ADP plays an integral role in the process of hemostasis by signaling through two platelet G-protein-coupled receptors, P2Y1 and P2Y12. The recent use of antagonists against these two receptors has contributed a substantial body of data characterizing the ADP signaling pathways in human platelets. Specifically, the results have indicated that although P2Y1 receptors are involved in the initiation of platelet aggregation, P2Y12 receptor activation appears to account for the bulk of the ADP-mediated effects. Based on this consideration, emphasis has been placed on the development of a new class of P2Y12 antagonists (separate from clopidogrel and ticlopidine) as an approach to the treatment of thromboembolic disorders. The present work examined the molecular mechanisms by which two of these widely used adenosine-based P2Y12 antagonists (2-methylthioadenosine 5′-monophosphate triethylammonium salt (2MeSAMP) and ARC69931MX), inhibit human platelet activation. It was found that both of these compounds raise platelet cAMP to levels that substantially inhibit platelet aggregation. Furthermore, the results demonstrated that this elevation of cAMP did not require Gi signaling or functional P2Y12 receptors but was mediated through activation of a separate G protein-coupled pathway, presumably involving Gs. However, additional experiments revealed that neither 2MeSAMP nor ARC69931MX (cangrelor) increased cAMP through activation of A2a, IP, DP, or EP2 receptors, which are known to couple to Gs. Collectively, these findings indicate that 2MeSAMP and ARC69931MX interact with an unidentified platelet G protein-coupled receptor that stimulates cAMP-mediated inhibition of platelet function. This inhibition is in addition to that derived from antagonism of P2Y12 receptors.Upon damage to the endothelial layer of the blood vessel wall, the underlying subendothelium is exposed to platelets in the blood, initiating a cascade of signaling events resulting in the transformation of “resting” platelets into “activated” platelets (1). One significant characteristic associated with these signaling events is the secretion of ADP from the platelet-dense granules (2). This released ADP acts to further amplify the platelet activation response by interacting with its G-protein-coupled receptors on the platelet surface, namely P2Y1 (coupled to Gq) and P2Y12 (coupled to Gi) (35). The consequence of platelet activation through ADP is a conformational change in the platelet membrane glycoprotein αIIbβ3 (6, 7), which then binds to fibrinogen present in the plasma. The binding of fibrinogen with αIIbβ3 on the surface of adjacent platelets results in fibrinogen-platelet cross-linking and the formation of a hemostatic plug at the site of vascular injury (8).Consequently, ADP is thought to play an integral role in the normal process of hemostasis. Of the two ADP-receptor signaling pathways in platelets, evidence has indicated that ADP-mediated P2Y12 signaling appears to play a more prominent role in platelet activation than ADP-mediated P2Y1 signaling (9, 10). For the most part, support for this notion derives from the use of the adenosine-based P2Y12 antagonists (i.e. 2MeSAMP4 and ARC69931MX), which have a much broader inhibitory profile than P2Y1 antagonists (e.g. A3P5P (adenosine-3′-phosphate-5′-phosphate) or MRS2179) (9). Thus, 2MeSAMP and ARC69931MX inhibit platelet aggregation in response to multiple agonists, such as thromboxane A2, collagen, thrombin, etc. (1113), whereas P2Y1 antagonists do not. On the other hand, this general requirement for P2Y12 signaling seems to be inconsistent with earlier reports indicating that activation of certain platelet receptors (e.g. thromboxane A2 receptor) can cause aggregation through ADP-independent mechanisms (14, 15). Based on this apparent inconsistency in the contribution of P2Y12 signaling to the overall platelet activation response, the present study investigated the possibility that the broad spectrum of inhibitory activity of this new generation of P2Y12 antagonists (i.e. MeSAMP and ARC69931MX) may derive from an elevation in platelet cAMP levels.Our data demonstrated that both 2MeSAMP and ARC69931MX do in fact significantly raise human platelet cAMP. Furthermore, this pharmacological effect is independent of P2Y12-Gi signaling and appears to proceed through activation of a separate Gs-coupled platelet receptor. Taken together, the results therefore indicate that these adenosine-based P2Y12 antagonists can produce their inhibition of platelet function through a cAMP-mediated mechanism.  相似文献

Supermatrix and supertree are two methods for constructing a phylogenetic tree by using multiple data sets. However, these methods are not a panacea, as conflicting signals between data sets can lead to misinterpret the evolutionary history of taxa. In particular, the supermatrix approach is expected to be misleading if the species-tree signal is not dominant after the combination of the data sets. Moreover, most current supertree methods suffer from two limitations: (i) they ignore or misinterpret secondary (non-dominant) phylogenetic signals of the different data sets; and (ii) the logical basis of node robustness measures is unclear.To overcome these limitations, we propose a new approach, called SuperTRI, which is based on the branch support analyses of the independent data sets, and where the reliability of the nodes is assessed using three measures: the supertree Bootstrap percentage and two other values calculated from the separate analyses: the mean branch support (mean Bootstrap percentage or mean posterior probability) and the reproducibility index.The SuperTRI approach is tested on a data matrix including seven genes for 82 taxa of the family Bovidae (Mammalia, Ruminantia), and the results are compared to those found with the supermatrix approach. The phylogenetic analyses of the supermatrix and independent data sets were done using four methods of tree reconstruction: Bayesian inference, maximum likelihood, and unweighted and weighted maximum parsimony. The results indicate, firstly, that the SuperTRI approach shows less sensitivity to the four phylogenetic methods, secondly, that it is more accurate to interpret the relationships among taxa, and thirdly, that interesting conclusions on introgression and radiation can be drawn from the comparisons between SuperTRI and supermatrix analyses. 1 Foliar nitrogen concentration, which can be manipulated in crop plants by fertilizer supply, has long been recognized as a major factor in phytophagous insect abundance and performance. More recently, the type of fertilizer supplied has been shown to influence the abundance of some herbivore species. The diamondback moth Plutella xylostella is a global pest of Brassica crops. Although it has been the subject of numerous studies on host-plant resistance and pest control, few studies have addressed the effect of abiotic factors, such as nutrient supply, on its performance and behaviour.
2 We assessed oviposition preference, larval feeding preference and larval performance of P. xylostella on two cultivars of Brassica oleracea . Plants were grown using two fertilizer types, John Innes fertilizer and an organic animal manure, at high and low concentrations.
3  Plutella xylostella laid more eggs on cultivar Derby Day than Drago. Derby Day was also the cultivar on which larval performance was maximized. However, differences in larval performance between cultivars were only found when plants were grown in compost with John Innes fertilizer, and not when fertilized with animal manure.
4 Foliar nitrogen concentration was greater in plants grown in high fertilizer treatments but did not differ between cultivars. The concentrations of three glucosinolate compounds (glucoiberin, sinigrin and glucobrassicin) were greater in the high fertilizer treatments. Glucosinolate concentrations were higher in the Drago than the Derby Day cultivar.
5 These results are discussed in relation to the preference-performance hypothesis, and the assessment of plant resistance differences between cultivars using different types of fertilizer.  相似文献

In this study, scaling down nanosuspension production to 10 mg of drug compound and evaluation of the nanosuspensions to 1 mg of drug compound per test were investigated. Media milling of seven model drug compounds (cinnarizine–indomethacin–itraconazole–loviride–mebendazole–naproxen–phenytoin) was evaluated in a 96-well plate setup (10, 20, and 30 mg) and a glass-vial-based system in a planetary mill (10, 100, and 1,000 mg). Physicochemical properties evaluated on 1 mg of drug compound were drug content (high-performance liquid chromatography), size [dynamic light scattering (DLS)], morphology (scanning electron microscopy), thermal characteristics (differential scanning calorimetry), and X-ray powder diffraction (XRPD). Scaling down nanosuspension production to 10 mg of drug compound was feasible for the seven model compounds using both designs, the planetary mill design being more robust. Similar results were obtained for both designs upon milling 10 mg of drug compound. Drug content determination was precise and accurate. DLS was the method of choice for size measurements. Morphology evaluation and thermal analysis were feasible, although sample preparation had a big influence on the results. XRPD in capillary mode was successfully performed, both in the suspended state and after freeze-drying in the capillary. Results obtained for the latter were superior. Both the production and the physicochemical evaluation of nanosuspensions can be successfully downscaled, enabling nanosuspension screening applications in preclinical development settings.  相似文献

Individuals in groups are often thought to scan their surroundings for threats independently of one another. Models, however, suggest that foragers should monitor the vigilance level of their neighbours to prevent cheating, and to gather information about incipient predation risk. Evidence for monitoring of vigilance is scant. Here, I examined changes in vigilance levels in sleeping gulls (Larus sp.) surrounded by neighbours in various states of alertness. Controlling for group size and neighbour density, gulls interrupted sleep more often to scan their surroundings, and were therefore more vigilant, when their neighbours were alert rather than sleeping or preening. The results provide evidence for copying of vigilance within groups of birds, suggesting a complex flow of information about predation risk in groups.  相似文献

Acoustic heart signals, generated by the mechanical processes of the cardiac cycle, carry significant information about the underlying functioning of the cardiovascular system. We describe a computational analysis framework for identifying distinct morphologies of heart sounds and classifying them into physiological states. The analysis framework is based on hierarchical clustering, compact data representation in the feature space of cluster distances and a classification algorithm. We applied the proposed framework on two heart sound datasets, acquired during controlled alternations of the physiological conditions, and analyzed the morphological changes induced to the first heart sound (S1), and the ability to predict physiological variables from the morphology of S1. On the first dataset of 12 subjects, acquired while modulating the respiratory pressure, the algorithm achieved an average accuracy of 82 ± 7% in classifying the level of breathing resistance, and was able to estimate the instantaneous breathing pressure with an average error of 19 ± 6%. A strong correlation of 0.92 was obtained between the estimated and the actual breathing efforts. On the second dataset of 11 subjects, acquired during pharmacological stress tests, the average accuracy in classifying the stress stage was 86 ± 7%. The effects of the chosen raw signal representation, distance metrics and classification algorithm on the performance were studied on both real and simulated data. The results suggest that quantitative heart sound analysis may provide a new non-invasive technique for continuous cardiac monitoring and improved detection of mechanical dysfunctions caused by cardiovascular and cardiopulmonary diseases.  相似文献

Undifferentiated pleomorphic sarcoma/Malignant Fibrous Histiocytoma (MFH) is one of the most common subtypes of human soft tissue sarcoma. Using cross species genomic analysis, we define a geneset from the LSL-KrasG12D; Trp53Flox/Flox mouse model of soft tissue sarcoma that is highly enriched in human MFH. With this mouse geneset as a filter, we identify expression of the RAS target FOXM1 in human MFH. Expression of Foxm1 is elevated in mouse sarcomas that metastasize to the lung and tissue microarray analysis of human MFH correlates overexpression of FOXM1 with metastasis. These results suggest that genomic alterations present in human MFH are conserved in the LSL-KrasG12D; p53Flox/Flox mouse model of soft tissue sarcoma and demonstrate the utility of this pre-clinical model.  相似文献

Trophic indicators were used to compare two Malian freshwater reservoirs whose main differences are based on their different fishing pressures. Data were collected from a scientific survey of small-scale fishery landings conducted in 2002/2003. The trophic levels of fish species caught by artisanal fisheries are estimated from observations of scientific fishing or from the metabase Fishbase. Important differences exist in the trophic structure of both reservoirs. In Selingue (with high fishing pressure), very few top predators are found in the catches while the low trophic level fishes increase in total catches. In Manantali (with low fishing pressure), the top predators contribute twice as much to catches compared to Selingue. Hence, the mean trophic level of catches in Selingue (2.80) is lower than in Manantali (2.97). When comparing these results with those of study made in 1994/1995, it clearly appears that the effects of the fishing pressure in Selingue are obvious through a decrease of 0.12 in the mean trophic level while in Manantali this mean level has increased by 0.33 due to a recent strategic targeting of top predators. Trophic spectra seem to be relevant tools to characterize exploited fish communities from multi-specific and multi-gear small-scale fisheries catch data.  相似文献

2'-Substituted analogs of triclosan have been synthesized to target inhibition of the key malarial enzyme Plasmodium falciparum enoyl acyl carrier protein reductase (PfENR). Many of these compounds exhibit good potency (EC50<500 nM) against in vitro cultures of drug-resistant and drug-sensitive strains of the P. falciparum parasite and modest (IC50=1-20 microM) potency against purified PfENR enzyme. Compared to triclosan, this survey of 2'-substituted derivatives has afforded gains in excess of 20- and 30-fold versus the 3D7 and Dd2 strains of parasite, respectively.  相似文献
