Blood, kidney and tick samples were obtained from 18 hunter-killed black bears (Ursus americanus) from three sites in northern Wisconsin. A Borrelia sp., morphologically and antigenically similar to Borrelia burgdorferi, was isolated from the blood of two of the animals, and from the kidney of a third. Ixodes dammini and Dermacentor variabilis were found on the bears. This is the first report of borreliosis in the Ursidae, and of the primary vector of Lyme disease, I. dammini, from this host.  相似文献

A specialized vector backbone from the Protein Structure Initiative was used to express full-length human cytochrome b5 as a C-terminal fusion to His8-maltose binding protein in Escherichia coli. The fusion protein could be completely cleaved by tobacco etch virus protease, and a yield of 18 mg of purified full-length human cytochrome b5 per liter of culture medium was obtained (2.3 mg per g of wet weight bacterial cells). In situ proteolysis of the fusion protein in the presence of chemically defined synthetic liposomes allowed facile spontaneous delivery of the functional peripheral membrane protein into a defined membrane environment without prior exposure to detergents or other lipids. The utility of this approach as a delivery method for production and incorporation of monotopic (peripheral) membrane proteins is discussed.  相似文献

A summary is provided of presentations and discussions at the NASA Radiation Biomarker Workshop held September 27-28, 2007 at NASA Ames Research Center in Mountain View, CA. Invited speakers were distinguished scientists representing key sectors of the radiation research community. Speakers addressed recent developments in the biomarker and biotechnology fields that may provide new opportunities for health-related assessment of radiation-exposed individuals, including those exposed during long-duration space travel. Topics discussed included the space radiation environment, biomarkers of radiation sensitivity and individual susceptibility, molecular signatures of low-dose responses, multivariate analysis of gene expression, biomarkers in biodefense, biomarkers in radiation oncology, biomarkers and triage after large-scale radiological incidents, integrated and multiple biomarker approaches, advances in whole-genome tiling arrays, advances in mass spectrometry proteomics, radiation biodosimetry for estimation of cancer risk in a rat skin model, and confounding factors. This study compared a conventional pull-up and chin-up with a rotational exercise using Perfect·Pullup? twisting handles. Twenty-one men (24.9 ± 2.4 years) and 4 women (23.5 ± 1 years) volunteered to participate. Electromyographic (EMG) signals were collected with DE-3.1 double-differential surface electrodes at a sampling frequency of 1,000 Hz. The EMG signals were normalized to peak activity in the maximum voluntary isometric contraction (MVIC) trial and expressed as a percentage. Motion analysis data of the elbow were obtained using Vicon Nexus software. One-factor repeated measures analysis of variance examined the muscle activation patterns and kinematic differences between the 3 pull-up exercises. Average EMG muscle activation values (%MVIC) were as follows: latissimus dorsi (117-130%), biceps brachii (78-96%), infraspinatus (71-79%), lower trapezius (45-56%), pectoralis major (44-57%), erector spinae (39-41%), and external oblique (31-35%). The pectoralis major and biceps brachii had significantly higher EMG activation during the chin-up than during the pull-up, whereas the lower trapezius was significantly more active during the pull-up. No differences were detected between the Perfect·Pullup? with twisting handles and the conventional pull-up and chin-up exercises. The mean absolute elbow joint range of motion was 93.4 ± 14.6°, 100.6 ± 14.5°, and 99.8 ± 11.7° for the pull-up, chin-up, and rotational exercise using the Perfect·Pullup? twisting handles, respectively. For each exercise condition, the timing of peak muscle activation was expressed as a percentage of the complete pull-up cycle. A general pattern of sequential activation occurred suggesting that pull-ups and chin-ups were initiated by the lower trapezius and pectoralis major and completed with biceps brachii and latissimus dorsi recruitment. The Perfect·Pullup? rotational device does not appear to enhance muscular recruitment when compared to the conventional pull-up or chin-up.  相似文献

The effect of climate and cultivation on soil organic C and N   总被引:6,自引:0,他引:6  
Here, we investigate the response of soil organic carbon (SOC) and soil organic nitrogen (SON) to cultivation within two different climatic regimes by comparing large soil data sets from India and the Great Plains. Multiple regression models for both regions show that SOC and SON, as well as C/N ratios, increase with decreasing temperatures and increasing precipitation, trends also noted in soil data organized by Holdridge life zones. The calculated difference between natural and cultivated soils in India revealed that the greatest absolute SOC and SON losses occurred in regions of low temperatures and high precipitation, while the C/N ratio decreased during cultivation only with decreasing temperature. In India, the fractional loss of SOC relative to undisturbed soils increases with decreasing temperature whereas, in the Great Plains, it increases with increasing precipitation. Also, the fractional loss of SOC increased in India with increasing amounts of original C, whereas no relationship between fractional loss and original C was noted for the Great Plains. The differential response of each region to cultivation is hypothesized to be due to differences in both climate and management practices (crop cycles, fertilization). These findings suggest that estimates of soil C loss due to cultivation should be based on an array of factors, and that it is unlikely that a constant relative C loss occurs in any region.  相似文献

The mechanism controlling G(2)/M checkpoint activation after DNA damage was thought to be mediated primarily by nuclear Chk1/Chk2 kinases. Recent evidence indicates that this checkpoint is more complex, involving at least two different biochemical systems that target the Cdc25B and Cdc25C phosphatases. Following genotoxic stress, different kinases integrate signaling from the damaged DNA and other damaged cellular components to regulate Cdc25 inactivation. Our current model for G(2)/M checkpoint activation after genotoxic stress is discussed emphasizing the roles for Chk1 and p38 kinases in checkpoint regulation.  相似文献

Effect of land use change on soil carbon in Hawaii   总被引:11,自引:3,他引:11  
Organic carbon storage and turnover were altered in soils formed from volcanic ash (Andisols) as a result of conversion of tropical forest to pasture and sugarcane cropland. Changes in soil carbon storage after approximately a century of each land use were estimated using stable carbon isotope values and carbon contents. Total organic carbon stored in soils varied as a result of management, with pasture soils showing net carbon gain and sugarcane soils showing net carbon loss. In pasture soils, increases in carbon at depth (40 to 80 cm) are below the rooting zone of the introduced (C4) vegetation, and have stable carbon isotopic values indicative of forest (C3) plants. Within the pasture rooting zone (0–40 cm) the isotopic data reveals that additions of pasture (C4) organic matter have been offset by losses of C3 carbon. The concentration of Fe/Al oxides (soil minerals that bind with organic matter to form oxide-humus complexes) appear to control the quantity of carbon stored in soils, as well as the difference in the depth and magnitude of carbon storage changes that occur with each type of land use change. Sugarcane land use appears to induce dissociation of Fe/Al oxide-humus complexes and loss of oxide-associated organic matter from the profile. In pastures, Fe/Al oxide-humus complexes are translocated to deeper horizons in the soils, resulting in greater profile carbon storage and longer apparent turnover time of carbon stored below 50 cm depth. In this high precipitation region, carbon losses from the soil appear to occur via downward transport, either as colloids or in solution, in addition to the generally assumed pathway of flux to the atmosphere as CO2.  相似文献
