Summary Wild strains of Saccharomyces cerevisiae were isolated from dairy products, bakery goods, fresh fruit and vegetables, and tested for killer activity. Four isolates out of 238 strains possessed killer activity. The best of these was converted to the petite form and hybridized with an industrial strain of Saccharomyces cerevisiae by protoplast fusion. Thirty-eight out of 104 isolates had killer activity, and some of these had good dough-raising activity as well. 相似文献

distributed into three groups of 10 animals each and treated with 4-nitroquinoline 1-oxide solution at 50 ppm through their
drinking water for 4, 12, and 20 weeks. Ten animals were used as control group. No histopathological abnormalities were induced
in the epithelium after 4 weeks of carcinogen exposure; however, immunoexpression of MMP-2 was noticed. The same picture occurred
to MMP-9, in which positive expression was detected for this immunomarker. MMP-2 and MMP-9 showed positive expression either
in pre-neoplastic lesions at 12 weeks following carcinogen exposure or in well-differentiated squamous cell carcinoma induced
after 20 weeks of treatment with 4NQO. Taken together, our results support the belief that MMP-2 and MMP-9 play important

Aspergillus terreus, Cladosporium cladosporioides, Fusarium
oxysporum, Gliocladium roseum, Penicillium spp., Talaromyces helicus and Trichoderma
koningii were isolated from heavily polluted streams near an industrial area in La Plata, Argentina. The fungi were obtained from sediments with 0.25–0.50 mg Cd/l and they were isolated in cadmium-basal medium. They were then cultivated to evaluate their Cd detoxification abilities. The biomass developed in static assays represented 5–53% of the yield of stirred cultures, for the different fungal species, although the cadmium absorption were similar in both cases. These soil fungi represented 50% of the total isolates and their mycelial growth was conspicuous in these polluted sediments. Although bacteria have been mentioned as active microorganisms against heavy metals, the mycelial fungi were able to develop a significantly higher mass to sequestrate more metals. Thus, they could be used in remediation biotechnology to improve the Cd detoxification of chronically contaminated habitats. 相似文献

The renal renin-angiotensin system plays a central role in the development of hypertension. The aim of this work was to verify the expression of angiotensin II receptors AT(1)R and AT(2)R in the microsomal fraction of renal cortex and correlate this with the development of hypertension and renal damage in spontaneously hypertensive rats (SHR) using Wistar-Kyoto rats (WKY) as controls. AT(1)R expression increased (126%) and AT(2)R expression decreased (66%) in 4-wk-old SHR; AT(2) expression decreased in 14-wk-old SHR (61%) compared with respective age-matched WKY. These modifications were correlated to the increase in protein kinase C activity and decrease in protein kinase A activity. Four-week-old SHR showed large accumulations of macrophages in kidney glomerulus and the tubulointerstitial area, dense cortical collagen deposition, and arterial proliferative changes in the walls of arterioles and medium-sized vessels. Similar modifications were also observed in 14-wk-old SHR. Four-week-old SHR treated with losartan (30 mg·kg(-1)·day(-1)) or hydralazine (15 and 30 mg·kg(-1)·day(-1)) by gavage for 10 wk did not develop hypertension. The decrease in AT(2)R expression and renal damage observed in SHR remained even after treatment with hydralazine. On the other hand, losartan treatment prevented the modifications observed in 14-wk-old SHR, indicating that renal injuries are caused specifically by AT(1) rather than an increase in blood pressure. Our results indicate that the imbalance in AT(1)R and AT(2)R expression is associated with an inflammatory process that contributes to renal injury in adult SHR and to the development of hypertension. 相似文献

Adipose tissue is considered an endocrine organ that promotes excessive production of reactive oxygen species when in excess, thus contributing to lipid peroxidation. Magnesium deficiency contributes to the development of oxidative stress in obese individuals, as this mineral plays a role as an antioxidant, participates as a cofactor of several enzymes, maintains cell membrane stability and mitigates the effects of oxidative stress. The objective of this review is to bring together updated information on the participation of magnesium in the oxidative stress present in obesity. We conducted a search of articles published in the PubMed, SciELO and LILACS databases, using the keywords ‘magnesium’, ‘oxidative stress’, ‘malondialdehyde’, ‘superoxide dismutase’, ‘glutathione peroxidase’, ‘reactive oxygen species’, ‘inflammation’ and ‘obesity’. The studies show that obese subjects have low serum concentrations of magnesium, as well as high concentrations of oxidative stress marker in these individuals. Furthermore, it is evident that the adequate intake of magnesium contributes to its appropriate homeostasis in the body. Thus, this review of current research can help define the need for intervention with supplementation of this mineral for the prevention and treatment of disorders associated with this chronic disease. 相似文献

The recombinant dihydropyrimidinase from Sinorhizobium meliloti CECT4114 (SmelDhp) has been characterised and its crystal structure elucidated at 1.85 Å. The global architecture of the protein is reminiscent of that of the amidohydrolase superfamily, consisting of two domains; an (α/β)8 TIM-like barrel domain, where the catalytic centre is located, and a smaller β-sheet sandwich domain of unknown function. The c-terminal tails of each subunit extend toward another monomer in a swapping-like manner, creating a hydrogen bond network which suggests its implication in protein oligomerisation. Mutational and structural evidence suggest the involvement of a conserved tyrosine in the reaction mechanism of the enzyme. SmelDhp presents both hydantoinase and dihydropyrimidinase activities, with higher affinity for the natural six-membered ring substrates. For the five-membered ring substrates, affinity was greater for those with aliphatic and apolar groups in the 5th carbon atom, with the highest rates of hydrolysis for d-5-methyl and d-5-ethyl hydantoin (kcat/Km = 2736 ± 380 and 944 ± 52 M?1 s?1, respectively). The optimal conditions for the enzyme activity were found to be 60 °C of temperature at pH 8.0. SmelDhp retains 95% of its activity after 6-hour preincubation at 60 °C. This is the first dihydropyrimidinase used for the hydrolytic opening of non-natural 6-monosubstituted dihydrouracils, which may be exploited for the production of β-amino acids. 相似文献

Oxidative stress is widely recognized as an important mediator of apoptosis in liver cells and plays a pivotal role in the pathogenesis of several diseases. Cocoa flavonoids have shown a powerful antioxidant activity providing protection against oxidation and helping prevent oxidative stress-related diseases. However, the molecular mechanisms responsible for this protection are not fully understood. Thus, in this study we investigated the protective effect of a cocoa polyphenolic extract (CPE) against tert-butyl hydroperoxide (t-BOOH)-induced apoptosis and the molecular mechanisms involved in this process. Incubation of HepG2 cells with t-BOOH induced apoptosis as evidenced by caspase-3 activation. This effect was accompanied by increased reactive oxygen species formation and by transient activation of the extracellular regulated kinases (ERKs) as well as sustained activation of the c-Jun N-terminal kinases (JNKs). On the contrary, pretreatment of HepG2 cells with CPE prevented apoptosis through the reduction of reactive oxygen species generation and the modulation of the apoptotic pathways activated by t-BOOH. CPE treatment also activated survival signaling proteins, such as protein kinase B (AKT) and ERKs, and increased the activities of two antioxidant enzymes, glutathione peroxidase (GPx) and glutathione reductase (GR). ERK's implication on GPx and GR induction and the protective effect of CPE against t-BOOH-induced oxidative stress and apoptosis were confirmed through experiments with selective inhibitors. These findings suggest that CPE is an effective inductor of GPx and GR activities via ERK activation and that this up-regulation seems to be required to attenuate t-BOOH-induced injury. 相似文献

The developmental profiles of the neurotransmitter recognition site, labeled by [3H]muscimol, and of the two main modulatory sites, labeled by [3H]flunitrazepam and [35S]t-butylbicyclophosphorothionate, respectively, within the GABAA receptor complex, have been determined in chick tectal lobes between embryonic day 8 and postnatal day 20. The consonance among the rates of appearance and accumulation of the three receptor sites in tectal membranes suggests a coordinated expression and assembly of the protein subunits involved in the spatial configuration of the complex and its three binding sites, although the existence of isolated muscimol binding subunits during early embryogenesis cannot be excluded at the present time. Furthermore, the total number of binding sites of each kind, per pair of lobes, is compatible with a 1/1/I stoichiometry. The GABAA receptor complex reaches a maximum of expression, relative to total membrane protein, immediately after hatching, suggesting that the tectal GABAergic system may be instrumental in damping the effects of sudden exposure to light of the chick visual system upon eye opening. 相似文献

Aim To determine whether the cover and growth habit of the main forest forming species (Polylepis australis BITT.) in a mountain range with low human population density is mainly affected by anthropogenic activities or by environmental influences. Location Central Argentina. Methods Using GIS and field surveys we established 146 plots of 30 × 30 m located in five river basins differing in human impact. We measured P. australis cover, growth habit of each individual (number of basal ramifications), index of long term human impact (percentage of rock exposed by soil erosion due to livestock and fires), evidence of logging, fire scars, local relief, percentage of rock outcrops and altitude above sea level. We analysed the influence of independent variables on P. australis cover and growth habit (average number of basal ramifications per plot) using correlations and General Linear Models. Results Polylepis australis cover was greater at intermediate altitudes above sea level and in areas with reduced long term human impact. Contrastingly local relief, percentage of rock outcrops and logging in the recent past did not have a major influence on P. australis abundance. Growth habit varied in complex patterns. Individuals with fewer ramifications were found in valley bottoms and more disturbed basins, while more ramifications were found at mid‐ and upper slopes and well preserved basins. In valley bottoms, ramifications decreased with increasing altitude whereas the opposite trend was observed for mid‐slopes. Ramifications were positively related to fires in two river basins and in mid‐ and upper slopes but not in valley bottoms. Fire impact was always less in valley bottoms than at mid‐ and upper slopes. Main conclusions Human impact had a major role on P. australis cover, while growth habit was determined by complex combinations of potentially cumulative natural and anthropogenic factors. Even in sparsely populated mountains, both human impact and their interaction with natural environmental gradients influence plant communities and need to be understood for effective management. 相似文献
