An artificial photosynthesis system based on N‐doped ZnTe nanorods decorated with an N‐doped carbon electrocatalyst layer is fabricated via an all‐solution process for the selective conversion of CO2 to CO. Substitutional N‐doping into the ZnTe lattice decreases the bandgap slightly and improves the charge transfer characteristics, leading to enhanced photoelectrochemical activity. Remarkable N‐doping effects are also demonstrated by the N‐doped carbon layer that promotes selective CO2‐to‐CO conversion instead of undesired water‐to‐H2 reduction by providing active sites for CO2 adsorption and activation, even in the absence of metallic redox centers. The photocathode shows promising performance in photocurrent generation (?1.21 mA cm?2 at ?0.11 VRHE), CO selectivity (dominant CO production of ≈72%), minor H2 reduction (≈20%), and stability (corrosion suppression). The metal‐free electrocatalyst/photocatalyst combination prepared via a cost‐effective solution process exhibits high performance due to synergistic effects between them, and thus may find application in practical solar fuel production. 相似文献

In eastern and southern part of India Dregea volubilis (Family Asclepediaceae) is widely used as anthelmintic in traditional system of medicine. The present study was conducted to evaluate the fasciocidal activity of the methanol extract of D. volubilis leaves (MEDV) and to observe the drug’s effect on organisms through scanning electron microscopic (SEM) study. Live parasites (Trematode: Fasciola gigantica) were collected in 0.9% phosphate-buffered saline from the bile ducts of buffalo. Those were incubated in the said media at 37 ± 1 °C either as control, or with MEDV at 5, 10, 25, 50 and 100 mg/ml as test groups or with albendazole at 10 mg/ml as standard group. The efficacy of the extract was determined on the basis of paralysis (temporary loss of spontaneous movement of the organisms) and/or death of the liver flukes. Death was confirmed when the organisms lost their motility permanently and their motility could not be revived even when vigorously shaken or dipped in warm water. MEDV at all concentration effectively paralyzed first and then killed the liver flukes (p < 0.001). Maximum fasciocidal activity was found with concentration of 100 mg/ml at 38.83 ± 3.41 min. Through SEM study, severe damages were observed in both the suckers as well as on the tegumental surfaces of the treated liver flukes. The study confirmed the fasciocidal activity of the MEDV. 相似文献

11-β hydroxysteroid dehydrogenase (11-βHSD1), tumor necrosis factor-α (TNF-α) and their role in obesity, regional adiposity and insulin resistance has been sparsely evaluated. We determined the polymorphic status of 11-βHSD1 4478T>G and TNF-α-308G>A in Asian Indians in north India. In this cross-sectional study (n = 498; 258 males, 240 females), association of genotypes (PCR–RFLP) of 11-βHSD1 and TNF-α were analyzed with obesity [BMI ≥ 25 kg/m2, percentage body fat (%BF by DEXA); subcutaneous and intra-abdominal fat area (L2–3 level by single slice MRI) in a sub sample] and insulin resistance. 46 percent subjects had generalized obesity, 55 % abdominal obesity and 23.8 % were insulin resistant. Frequencies (%) of [T/T] and [T/G] genotypes of 11-βHSD1 were 89.57 and 10.43 respectively. Homozygosity for 11-βHSD1 4478G/G was absent with no association with parameters of obesity and insulin resistance. Frequencies (%) of TNF-α [G] and [A] alleles were 88 and 12 respectively. Higher frequency of variant -308[A/A] was observed in females versus males (p = 0.01). Females with at least one single A allele of TNF-α-308G>A had significantly high %BF and total skinfold, whereas higher values of waist hip ratio, total cholesterol, triglycerides and VLDL were observed in males. Subjects with even a single A allele in TNF-α genotype showed higher subscapular skinfold predisposing them to truncal subcutaneous adiposity (p = 0.02). Our findings of association of TNF-α-308G>A variant in females with obesity indices suggests a gender-specific role of this polymorphism in obesity. High truncal subcutaneous adiposity is associated with A allele of TNF-α-308G>A in this population. 相似文献

Abstract A study of the temporal synergism of serotonergic and dopaminergic activity in the regulation of reproductive seasonality of photosensitive and photorefractory migratory Red headed bunting was undertaken. In experiment 1, groups of birds kept in natural day length (NDL) in their photosensitive phase (April) received daily injections of L‐DOPA (L‐dihydroxyphenylalanine, a dopamine precursor) at 8 and 12 hr after 5‐HTP (5 hydroxytryptophan, a serotonin precursor) administration. One control group received two daily injections of saline. The injections with 12‐hr temporal relation induced early recrudescence and full gonadal growth was achieved much in advance to that of control birds. The applications with 8‐hr temporal relation suppressed the testicular growth initially but later on this effect was eliminated by NDL of May‐June. In Experiment 2, photorefractory birds also received the treatments but the temporal relation of 5‐HTP and L‐DOPA had no effect on the regressed gonads of these birds. This study indicates that injections with specific phase relations of dopamine and serotonin precursors may alter seasonal reproductive conditions only in the photosensitive phase but had no effect in photorefractory birds. 相似文献

Hereditary Hemochromatosis is an iron overload disease most frequently associated with mutations in the HFE gene. While clinical studies of the disease have received extensive attention by various groups, the localisation, trafficking and function of the HFE protein, and its chaperone beta2-microglobulin (beta2M), require further investigation. In this study, we present data on the cellular localisation of HFE and its clinically relevant mutants in HuTu 80 cells. We find by confocal microscopy that HFE localises to the endosomal-recycling compartment (ERC), with minimal localisation to sorting or late endosomes. Interestingly, we also demonstrate that beta2M localises to the ERC where it co-localises with HFE. We find that exogenous expression of HFE results in enhanced beta2M cellular levels and that beta2M is necessary for cell surface expression of HFE. Finally, we have analysed the functional effects of exogenous expression of HFE and beta2M on transferrin binding to the cell surface. In summary, our study sheds light on the localisation and functional effects of the HFE and its chaperone protein beta2M. 相似文献

Glutathione S-transferases play an important role in cellular detoxification and may have evolved to protect cells against reactive oxygen metabolites. In this study, we report the molecular characterization of glutathione s-transferase-theta (GST-θ) from freshwater prawn Macrobrachium rosenbergii. A full length cDNA of GSTT (1417 base pairs) was isolated and characterized bioinformatically. Exposure to virus (white spot syndrome baculovirus or M. rosenbergii nodovirus), bacteria (Aeromonas hydrophila or Vibrio harveyi) or heavy metals (cadmium or lead) significantly increased the expression of GSTT (P < 0.05) in hepatopancreas. Recombinant GST-θ with monochlorobimane substrate had an optimum activity at pH 7.5 and 35 °C. Furthermore recombinant GST-θ activity was abolished by the denaturants triton X-100, Gua-HCl, Gua-thiocyanate, SDS and urea in a dose-dependent manner. Overall, the results suggest a potential role for M. rosenbergii GST-θ in detoxification and possibly conferring immune protection. 相似文献

Rationale
Given that the diagnosis of chronic obstructive pulmonary disease (COPD) relies on demonstrating airflow limitation by spirometry, which is known to be poorly sensitive to early disease, and to regional differences in emphysema, we sought to evaluate individual lobar contributions to global spirometric measures.Methods
Subjects with COPD were compared with smokers without airflow obstruction, and non-smokers. Emphysema (% low attenuation area, LAAinsp<−950 HU, at end-inspiration) and gas trapping (%LAAexp<−856 HU at end-expiration) on CT were quantified using density mask analyses for the whole lung and for individual lobes, and distribution across lobes and strength of correlation with spirometry were compared.Results
The right middle lobe had the highest %LAAinsp<−950 HU in smokers and controls, and the highest %LAAexp<−856 HU in all three groups. While RML contributed to emphysema and gas trapping disproportionately to its relatively small size, it also showed the least correlation with spirometry. There was no change in correlation of whole lung CT metrics with spirometry when the middle lobe was excluded from analyses. Similarly, RML had the highest %LAAexp<−856 HU while having the least correlation with spirometry.Conclusions
Because of the right middle lobe’s disproportionate contribution to CT-based emphysema measurements, and low contribution to spirometry, longitudinal studies of emphysema progression may benefit from independent analysis of the middle lobe in whole lung quantitative CT assessments. Our findings may also have implications for heterogeneity assessments and target lobe selection for lung volume reduction.Clinical Trial Registration

The human pathogen Mycobacterium tuberculosis is the causative agent of pulmonary tuberculosis (TB), a disease with high worldwide mortality rates. Current treatment programs are under significant threat from multi-drug and extensively-drug resistant strains of M. tuberculosis, and it is essential to identify new inhibitors and their targets. We generated spontaneous resistant mutants in Mycobacterium bovis BCG in the presence of 10× the minimum inhibitory concentration (MIC) of compound 1, a previously identified potent inhibitor of mycobacterial growth in culture. Whole genome sequencing of two resistant mutants revealed in one case a single nucleotide polymorphism in the gene aspS at 535GAC>535AAC (D179N), while in the second mutant a single nucleotide polymorphism was identified upstream of the aspS promoter region. We probed whole cell target engagement by overexpressing either M. bovis BCG aspS or Mycobacterium smegmatis aspS, which resulted in a ten-fold and greater than ten-fold increase, respectively, of the MIC against compound 1. To analyse the impact of inhibitor 1 on M. tuberculosis AspS (Mt-AspS) activity we over-expressed, purified and characterised the kinetics of this enzyme using a robust tRNA-independent assay adapted to a high-throughput screening format. Finally, to aid hit-to-lead optimization, the crystal structure of apo M. smegmatis AspS was determined to a resolution of 2.4 Å. 相似文献

Soil samples isolated from dye-contaminated sites were exploited for isolation of dye decolorizing microorganisms. A novel bacterial mixture, RkNb1, was selected based on its efficiency, showing maximum and faster decolorization of textile dyes. Seven bacterial strains were isolated and identified from the bacterial mixture as Ochrobactrum intermedium (HM480365), Ochrobactrum intermedium strain M16-10-4 (HM030758), Enterococcus faecalis (HM480367), Arthrobacter crystallopoietes (HM480368), Kocuria flavus (HM480369), Bacillus beijingensis (HM480370), and Citrobacter freundii (HM480371) by 16S rRNA gene sequence analysis. This bacterial mixture showed 98.17% decolorization of Reactive Violet 5 (400 mg L?1) within 8 h. The culture exhibited good decolorization ability at pH 8 and at a temperature of 37°C. Malt extract and peptone was found to enhance the decolorization rate of Reactive Violet 5. Plackett-Burman experimental design was used for elucidation of medium components affecting Reactive Violet 5 decolorization. Dye degradation products obtained during the course of decolorization were analyzed by high-performance thin-layer chromatography (HPTLC), Fourier transform infrared (FTIR), and nuclear magnetic resonance (NMR). The potential of this bacterial mixture to decolorize Reactive Violet 5 dye from manufacturing industry effluent is to be carried out using appropriate bioreactors. 相似文献
