Organoseleno-compounds have been investigated for its beneficial effects against methylmercury toxicity. In this way, diphenyl diselenide (PhSe)2 was demonstrated to decrease Hg accumulation in mice, protect against MeHg-induced mitochondrial dysfunction, and protect against the overall toxicity of this metal. In the present study we aimed to investigate if co-treatment with (PhSe)2 and MeHg could decrease accumulation of Hg in liver slices of rats. Rat liver slices were co-treated with (PhSe)2 (0.5; 5 µM) and/or MeHg (25 µM) for 30 min at 37 °C and Se and Hg levels were measured by inductively coupled plasma mass spectrometry (ICP-MS) in the slices homogenate, P1 fraction, mitochondria and incubation medium. Co-treatment with (PhSe)2 and MeHg did not significantly alter Se levels in any of the samples when compared with compounds alone. In addition, co-treatment with (PhSe)2 and MeHg did not decrease Hg levels in any of the samples tested, although, co-incubation significantly increased Hg levels in homogenate. We suggest here that (PhSe)2 could exert its previously demonstrated protective effects not by reducing MeHg levels, but forming a complex with MeHg avoiding it to bind to critical molecules in cell. 相似文献

Many animal taxa that display sexual size dimorphism (SSD) exhibit a positive allometric relationship in which the degree of dimorphism increases with body size. This macroevolutionary pattern is known as Rensch's rule. Although sexual selection is hypothesized to be the main mechanism causing this pattern, body size is influenced by several selective forces, including natural and sexual selection. Therefore, by focusing exclusively on SSD one cannot ascertain which of these selective forces drives Rensch's rule. If sexual selection is indeed the main mechanism underlying Rensch's rule, we predict that other sexually selected traits, including coloration‐based ornaments, will also exhibit interspecific allometric scaling consistent with Rensch's rule. We tested this prediction using wing pigmentation of 89 species of dragonflies. Studies show that male wing pigmentation is generally under strong intra‐ and intersexual selection, so that sexual dichromatism in this trait should follow Rensch's rule. Conversely, the available evidence suggests that male body size is usually not sexually selected in dragonflies, so we do not expect SSD to follow Rensch's rule. First, we found that sexual dichromatism in wing pigmentation was consistent with Rensch's rule. The phylogenetic major axis regression slope was significantly greater than one. We also showed that the allometric slope for SSD was not different from unity, providing no support for Rensch's rule. Our results provide the first evidence that a trait which appears to be under strong sexual selection exhibits a pattern consistent with Rensch's rule. 相似文献

The aim of this study was to assess stress/strain of different implant modeling simplifications by 3D-FEA. Three variation of external hexagon implant (Ø3.75?×?10 mm) supporting one molar crown were simulated: A (no threads); B (slightly threads simplification); C (original design). 200 N (axial) and 100 N (oblique) were applied. Cortical bone was evaluated by maximum principal stress and microstrain qualitatively and quantitatively (ANOVA and Tukey post hoc (p < 0.05)). Higher stress levels (p < 0.05) were observed in model A. Models B and C presented similar stress transmission. It was possible to conclude that slightly simplification should be used for studies evaluating stress transferring for bone tissue. 相似文献

Effective ecological restoration actions should be able to recover ecosystem processes that influence community development in the long term. However, there is scarce information on how landscape factors promote or accelerate fauna recovery. We used a landscape framework to evaluate how functional groups respond to natural regeneration in a highly fragmented region of Atlantic Forest. Using bird functional groups sampled in 15 regenerating forest fragments, we built and ranked models using a model selection approach to test the relative effect of landscape variables on each group. Our results showed that bird community recovery is not determined by the duration of the regeneration process (i.e. forest age), but by how the species responds to the landscape context. Functional diversity and the abundance of the functional groups were mainly related to composition metrics, whereas the functional metric affected only specific groups. Our findings highlight the importance of considering the landscape level to ensure both the colonization of fauna and the restoration of ecological functions. 相似文献

Intensive land use of the Brazilian Atlantic Forest accelerated with the rise of sugar cane plantations in the northeastern part of Brazil. Consequently, many ecosystems were destroyed, including riparian forests. The number of studies of riparian restoration has increased but comparative studies on the belowground effects of common reforestation strategies are rare. Here, we compared soil microbial properties among four different land use types: native rainforest, sugar cane plantation, single species reforestation, and mixed species reforestation, each replicated at two spatially independent sites. Soil samples were taken in 2013 and 2014, that is 2 and 3 years after reforestation, respectively. In both years, land use types had a significant effect on basal respiration, microbial biomass, and specific respiration (whereas specific respiration was marginally affected in 2014). In 2013, basal respiration in sugar cane plantations was significantly lower (?65%) when compared to native forests. In 2014, basal respiration (+60%) and soil microbial biomass (+90%) were significantly higher in mixed species reforestation compared to sugar cane, whereas single species reforestation had comparable values as in sugar cane plantations. Our results indicate that soil microbial biomass and activity respond rapidly to land use change when mixed species reforestation is used. Thus, using mixed species reforestation may enhance the provisioning of ecosystem services already in the short term. 相似文献

The intercellular communication mediated by extracellular vesicles (EVs) has gained international interest during the last decade. Interfering with the mechanisms regulating this cellular process might find application particularly in oncology where cancer cell‐derived EVs play a role in tumour microenvironment transformation. Although several mechanisms were ascribed to explain the internalization of EVs, little is our knowledge about the fate of their cargos, which are crucial to mediate their function. We recently demonstrated a new intracellular pathway in which a fraction of endocytosed EV‐associated proteins is transported into the nucleoplasm of the host cell via a subpopulation of late endosomes penetrating into the nucleoplasmic reticulum. Silencing tetraspanin CD9 both in EVs and recipient cells strongly decreased the endocytosis of EVs and abolished the nuclear transfer of their cargos. Here, we investigated whether monovalent Fab fragments derived from 5H9 anti‐CD9 monoclonal antibody (referred hereafter as CD9 Fab) interfered with these cellular processes. To monitor the intracellular transport of proteins, we used fluorescent EVs containing CD9‐green fluorescent protein fusion protein and various melanoma cell lines and bone marrow‐derived mesenchymal stromal cells as recipient cells. Interestingly, CD9 Fab considerably reduced EV uptake and the nuclear transfer of their proteins in all examined cells. In contrast, the divalent CD9 antibody stimulated both events. By impeding intercellular communication in the tumour microenvironment, CD9 Fab‐mediated inhibition of EV uptake, combined with direct targeting of cancerous cells could lead to the development of novel anti‐melanoma therapeutic strategies. 相似文献
