Effects of transcutaneous electrical spinal cord stimulation (tESCS) on the parameters of stepping movements in healthy subjects were investigated during two kinds of activity: walking on a moving treadmill belt (active treadmill) as well as pushing the treadmill belt by effort of the legs (passive treadmill). It was found that the total interference electromyogram (EMG) activity during stepping performance on a passive treadmill was 1.5–2 times higher than during stepping on an active treadmill. In addition, the amplitude of angular displacement of the hip joint and ankle was 2.5 times and 1.7 times higher, respectively, during passive vs. active treadmill, while the duration of stepping cycle decreased by 19%. Although the muscles were exposed to different load and the parameters of motion on the active and passive treadmill were different, tESCS caused an increase in the total EMG activity in 96% of cases both on the active and on the passive treadmill. In both cases, the stepping cycle period decreased by 4–43% in all subjects. These results suggest that tESCS can affect voluntary stepping patterns under conditions of different afferent control.  相似文献

The effects of local anesthetics (LAs), including aminoamides and aminoesters, on the characteristics of single gramicidin A (GA) channels in 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) bilayers were studied. Aminoamides, namely lidocaine (LDC), prilocaine (PLC), mepivacaine (MPV), and bupivacaine (BPV), reduced the conductance of GA channels. Aminoesters influenced the current fluctuations induced by GA differently; procaine (PC) did not affect the fluctuations, whereas tetracaine (TTC) distinctly reduced the conductance of single GA channels. Using electrophysiological technique, we estimated the changes in the membrane boundary potential at the adsorption of LAs; LDC, PLC, MPV, BPV, and TTC substantially increased, while PC did not affect it. To elucidate which component of the membrane boundary potential, the surface or dipole potential, is responsible for the observed effects of LAs, we employed a fluorescence assay. We found that TTC led to a significant increase in the membrane dipole potential, whereas the adsorption of LDC, PLC, MPV, BPV, and PC did not produce any changes in the membrane dipole potential. We concluded that aminoamides affected the surface potential of lipid bilayers. Together, these data suggest that the effects of LAs on the conductance of single GA channels are caused by their influence on membrane electrostatic potentials; the regulation of GA pores by aminoamides is associated with the surface potential of membranes, whereas TTC modulation of channel properties is predominantly due to changes in dipole potential of lipid bilayers. These data might provide some significant implications for voltage-gated ion channels of cell membranes.  相似文献

The cells of haploid Aspergillus niger strains contain, on the average, 7-9 nuclei, a fragment of a thin hypha 100 me long comprising 11-19 nuclei. The cells of a diploid strain are 1.5-2.6 times larger in volume. The diploid cells contain less nuclei and more cytoplasm per nucleus as compared to the haploid strains. The primary sterigmae of Aspergillus niger comprise 3-13 nuclei, the secondary sterigmae and conidia, one nucleus. The conidia of the diploid strains are 1.8-2.0 times larger in volume and contain twice as much DNA as compared to the haploid strains.  相似文献

Equal partitioning of the multi-copy 2-micron plasmid of the budding yeast Saccharomyces cerevisiae requires association of the plasmid Rep1 and Rep2 proteins with the plasmid STB partitioning locus. Determining how the Rep proteins contribute has been complicated by interactions between the components. Here, each Rep protein was expressed fused to the DNA-binding domain of the bacterial repressor protein LexA in yeast harboring a replication-competent plasmid that had LexA-binding sites but lacked STB. Plasmid transmission to daughter cells was increased only by Rep2 fusion expression. Neither Rep1 nor a functional RSC2 complex (a chromatin remodeler required for 2-micron plasmid partitioning) were needed for the improvement. Deletion analysis showed the carboxy-terminal 65 residues of Rep2 were required and sufficient for this Rep1-independent inheritance. Mutation of a conserved basic motif in this domain impaired Rep1-independent and Rep protein/STB-dependent plasmid partitioning. Our findings suggest Rep2, which requires Rep1 and the RSC2 complex for functional association with STB, directly participates in 2-micron plasmid partitioning by linking the plasmid to a host component that is efficiently partitioned during cell division. Further investigation is needed to reveal the host factor targeted by Rep2 that contributes to the survival of these plasmids in their budding yeast hosts.  相似文献

The class Kinetoplastea encompasses both free-living and parasitic species from awide range of hosts. Several representatives of this group are responsible for severehuman diseases and for economic losses in agriculture and livestock. While this groupencompasses over 30 genera, most of the available information has been derived fromthe vertebrate pathogenic genera LeishmaniaandTrypanosoma. Recent studies of the previously neglected groups ofKinetoplastea indicated that the actual diversity is much higher than previouslythought. This article discusses the known segment of kinetoplastid diversity and howgene-directed Sanger sequencing and next-generation sequencing methods can help todeepen our knowledge of these interesting protists.  相似文献

A gram-positive, motile, strict anaerobic spore-forming bacterium was isolated from the over-cooled brine in the permafrost. The optimal temperature for isolate growth was 5-6 degrees C at pH 6.8-7.2. The bacterium was growing on the medium rich in saccharides and disaccharides. Out of polysaccharides tested, only xylan sustained the growth. Fermentation of the hexoses led to the formation of acetate, butyrate, lactate, H2,CO2 and some formate and ethanol. Cell wall peptidoglycan contained meso-diaminopimelic acid. The major fatty acids of the cell wall were C(14:0) and C(16:1c9). The content of G-C pairs in DNA was 31.4 mol%. As phylogenetic analysis has shown, it is closely linked to the members of cluster 1 of Clostridium. It differs from the other species of the genus by the substrates necessary for the growth, products forming as a result of the fermentation and content of the fatty acids in the cell wall. Thus, it was suggested to describe this strain as a new species named Clostridium algoriphilum. Type strain 14D1 was deposited into the Russian Collection of the Microorganisms VKM B-2271T and German Collection of the Microorganisms DSM 16153T .  相似文献

The paper deals with the microbiological characterization of water-saturated horizons in permafrost soils (cryopegs) found on the Varandei Peninsula (Barents Sea coast), 4–20 m deep. The total quantity of bacteria in the water of cryopegs was 3.5 × 108 cells/ml. The population of cultivated aerobic heterotrophic bacteria was 3–4 × 107 cells/ml and the number of anaerobic heterotrophic bacteria varied from 102 to 105 cells/ml depending on cultivation temperature and salinity. Sulfate-reducing bacteria and methanogenic archaea were found as hundreds and tens of cells per ml of water, respectively. A pure culture of a sulfate-reducing strain B15 was isolated from borehole 21 and characterized. Phylogenetic analysis has shown that the new bacterium is a member of the genus Desulfovibrio with Desulfovibrio mexicanus as its closest relative (96.5% similarity). However, the significant phenotypic differences suggest that strain B15 is a new species of sulfate-reducing bacteria.  相似文献
