A digital imaging system was developed for measuring various physical characteristics of individual nematodes and for comparing groups of nematodes. The equipment consists of a microscope, a video camera, a video digitizer, interactive displays, and a computer. Various physical and mathematical methods were incorporated, algorithms devised, and computer software written for image acquisition, editing, and analysis. To test the system, four populations of an isolate of the pinewood nematode, Bursaphelenchus xylophilus, subjected to 100% relative humidity at 22 C for 0, 12, 24, or 48 hours were compared. The results showed that the system can be used to measure physical parameters of individual nematodes and to differentiate groups of nematodes.  相似文献

The alkaloids present in the seeds of four Erythrina species, E. brucei, E. cochleata, E. thollonia and E. caribaea have been screened by GC/MS. A new alkaloid, erythrocarine, has been isolated from E. caribaea and characterized as a methylenedioxy analogue of the known dienoid alkaloids erysoline and erysonine.  相似文献

The aromatase and estrone sulfatase enzymes are important sources of biologically active estrogens in postmenopausal women with breast cancer. Promising initial results in the treatment of endocrine-responsive breast cancer have been exhibited by 125-dihydroxyvitamin D3 and the synthetic vitamin D analogues MC903 and EB1089. However, these compounds together with vitamin D3 and vitamin D3 sulfate did not inhibit the human placental aromatase enzyme when assayed up to 20 μm. Only vitamin D3 sulfate and 125-dihydroxyvitamin D inhibited the estrone sulfatase activity in human placental microsomes, albeit at high concentration (32 and 37% inhibition, respectively with 50 μm each inhibitor). It is unlikely that inhibition of aromatase or estrone sulfatase enzymes contribute to the inhibitory effect of this group of compounds on breast cancer cells in vivo.  相似文献

The capacity of tumour cells to maintain continual overgrowth potential has been linked to the commandeering of normal self-renewal pathways. Using an epithelial cancer model in Drosophila melanogaster, we carried out an overexpression screen for oncogenes capable of cooperating with the loss of the epithelial apico-basal cell polarity regulator, scribbled (scrib), and identified the cell fate regulator, Abrupt, a BTB-zinc finger protein. Abrupt overexpression alone is insufficient to transform cells, but in cooperation with scrib loss of function, Abrupt promotes the formation of massive tumours in the eye/antennal disc. The steroid hormone receptor coactivator, Taiman (a homologue of SRC3/AIB1), is known to associate with Abrupt, and Taiman overexpression also drives tumour formation in cooperation with the loss of Scrib. Expression arrays and ChIP-Seq indicates that Abrupt overexpression represses a large number of genes, including steroid hormone-response genes and multiple cell fate regulators, thereby maintaining cells within an epithelial progenitor-like state. The progenitor-like state is characterised by the failure to express the conserved Eyes absent/Dachshund regulatory complex in the eye disc, and in the antennal disc by the failure to express cell fate regulators that define the temporal elaboration of the appendage along the proximo-distal axis downstream of Distalless. Loss of scrib promotes cooperation with Abrupt through impaired Hippo signalling, which is required and sufficient for cooperative overgrowth with Abrupt, and JNK (Jun kinase) signalling, which is required for tumour cell migration/invasion but not overgrowth. These results thus identify a novel cooperating oncogene, identify mammalian family members of which are also known oncogenes, and demonstrate that epithelial tumours in Drosophila can be characterised by the maintenance of a progenitor-like state.  相似文献

The present study shows the feasibility of a newly isolated strain Acinetobacter sp. B9 for concurrent removal of phenol and Cr (VI) from wastewater. The experiments were conducted in a batch reactor under aerobic conditions. Initially, when mineral salt solution was used as the culture medium, the strain was found to utilize phenol as sole carbon and energy source while no Cr (VI) removal was observed. However, the addition of glucose as co-carbon source resulted in the removal of both toxicants. This co-removal efficiency of the strain was further improved with nutrient-rich media (NB). Optimum co-removal was determined at 188 mg L?1 of phenol and 3.5 mg L?1 of Cr (VI) concentrations at pH 7.0. Strain B9 followed the orthometabolic pathway for phenol degradation. Transmission electron microscopy (TEM) and Fourier transform infrared spectroscopy (FT-IR) studies showed sorption of chromium as one of the major mechanisms for Cr (VI) removal by B9 cells. Acinetobacter sp. B9 was later on checked for bioremediation of real tannery wastewater. After 96 h of batch treatment of tannery effluent containing an initial 47 mg L?1 phenol and 16 mg L?1 Cr (VI), complete removal of phenol and 87 % reduction of Cr (VI) were attained, showing high efficiency of the bacterial strain for potential application in industrial pollution control.  相似文献

Endothelial cell migration is critical for vascular angiogenesis and is compromised to facilitate tumor metastasis. The migratory process requires the coordinated assembly and disassembly of focal adhesions (FA), actin, and microtubules (MT). MT dynamics at FAs deliver vesicular cargoes and enhance actomyosin contractility to promote FA turnover and facilitate cell advance. Noncentrosomal (NC) MTs regulate FA dynamics and are sufficient to drive cell polarity, but how NC MTs target FAs to control FA turnover is not understood. Here, we show that Rac1 induces the assembly of FA-proximal septin filaments that promote NC MT growth into FAs and inhibit mitotic centromere-associated kinesin (MCAK)-associated MT disassembly, thereby maintaining intact MT plus ends proximal to FAs. Septin-associated MT rescue is coupled with accumulation of Aurora-A kinase and cytoplasmic linker-associated protein (CLASP) localization to the MT between septin and FAs. In this way, NC MTs are strategically positioned to undergo MCAK- and CLASP-regulated bouts of assembly and disassembly into FAs, thereby regulating FA turnover and cell migration.  相似文献

RNA synthesis and morphological changes in the follicular epithelial cells of oöcytes of Douglas-fir beetle, Dendroctonus pseudotsugae were studied during the reproductive phase. Inhibition of synthesis of DNA dependent RNA by actinomycin D injections blocked yolk deposition in the oöcytes as well as oviposition within the normal period. A mixture of radioactively labelled haemolymph and ovarial proteins was deposited as yolk proteins in the oöcytes of normal beetles. Such proteins were not deposited in the oöcytes of females injected with actinomycin D; the blockage of yolk deposition persisted even when such females were treated extraneously with juvenile hormone.  相似文献

The present investigation was aimed towards analyzing the potential of consortia of native filamentous microalgal strains (MC2), native unicellular microalgal strains (MC3), and selected microalgae from germplasm (MC1) in terms of nutrient removal, water quality improvement, and biomass production using primary treated sewage water. Highest NO3-N (90 %) and PO4-P (97.8 %) removal was obtained with MC2-inoculated sewage water. Highest decrease in total dissolved solids to 806 from 1,120 mg L?1 and highest increase in dissolved oxygen of 9.0 from 0.4 mg L?1 were obtained using MC2-inoculated sewage water on the sixth day. The biomass production was also highest in MC2 (1.07 g L?1) followed by MC1 and MC3 (0.90 and 0.94 g L?1, respectively) on the sixth day. The consortium of filamentous strains from native environment not only proved promising in nutrient removal efficiency but also led to enhanced biomass. The present study highlighted the utility of such a consortium for sewage wastewater treatment and the promise of sewage water as a growth medium for biomass production.  相似文献
