The interactive effects of light intensity, NaCl, nitrogen, and phosphorus on intracellular biomass content and extracellular polymeric substance production were assessed for Arthrospira sp. (Spirulina) in a two-phase culture process using principal component analysis and central composite face design. Under high light intensity (120 μmol photons m?2?s?1) and low NaCl (1 gL?1), NaNO3, and K2HPO4 (0.5 g L?1), the carbohydrate content was maximized to 26.61%. Interaction of both K2HPO4 (1.6 gL?1) and NaCl (1.19 gL?1) with low NaNO3 (0.5 gL?1) achieved the maximum content of lipids (15.62%), while high NaCl (40 gL?1), K2HPO4, and NaNO3 (4.5 gL?1) enhanced mainly total carotenoids (0.85%). Conversely, under low light intensity of 10 μmol photons m?2?s?1 combined with 11.76 gL?1 of NaCl, 0.5 gL?1 of NaNO3, and 2.68 gL?1 of K2HPO4, the phycobiliprotein content reached its highest level (16.09%). The maximum extracellular polymeric substance (EPS) production (0.902 gg?1?DW) was triggered under moderate light of 57.25 μmol photons m?2?s?1 and interaction of high NaCl (40 gL?1) and K2HPO4 (4.5 gL?1) with low NaNO3 (0.5 gL?1). The maximization ratios of intracellular biomass content in terms of carbohydrate, lipid, total carotenoid, phycobiliprotein, and EPS production were 3.55-, 1.73-, 9.55-, 2.92-, and 1.46-fold, respectively, greater than those obtained at optimal growth conditions. This study demonstrated that the multiple stress factors applied to the adopted two-phase culture process could be a promising strategy to produce biomass enriched in various high-value compound.  相似文献

Purpose

Several efforts have attempted to incorporate the sources of uncertainty and variability into the life cycle assessment (LCA) of pavements. However, no method has been proposed that can simultaneously consider data quality, methodological choices, and variability in inputs and outputs without the need for complementary software. This study aims to develop and implement a flexible method that can be used in the LCA software to assess the effects of these sources on the conclusions.

Methods

A Monte Carlo analysis was conducted and applied in a comparative LCA of pavements to assess the preferred scenario. The uncertainty of the results was first estimated by considering the data quality using the ecoinvent database. Moreover, the variabilities of the materials, construction methods, and repair stages of the pavement life cycle were included in the analysis by assigning continuous uniform probability distributions to each variable. Ultimately, the probability of methodological choices was modeled using uniform distributions and assigning a portion of the area of the distribution to each scenario. The individual and combined effects of these uncertainty and variability sources were assessed in a comparative LCA of asphalt and concrete pavements in a cold region such as Quebec (Canada).

Results and discussion

The results of the Monte Carlo analysis show that the allocation choices can change the environmentally preferred scenario in four midpoint categories. These categories are significantly dominated by the crude oil supply chain. The variability in construction materials and methods can change the preferred scenario in the damage categories, namely, human health and global warming. Additionally, parameter uncertainty has a significant effect on the conclusion of the preferred scenario in ecosystem quality. The worst qualitative scores are given to the geographical uncertainty of the elementary flow that primarily contributes to this category (i.e., zinc). The simultaneous effect of the uncertainty and variability sources prevents the decision-maker from reaching a less uncertain conclusion about ecosystem quality, human health, and global warming effects.

Conclusions

This study demonstrates that it is feasible to assess the cumulative effects of common uncertainty and variability sources using commercial LCA software, including Monte Carlo simulation. Based on the variability and uncertainty of the results, the identification of a certain conclusion is case specific at both the midpoint and endpoint levels. Increasing the quality of the inventory is one solution to decreasing the uncertainties related to human health, ecosystem quality, and global warming regarding pavement LCA. This improvement can be achieved by avoiding the adaptation of a similar process to match the considered process and using practical construction efficiencies and realistic construction materials. The effectiveness of these tasks must be assessed in future studies. It should be noted that these conclusions were determined regardless of the uncertainty in the characterization factors of the impact assessment method.

Mitochondrial import of pyruvate by the mitochondrial pyruvate carrier (MPC) is a central step which links cytosolic and mitochondrial intermediary metabolism. To investigate the role of the MPC in mammalian physiology and development, we generated a mouse strain with complete loss of MPC1 expression. This resulted in embryonic lethality at around E13.5. Mouse embryonic fibroblasts (MEFs) derived from mutant mice displayed defective pyruvate-driven respiration as well as perturbed metabolic profiles, and both defects could be restored by reexpression of MPC1. Labeling experiments using 13C-labeled glucose and glutamine demonstrated that MPC deficiency causes increased glutaminolysis and reduced contribution of glucose-derived pyruvate to the TCA cycle. Morphological defects were observed in mutant embryonic brains, together with major alterations of their metabolome including lactic acidosis, diminished TCA cycle intermediates, energy deficit and a perturbed balance of neurotransmitters. Strikingly, these changes were reversed when the pregnant dams were fed a ketogenic diet, which provides acetyl-CoA directly to the TCA cycle and bypasses the need for a functional MPC. This allowed the normal gestation and development of MPC deficient pups, even though they all died within a few minutes post-delivery. This study establishes the MPC as a key player in regulating the metabolic state necessary for embryonic development, neurotransmitter balance and post-natal survival.  相似文献

Dengue virus (DENV) and Zika virus (ZIKV) are flaviviruses transmitted to humans by their common vector, Aedes mosquitoes. DENV infection represents one of the most widely spread mosquito‐borne diseases whereas ZIKV infection occasionally re‐emerged in the past causing outbreaks. Although there have been considerable advances in understanding the pathophysiology of these viruses, no effective vaccines or antiviral drugs are currently available. In this study, we evaluated the antiviral activity of carnosine, an endogenous dipeptide (β‐alanyl‐l ‐histidine), against DENV serotype 2 (DENV2) and ZIKV infection in human liver cells (Huh7). Computational studies were performed to predict the potential interactions between carnosine and viral proteins. Biochemical and cell‐based assays were performed to validate the computational results. Mode‐of‐inhibition, plaque reduction, and immunostaining assays were performed to determine the antiviral activity of carnosine. Exogenous carnosine showed minimal cytotoxicity in Huh7 cells and rescued the viability of infected cells with EC50 values of 52.3 and 59.5 μM for DENV2 and ZIKV infection, respectively. Based on the mode‐of‐inhibition assays, carnosine inhibited DENV2 mainly by inhibiting viral genome replication and interfering with virus entry. Carnosine antiviral activity was verified with immunostaining assay where carnosine treatment diminished viral fluorescence signal. In conclusion, carnosine exhibited significant inhibitory effects against DENV2 and ZIKV replication in human liver cells and could be utilized as a lead peptide for the development of effective and safe antiviral agents against DENV and ZIKV.  相似文献

Gravity is a constant force guiding the direction of plant growth. In young poplar stem, reorientation of the apical region is mainly obtained by differential growth of elongating primary tissues. At the base, where elongation is achieved but where the cambium is active, reorientation is due to asymmetrical formation of reaction wood. After 45 min of gravistimulation, the stem showed no reorientation, but 1 week later, reaction wood was observed at the base of the stem. To determine the molecular mechanisms taking place at the top and base of the stem, after 45 min or 1 week of inclination, the changes induced in protein accumulation were studied by two-dimensional polyacrylamide gel electrophoresis and quantitatively analyzed using image analysis software. Around 300 protein spots were reproducibly detected and analyzed. Forty percent of these proteins showed significant changes after inclination. Mass spectrometry analysis of 135 spots led to the identification of 60 proteins involved in a wide range of activities and metabolisms. Very different patterns of protein expression were obtained according to conditions tested, highlighting the complexity of gravitropic responses. Our results suggest that primary and secondary tissues present specific mechanisms to sense reorientation and to respond to inclination. Some selected proteins are discussed.  相似文献

Soil salinity and sodicity are major constraints to rice production in about twenty per cent of the irrigated crop land. Inbuilt genetic tolerance to salinity is the most economical and environmentally sustainable way to solve this problem. A mapping population of 200 F2 plants and their corresponding F3 families, derived from a cross between a salt tolerant indica rice variety CSR27 and a salt sensitive variety MI48 were used to map OTLs for salt tolerance. Seventeen different parameters, including seedling salt injury score, Na+, K+, CI? concentrations and Na+/K+ ratio in leaf and stem tissues at vegetative and reproductive stages were mapped. A framework linkage map was constructed using 79 SSR and EST markers distributed over the twelve rice chromosomes at an average interval of 20.7cM and total map distance of 1634.5 cM. Twenty five major OTLs, each explaining more than ten per cent of the trait phenotypic variance, were mapped on chromosomes 1, 2, 3 and 8. These included one OTL for seedling salt injury score, nine for Na+ concentration, three for K+ concentration and four for Cl? concentration in leaf and stem tissues at vegetative and reproductive stages. The Na+/K+ ratio, an important ion balancing parameter for the salt tolerance, was controlled by eight OTLs explaining phenotypic variance in the range of 42.88–52.63%. Four OTL intervals were robust with major effect and having OTLs for multiple salt tolerance parameters that might be governed by common or tightly linked genes. One major OTL for multiple salt tolerance parameters on chromosome 8 and three major OTLs for CI? ion concentration are novel for this study. The OTLs identified here will serve as a base for fine mapping, gene tagging and marker assisted selection for salt tolerance in rice.  相似文献
