs. 80 ± 7 g day‐1) between 3M and 6M, respectively. There was no difference in fullness 24‐h AUC, but hunger AUC (41850 ± 2255 vs. 36612 ± 2556 mm.24 h, P = 0.03) and “desire to eat” AUC (47061 ± 1791 vs. 41170 ± 2574 mm.24 h, P = 0.03) were greater during 6M than 3M.Conclusion:
We conclude that increasing meal frequency from three to six per day has no significant effect on 24‐h fat oxidation, but may increase hunger and the desire to eat.86.

The parapagurid hermit crabs Sympagurus dimorphus (Studer, 1883) and Parapagurus bouvieri (Stebbing, 1910) form dominant components of South African deeper-water benthic invertebrate communities and, as adults, forego the use of “traditional” gastropod shells in favour of symbiotically associated Epizoanthus carcinoecia. A total of 1,122 adult S. dimorphus and 549 adult P. bouvieri derived from demersal research trawl surveys were examined to investigate parameters of population structure, carcinoecia association and morphology. S. dimorphus had a female-biased sex ratio of 1:1.5, and P. bouvieri an equal sex ratio of 1:1. The proportion of males of both species increased with cephalothoracic shield length (CL). The lengths of the major right chelipeds of both S. dimorphus males and females, and P. bouvieri males were positively allometric, and negatively allometric in P. bouvieri females. All P. bouvieri inhabited carcinoecia, while 3.39% (n = 38) of S. dimorphus specimens occupied gastropod shells. There was a positive relationship between S. dimorphus CL and rate of gastropod shell occupation (r2 = 0.96). In S. dimorphus, number of zoanthid polyps forming the carcinoecia pseudoshell increased with both total shell volume (R2 = 0.73) and colony height (R2 = 0.61), but no such relationships were found for P. bouvieri. Euspira napus was the most common original shell around which carcinoecia developed in both S. dimorphus (45.45%) and P. bouvieri (39.00%). We speculate that these hermit species exist along an evolutionary continuum of carcinoecia use. Sympagurus dimorphus retains the ability to occupy both gastropod shells and pseudoshells and, like most other hermit crabs, continue to use shells/carcinoecia as shelters to protect the entire body. In contrast, adult P. bouvieri appear to be totally reliant on carcinoecia, which they use more as a brood pouch covering only the abdomen, rather than as a shelter into which they can fully retract to escape predation. 相似文献

Src tyrosine kinases are a family of non-receptor proteins that are responsible for the growth process, cellular proliferation, differentiation and survival. Lack of Src kinase control has been associated with the development of certain human cancers. This family of proteins is constituted of four domains, with SH1 being the kinase or catalytic domain. SH1 also presents three important regulatory sites. Two residues, Tyr416 and Tyr527, are responsible for important phosphorylation processes that lead to, respectively, activation and deactivation of these kinases. More recently, however, a set of four cysteine residues located near the C-terminus-Cys483, Cys487, Cys496 and Cys498-has been associated with the activation of the Src kinases through S-nitrosylation reactions. Particularly, the Cys498 has been specified as a fundamental residue when considering this regulatory mechanism. Aiming to understand the role of these four cysteines in S-nitrosylation, theoretical studies of electrostatic, steric and hydrophobic properties were performed with a sequence of 20 amino acids, enclosing the four cysteine residues under study, extracted from the PDB coordinates of the crystal obtained from the inactive state of Src kinase. Results indicate that Cys498 is buried deeply in the protein, in hydrophobic surroundings in which NO is more likely to suffer decomposition into the electrophilic intermediates known to be responsible for S-nitrosylation reactions. Electronic calculated properties, such as punctual atomic charges, electrostatic potentials and molecular orbital energy, also demonstrated the good nucleophilic potential of Cys498. 相似文献

Increasing production of biofuels has led to concerns about indirect land‐use change (ILUC). So far, significant efforts have been made to assess potential ILUC effects. But limited attention has been paid to strategies for reducing the extent of ILUC and controlling the type of LUC. This case study assesses five key ILUC mitigation measures to quantify the low‐ILUC‐risk production potential of miscanthus‐based bioethanol in Lublin province (Poland) in 2020. In 2020, a total area of 196 to 818 thousand hectare of agricultural land could be made available for biomass production by realizing above‐baseline yield developments (95–413 thousand ha), increased food chain efficiencies (9–30 thousand ha) and biofuel feedstock production on underutilized lands (92–375 thousand ha). However, a maximum 203–269 thousand hectare is considered legally available (not protected) and biophysically suitable for miscanthus production. The resulting low‐ILUC‐risk bioethanol production potential ranges from 12 to 35 PJ per year. The potential from this region alone is higher than the national Polish target for second‐generation bioethanol consumption of 9 PJ in 2020. Although the sustainable implementation potential may be lower, the province of Lublin could play a key role in achieving this target. This study shows that the mitigation or prevention of ILUC from bioenergy is only possible when an integrated perspective is adopted on the agricultural and bioenergy sectors. Governance and policies on planning and implementing ILUC mitigation are considered vital for realizing a significant bioenergy potential with low ILUC risk. One important aspect in this regard is monitoring the risk of ILUC and the implementation of ILUC mitigation measures. Key parameters for monitoring are land use, land cover and crop yields. 相似文献
