Human transfer factor (TF) was fractionated by exclusion chromatography and the fractions were tested for biologic activity in vivo and in vitro. Specific TF activity in vivo was found to reside in the major UV-absorbing peak (Fraction III). Fraction III eluted at 2.7 X V(O) and transferred tuberculin, candida, or KLH-reactivity to previously negative recipients. Fraction III from nonreactive donors was ineffective. When the fractions were tested in vitro, we found that both the mitogenic activity of whole TF and the suppressive activity to mitogen activation when present in TF was found in Fraction I. Fraction III contained components responsible for augmentation of PHA and PWM responses. In addition, Fraction III contained the component responsible for antigen-dependent augmentation of lymphocyte transformation. Fraction IV was suppressive to antigen-induced lymphocyte transformation. These data suggest that TF preparations contain components which can affect immune reactions in both specific and nonspecific ways. 相似文献

Both the hormone dependency and the morphological details of estrogen-dependent ciliogenesis in the shell gland of the chick oviduct were investigated. Ciliogenesis was initiated on day 3 of estrogen treatment, and progressively more cells became differentiated until, on day 10, 55% ciliation occurred with 17-estradiol (1 mg/day) and 75% ciliation occurred with diethylstilbestrol (1 mg/day). Simultaneous administration of progesterone with diethylstilbestrol (1 mg each/day for 10 days) caused a 50% depression in the number of ciliated cells on day 10. The rate of ciliogenesis was found to be affected by progesterone and the type of estrogen administered. The minimum stimulatory dose of estradiol was found to be between 0.01 mg/day and 0.05 mg/day. Ciliogenic cells were first recognized by the appearance of pro-basal bodies in the apical portion of the cell. Pro-basal body maturation and cilium formation were the same as those described for the chick trachea. Ciliogenesis in the chick was found to be homologous to estrogen-dependent ciliogenesis in various mammalian oviducts. 相似文献

Aspects of the physiology of iron deposition of an iron-precipitating strain of Sphaerotilus were investigated in laboratory culture to characterize the process. Measurement of growth (incorporated L-[3H]alanine) and iron deposition (incorporated 59Fe) demonstrated that Sphaerotilus exhibit a characteristic temporal pattern of iron deposition, which is delayed until the latter portion of the exponential or the onset of the stationary growth phase. The growth rate (mu=0.17 h-1) was apparently independent of the iron concentration in the medium. There was, furthermore, no direct correlation between the iron concentration and final cell protein yield. It was concluded from experiments involving growth on artificial substrata (glass cover slips) that sessile populations derived no phsiological advantage (manifested as differences in growth rates) over free-living cells. There was no difference in the rate or onset of iron deposition of attached compared to suspended cells. Blocking of protein synthesis by the addition of chloramphenicol suggested that, once iron deposition was initiated, continued protein synthesis was not required for full expression of this capability. The results suggested that iron deposition may be possibly mediated by certain of the constituents of the organism's sheath. 相似文献

The development of a Holocene peat near Marudi (Sarawak) and a Miocene coal near Berakas (Brunei) is studied pollen-analytically and stratigraphically by comparison with present-day swamp vegetation along the NW Bornean coast. A number of 76 pollen and spore types were recognized and are briefly diagnosed.The Marudi profile starts on mangrove sediment and shows a succession which is closely comparable to the present-day zonation of the large raised bog of which it forms part.The Berakas profile starts on fluviatile sediments and remains in the mixed swamp forest stage. This small deposit has remained close to the coast and peat growth is terminated by marine transgression.The floristic composition of the mixed swamp forest stage in both bogs, separated in age by approximately seven million years is closely comparable. Minor differences which exist can be attributed to differences in size, ecologic conditions and accessibility from mainland reservoirs.Only on spore type, Stenochlaena areolaris has become extinct in Borneo. 相似文献

Aedes aegypti and Ae. albopictus are the main vectors of mosquito-borne viruses of medical and veterinary significance. Many of these viruses have RNA genomes. Exogenously provided, e.g. transgene encoded, small RNAs could be used to inhibit virus replication, breaking the transmission cycle. We tested, in Ae. aegypti and Ae. albopictus cell lines, reporter-based strategies for assessing the ability of two types of small RNAs to inhibit a chikungunya virus (CHIKV) derived target. Both types of small RNAs use a Drosophila melanogaster pre-miRNA-1 based hairpin for their expression, either with perfect base-pairing in the stem region (shRNA-like) or containing two mismatches (miRNA-like). The pre-miRNA-1 stem loop structure was encoded within an intron; this allows co-expression of one or more proteins, e.g. a fluorescent protein marker tracking the temporal and spatial expression of the small RNAs in vivo. Three reporter-based systems were used to assess the relative silencing efficiency of ten shRNA-like siRNAs and corresponding miRNA-like designs. Two systems used a luciferase reporter RNA with CHIKV RNA inserted either in the coding sequence or within the 3’ UTR. A third reporter used a CHIKV derived split replication system. All three reporters demonstrated that while silencing could be achieved with both miRNA-like and shRNA-like designs, the latter were substantially more effective. Dcr-2 was required for the shRNA-like siRNAs as demonstrated by loss of inhibition of the reporters in Dcr-2 deficient cell lines. Neuronal hippocampal Ca2+ dysregulation is a critical component of cognitive decline in brain aging and Alzheimer''s disease and is suggested to impact communication and excitability through the activation of a larger after hyperpolarization. However, few studies have tested for the presence of Ca2+ dysregulation in vivo, how it manifests, and whether it impacts network function across hundreds of neurons. Here, we tested for neuronal Ca2+ network dysregulation in vivo in the primary somatosensory cortex (S1) of anesthetized young and aged male Fisher 344 rats using single‐cell resolution techniques. Because S1 is involved in sensory discrimination and proprioception, we tested for alterations in ambulatory performance in the aged animal and investigated two potential pathways underlying these central aging‐ and Ca2+‐dependent changes. Compared to young, aged animals displayed increased overall activity and connectivity of the network as well as decreased ambulatory speed. In aged animals, intranasal insulin (INI) increased network synchronicity and ambulatory speed. Importantly, in young animals, delivery of the L‐type voltage‐gated Ca2+ channel modifier Bay‐K 8644 altered network properties, replicating some of the changes seen in the older animal. These results suggest that hippocampal Ca2+ dysregulation may be generalizable to other areas, such as S1, and might engage modalities that are associated with locomotor stability and motivation to ambulate. Further, given the safety profile of INI in the clinic and the evidence presented here showing that this central dysregulation is sensitive to insulin, we suggest that these processes can be targeted to potentially increase motivation and coordination while also reducing fall frequency with age. 相似文献
