Leishmaniasis and Chagas disease afflict the poorest countries in the world. The Brazilian flora represents a rich source for the screening of potential antiparasitic compounds. In this work, we tested the total alkaloid and ethanol extracts of nine different plants from Brazilian families which produce isoquinoline alkaloids, to determine their in vitro antiparasitic effect against L. chagasi and T. cruzi parasites. Promastigotes of L. chagasi were shown to be susceptible only to the total alkaloid extracts of A. crassiflora (EC50 value = 24.89 microg/ml), A. coriacea (EC50 value = 41.60 microg/ml), C. ovalifolia (EC50 value = 63.88 microg/ml) and G. australis (EC50 value = 37.88 microg/ml). Except for the G. australis total alkaloids, all the three extracts presented a considerable activity when tested against intracellular amastigotes. The most effective alkaloid extracts were those from A. crassiflora and C. ovalifolia, which reduced the number of infected macrophages at 25 microg/ml by 86.1% and 89.8%, respectively. Among the 18 tested extracts, 16 showed anti-Trypanosoma activity. Eight extracts (A. crassiflora, A. coriacea, C. ovalifolia, D. furfuracea, D. lanceolata, S. guianensis, X. emarginata and G. australis) were the most effective against the trypomastigotes, killing approximately 100% of the parasites at the maximal concentration of 100 microg/ml. Cytotoxicity against mammalian cells was evaluated for all extracts, but potential ones showed little or no cytotoxicity and a considerable antiparasitic effect, including D. furfuracea, D. lanceolata, G. australis, S. guianensis and X. emarginata. In the present study, we aimed to compare the thermoregulatory response and soccer-specific training performance aspects of two commercially available sport drinks, both of similar carbohydrate concentration, but one containing 5.2% glycerol. Ten players participated in two similar outdoor training sessions and were randomly assigned to each of two drinks: a carbohydrate (C) beverage or a carbohydrate-glycerol (CG) beverage. Players consumed 500 mL of C or CG 30 minutes pre-exercise and at half-time. Pre- and postexercise body mass, core temperature (CT), and heart rate (HR) were recorded, and urine and blood samples were taken. No difference was observed between days for wet bulb globe temperature (session 1: 17.0 +/- 1.1 degrees C, session 2: 16.9 +/- 1.1 degrees C; P = 0.944). The degree of dehydration (% Delta BM) was greater after the C trial (P = 0.041). Similarly, percent change in plasma volume was greater in the C trial (P = 0.049). No overall main affect was observed between CT and mean exercise HRs during either training session (CT: P = 0.350; mean HR: P = 0.256), and there was no difference observed between groups in time to failure during the session-ending fatigue test (P = 0.547). Ingestion of a CG beverage provided players with better hydration than C alone. However, if training sessions are short (<75 minute), with adequate time for recovery, both drinks are sufficient for maintaining performance intensities during soccer-specific training. 相似文献

The present study investigated the effects of carbofuran on the energy metabolism (levels of glycogen, total proteins, total lipids, triglycerides, and lipoperoxidation), Na+/K+ATPase activity, and reproductive parameters (formation of couples, ovigerous females, and mean number of eggs) in the freshwater amphipods Hyalella pleoacuta and Hyalella curvispina. These crustaceans live in limnetic environments of the plateau (H. pleoacuta) and coastal plain (H. curvispina) of the state of Rio Grande do Sul in southern Brazil. The animals were collected in the winter of 2006 in the Vale das Trutas (28 degrees 47'00'S-49 degrees 50'53'W) in the Municipality of S?o José dos Ausentes, and in Gentil Lagoon (29 degrees 56'30'S, 50 degrees 07'50'W) in the Municipality of Tramandaí. In the laboratory, the amphipods were kept submerged in aquariums under controlled conditions of photoperiod (12 h light: 12 h dark), temperature (23 degrees C+/-1), and constant oxygenation. Animals were exposed to carbofuran at a dose of 5 or 50 microg/L for a period of 7 days. At the end of this period, the animals were immediately frozen for determination of the biochemical parameters, lipoperoxidation levels (TBARS), and enzyme Na+/K+ATPase activity. During each day of culture, several reproductive parameters were observed. Statistical analysis (ANOVA) revealed that carbofuran induces significant decreases in glycogen, proteins, lipids, triglycerides, and Na+/K+ATPase, as well as a significant increase in lipoperoxidation levels. Studies of all the biochemical parameters seem to be quite promising, in order to assess and predict the effects of toxicants on non-target organisms. The results also suggest that the reproductive parameters (formation of couples, ovigerous females and mean number of eggs) may provide sensitive criteria for assessing ecotoxicological effects. Furthermore, H. pleoacuta and H. curvispina are suitable organisms for use in toxicity tests, and we suggest that they are sensitive species that could be used in monitoring studies. 相似文献

The objective of the present study was to evaluate the endocrine and behavioral features of estrous-induced Alpine goats. A total of 36 nulliparous, 40 non-lactating and 42 lactating does were treated with intravaginal 60 mg medroxyprogesterone acetate sponges for 9 d plus 200 IU eCG and 22.5 microg d-cloprostenol 24 h before sponge removal. Plasma progesterone concentration was analyzed from blood sampled on days 0 (sponge insertion), 5, 8 (cloprostenol administration) and 9 (sponge removal) in 11 nulliparous, 13 non-lactating and 11 lactating does. Estrous response did not differ (P>0.05) among nulliparous (97.2%), non-lactating (90.00%) and lactating does (85.7%). Interval to estrus and duration of estrus did not differ (P>0.05) among nulliparous (22.8+/-9.9 and 25.6+/-6.8h), non-lactating (23.7+/-15.8 and 25.0+/-6.0 h) and lactating does (22.2+/-10.4 and 24.9+/-4.2h). The accumulative percentage of does in estrus during the first 36 h after sponge removal was 88.1%. The correlation between interval to estrus and duration of estrus was r=-0.32 (P<0.001). Endogenous progesterone production is decreased until day 8 or suppressed by MAP on day 9. Conception rate was greater (P<0.01) in lactating (77.8%) than non-lactating (44.4%) but similar (P>0.05) to nulliparous (60.0%) goats. Estrus can be efficiently induced by means of hormonal treatment in goats and acceptable fertility can be obtained regardless of animal category. 相似文献

Spinocerebellar ataxia type 10 (SCA10) is an autosomal dominant neurodegenerative disease characterized by cerebellar ataxia and seizures. The disease is caused by a large ATTCT repeat expansion in the ATXN10 gene. The first families reported with SCA10 were of Mexican origin, but the disease was soon after described in Brazilian families of mixed Portuguese and Amerindian ancestry. The origin of the SCA10 expansion and a possible founder effect that would account for its geographical distribution have been the source of speculation over the last years. To unravel the mutational origin and spread of the SCA10 expansion, we performed an extensive haplotype study, using closely linked STR markers and intragenic SNPs, in families from Brazil and Mexico. Our results showed (1) a shared disease haplotype for all Brazilian and one of the Mexican families, and (2) closely-related haplotypes for the additional SCA10 Mexican families; (3) little or null genetic distance in small normal alleles of different repeat sizes, from the same SNP lineage, indicating that they are being originated by a single step mechanism; and (4) a shared haplotype for pure and interrupted expanded alleles, pointing to a gene conversion model for its generation. In conclusion, we show evidence for an ancestral common origin for SCA10 in Latin America, which might have arisen in an ancestral Amerindian population and later have been spread into the mixed populations of Mexico and Brazil. 相似文献

Background
The arboviruses Zika virus (ZIKV) and Dengue virus (DENV) have important epidemiological impact in Brazil and other tropical regions of the world. Recently, it was shown that previous humoral immunity to DENV enhances ZIKV replication in vitro, which may lead to more severe forms of the disease. Thus, traditional approaches of vaccine development aiming to control viral infection through neutralizing antibodies may induce cross-reactive enhancing antibodies. In contrast, cellular immune response was shown to be capable of controlling DENV infection independently of antibodies. The aim of the present study was to design a flavivirus NS5 protein capable of inducing a cellular immune response against DENV and ZIKV.Methods
A consensus sequence of ZIKV NS5 protein was designed among isolates from various continents. Epitopes were predicted for the most prevalent alleles of class I and II HLA in the Brazilian population. Then, this epitopes were analyzed with regard to their conservation, population coverage and distribution along the whole antigen.Results
Nineteen epitopes predicted to be more reactive (percentile rank <1) and 100% conserved among ZIKV and DENV serotypes were selected. The distribution of such epitopes along the protein was shown on a three-dimensional model and population coverage was calculated for different regions of the world. The designed protein was predicted to be stable and the distribution of selected epitopes was shown to be homogeneous along domains. The population coverage of selected epitopes was higher than 50% for most of tropical areas of the world.Conclusion
Such results indicate that the proposed antigen has the potential to induce protective cellular immune response to ZIKV and DENV in different human populations of the world.98.

Dysregulation of cellular iron homeostasis in human breast cancer is reflected by the altered expression of regulatory proteins. The expressions of iron-related proteins in the mammary glands of cats and dogs have not been assessed. We evaluated the expressions of ferritin, ferroportin, hepcidin and transferrin receptor 1 in benign and malignant mammary gland lesions in cats and dogs. Iron deposition was detected using Perls’ Prussian blue staining. We found no major differences in the expression of iron-related proteins between benign and malignant mammary gland lesions in either cats or dogs; however, these species exhibited accumulation of iron in benign lesions. Our findings provide an explanation for the absence of higher iron requirements by tumor cells in these animals. Further investigation of local iron homeostasis in cats and dogs and differences in their physiology compared to human breast cancer is required. 相似文献
