Heavy metals are required by all organisms for normal function, but high levels of heavy metals are toxic. Therefore, homeostasis of these metals is crucial. In the human malaria-causing agent Plasmodium falciparum, the mechanisms of heavy metal transport have yet to be characterized. We have developed a P. falciparum line resistant to heavy metals from a wild-type line sensitive to heavy metals. A molecular and biochemical analysis of the involvement of the P. falciparum multidrug resistance 2 (pfmdr2) gene, an ABC-type transporter, in heavy metal homeostasis was studied. Using a novel uptake assay applied on these two strains, it was demonstrated that, when exposed to heavy metals, the sensitive line accumulates metal, whereas no accumulation was observed in the resistant line. The accumulation occurs within the parasite itself and not in the cytoplasm of the red blood cell. This difference in the accumulation pattern is not a result of amplification of the pfmdr2 gene or of a change in the expression pattern of the gene in the two lines. Sequencing of the gene from both lines revealed a major difference; a stop codon is found in the sensitive line upstream of the normal termination, resulting in a truncated protein that lacks 188 amino acids that contain a portion of the essential cytoplasmatic transporter domain, thereby rendering it inactive. In contrast, the resistant line harbors a full-length, active protein. These findings strongly suggest that the PFMDR2 protein acts as an efflux pump of heavy metals.  相似文献

Photosystem I reaction centers of the cyanobacterium Thermosynechococcus elongatus have been investigated using single-molecule spectroscopy. Single-molecule fluorescence emission spectra reveal a new fluorescence band located at 745 nm. Fluorescence polarization spectroscopy and fluorescence autocorrelation analysis show that only a few chlorophylls are responsible for the photoemission from the Photosystem I trimer at low temperature. Intersystem crossing parameters of the red pool chlorophylls have been determined via fluorescence autocorrelation measurements. The triplet yield of the red chlorophylls is strongly reduced in comparison to chlorophyll a in solution. Strong quenching of the triplet state indicates that the red chlorophylls are located in close contact to carotenoids.  相似文献

Our aim was to assess the differential effect of waist circumference on left-ventricular (LV) structural and functional alterations, in hypertensive males and females. One thousand seven hundred and eighty nine consecutive, nondiabetic, essential hypertensives (aged 55.8 +/- 13.5 years, 966 females), included in the 3H Study, an ongoing registry of hypertension-related-target-organ damage, were classified to obese and nonobese groups according to Adult Treatment Panel III criteria. All participants underwent complete echocardiographic study including LV diastolic function evaluation by means of conventional and tissue Doppler imaging (TDI) methods, averaging early and late diastolic mitral annular peak velocities (Em, Am, Em/Am) from four separate sites of measurement. Hypertensive obese women compared with nonobese exhibited significantly greater LV mass index and prevalence of LV hypertrophy (by 5.5 g/m(2), P = 0.003, and 8.8%, P = 0.005, respectively), while such differences were not present among men. Obese women compared to nonobese ones were accompanied by lower transmitral E/A (by 0.08, P < 0.001), TDI-derived Em/Am (by 0.12, P < 0.001), and higher E/Em ratio (by 0.8, P = 0.016). In contrast, hypertensive obese men compared to nonobese ones exhibited lower E and Em (by 0.04 m/s and 0.6 cm/s, both P < 0.05). A significant interaction between sex and abdominal obesity was observed only regarding TDI-derived Am and Em/Am. Furthermore, waist circumference was a predictor of E/A (beta = -0.097, P = 0.002) and Em/Am (beta = -0.116, P = 0.001), independently of body size, in females but not in males. The adverse effect of abdominal obesity on LV alterations is more pronounced among female hypertensives, suggesting that routine measurement of waist circumference provides additional information on cardiac phenotype especially in women.  相似文献

Nutrition fuels any activity performed by an organism and has been shown to affect its ability to withstand pathogens and parasites. Furthermore, animals over a wide range of taxa have been shown to exhibit a choice of foods and nutrients that are beneficial to their fitness. Saprophagous animals most often feed on microorganisms growing on dead organic matter rather than the organic matter itself. Various yeast species play an important role in both larval and adult nutrition of saprophagous Drosophila melanogaster. We hypothesised the dietary microbial species to affect life‐history traits of D. melanogaster, including the ability to fend offparasitoids, and larvae to prefer to devour those yeast species beneficial to their development and immunocompetence. Particular yeast species known to be associated with D. melanogaster could be shown to have a substantial influence on various larval and adult fitness traits including the ability to encapsulate eggs of the parasitoid wasp Asobara tabida. It also turned out that larvae chose to devour those yeast species which supported their ability to encapsulate parasitoid eggs. Which yeast species was preferred and had a beneficial impact on encapsulation ability, was subject to inter‐individual variability within the investigated population, hinting at the existence of an adaptive heritable variability regarding individual choice and salubriousness of food. The results suggest that the dietary microbial species of saprophagous insects may influence the resistance against parasitoid attacks and thus the outcome of the interaction between a saprophagous host and its parasitoids.  相似文献

Lymphomas originating from the lymphatic system comprise about 30 entities classified according to the World Health Organization (WHO). The histopathological diagnosis is generally considered difficult and prone to mistakes. Since non-random chromosomal translocations are specifically involved in different lymphoma entities, their detection will be increasingly important. Hence, a split-signal fluorescence in situ hybridisation (FISH) procedure would be helpful in discriminating the most difficult classifications. The Euro-FISH programme, a concerted action of nine European laboratories, has validated a robust, standardised protocol to improve the diagnostic approach on lymphoma entities. Therefore, 16 fluorescent probes and 10 WHO entities, supplemented with reactive cases, were selected. The results of the Euro-FISH programme show that all probes were correctly cytogenetically located, that the standardised protocol is robust, resulting in reliable results in approximately 90% of cases, and that the procedure could be implemented in every laboratory, bringing the relatively easy interpretation of split-signal probes within the reach of many pathology laboratories.  相似文献

Aggregation process affects the ability of Lactobacillus crispatus, a probiotic, to survive into the gastro-intestinal environment and to adhere to the intestinal mucosa. To elucidate mechanisms underlying this process, a comparative proteomic study was carried out on a wild type strain M247 and its spontaneous isogenic mutant Mu5, which had lost the aggregative phenotype. Results highlighted an overall lower amount of enzymes involved in carbohydrate transport and metabolism in strain M247 compared to strain Mu5, suggesting a reduction in the general growth rate, probably caused by nutrient limitation in cell aggregates, coherently with the phenotypic traits of the strains. Moreover, the up-regulation of a putative elongation factor Tu in the wild type M247 strain could suggest a role of this particular protein in the adhesion mechanism of L. crispatus.  相似文献

Populations at geographic range limits may frequently encounter changing biotic interactions as well as abiotic constraints. A conundrum in plant population biology, the frequent apparent overproduction of flowers, is thought to be a response by plant populations to unpredictable pollinator environments, such as those that may be expected at the range limit. We studied pollinator visitation rates and pollen limitation of fruit set in populations of Witheringia solanacea , a widespread bee-pollinated species. We compared two populations in the south and two in the north of Costa Rica, with each region encompassing a lower montane site at the upper elevational limit of the species range with small plant populations, and a premontane site where population densities were high. Populations in the south, where the climate was favorable, had higher pollinator visitation and higher natural fruit set, and showed only minor pollen limitation as compared with northern populations at windy sites near the Continental Divide. Lower montane populations had lower pollinator visitation rates than premontane populations but did not show more pollen limitation of fruit set. In general, pollen limitation was not inversely proportional to pollinator abundance but was higher where the climate is unpredictable.  相似文献

The aim of this paper was to propose a granulation mechanism in order to interpret all the experimental observations that arose during experiments on two UASB reactors, where one was supplemented with ferrous iron at a dose of 0.01 g Fe(2+) per g COD feed. This supplementation with ferrous iron allowed COD removal of more than 98% at a loading rate of 9 g COD/L per day, which was 24% higher than for the reactor not receiving the ferrous iron. Moreover, in the Fe(2+)-dosed reactor, a higher increase of the granule diameter was observed. Indeed, the granule diameter in the Fe(2+)-dosed reactor at the end of the experiments was 56% greater than that of the control reactor. This mechanism describes the course of anaerobic granule growth. The formation of the inorganic precipitate of ferrous sulphide constitutes the inert nuclei around which the biomass is attached. This initiates the formation of new granules.  相似文献

Platelet Activating Factor (PAF) is a bioactive phospholipid, which exhibits a variety of biological activities and plays a significant role in all aspects of reproduction. In this work, a single intravenous injection of various concentrations of PAF shortly after Human Chorionic Gonadotropin (HCG) administration as well as 24 and 48 h before HCG administration was studied in NZB x NZW F1 hybrid mice. Optimum results were observed when PAF was injected just after the administration of HCG. In this protocol, the concentrations of PAF exhibited bell-shaped response to every stage of development. Any concentration of PAF between 5.5 x 10(-11) and 5.5 x 10(-15)g/g b.w., caused an improved ovulation rate, an increased fertilization rate, an increased rate of cell cycle and an enhanced hatching blastocyst rate (P<0.05 for all stages). Injection of lyso-PAF had no effect in any stage. Our data show that the effect of PAF on early stages of embryo development in vitro is dependent on its way of administration, on the concentrations used as well as on the time PAF is injected.  相似文献
