Actin filaments play an essential role in cell movement, and many posttranslational modifications regulate actin filament assembly. Here we report that prolyl hydroxylase 3 (PHD3) interacts with nonmuscle actin in human cells and catalyzes hydroxylation of actin at proline residues 307 and 322. Blocking PHD3 expression or catalytic activity by short hairpin RNA knockdown or pharmacological inhibition, respectively, decreased actin prolyl hydroxylation. PHD3 knockdown increased filamentous F-actin assembly, which was reversed by PHD3 overexpression. PHD3 knockdown increased cell velocity and migration distance. Inhibition of PHD3 prolyl hydroxylase activity by dimethyloxalylglycine also increased actin polymerization and cell migration. These data reveal a novel role for PHD3 as a negative regulator of cell motility through posttranslational modification of nonmuscle actins. 相似文献

Age‐associated ventriculomegaly is typically attributed to neurodegeneration; however, additional factors might initiate or contribute to progressive ventricular expansion. By directly linking postmortem human MRI sequences with histological features of periventricular tissue, we show that substantial lateral ventricle surface gliosis is associated with ventriculomegaly. To examine whether loss of ependymal cell coverage resulting in ventricle surface glial scarring can lead directly to ventricle enlargement independent of any other injury or degenerative loss, we modeled in mice the glial scarring found along the lateral ventricle surface in aged humans. Neuraminidase, which cleaves glycosidic linkages of apical adherens junction proteins, was administered intracerebroventricularly to denude areas of ependymal cells. Substantial ependymal cell loss resulted in reactive gliosis rather than stem cell‐mediated regenerative repair of the ventricle lining, and the gliotic regions showed morphologic and phenotypic characteristics similar to those found in aged humans. Increased levels of aquaporin‐4, indicative of edema, observed in regions of periventricular gliosis in human tissue were also replicated in our mouse model. 3D modeling together with volume measurements revealed that mice with ventricle surface scarring developed expanded ventricles, independent of neurodegeneration. Through a comprehensive, comparative analysis of the lateral ventricles and associated periventricular tissue in aged humans and mouse, followed by modeling of surface gliosis in mice, we have demonstrated a direct link between lateral ventricle surface gliosis and ventricle enlargement. Waste gas containing diethyldisulphide (DEDS) is generated from various industries including pulp and paper, refinery, rayon and molasses based distilleries, etc. DEDS has odour threshold detection with an average concentration of 10(-9)mg/m(3) at 25 degrees C. DEDS is toxic to bacteria, fungus and also to mammals when exposed for a long period. Waste gas containing DEDS require proper treatment prior to discharge into the environment. DEDS containing waste gas was treated in a biofilter, packed with compost along with wooden chips and enriched with DEDS degrading microorganisms. The biofilter could remove DEDS to the extent of 94+/-5% at a loading of 1.60 g/m(3)/h with an empty bed retention time of 150s. At optimal operating conditions, the average moisture content required by the biofilter was in the range of 60-65%. The biodegradative products of DEDS were thiosulphate and sulphate. 相似文献

Fire is considered as an extreme disturbance in Mediterranean grasslands or shrublands as it often brings about many sudden changes in the vegetation structure, composition, and diversity patterns. In addition, it creates opportunities for exotic plant species to establish successfully in foreign habitat, and to outperform dominating native species. Monitoring and simulating post-fire successional changes, therefore, are essential tasks to efficiently restore native grasslands or shrublands. In this paper, we develop a theoretical framework for simulating fire-induced successional changes, mainly for Mediterranean vegetation, based on a three-level hierarchy of successional causes. Within this proposed framework, fire effects are considered by associating it with the number of burned sites open-up and specific changes at the burned sites relative to unburned sites. Three distinct site-specific neighborhoods are constructed; changes within each neighborhood allow sequential replacement of plant species by another plant species with greater maximum size, age and lower maximum growth rates and dispersal abilities. The proposed framework can be used to develop a spatially explicit individual-based model which will be useful for monitoring and predicting successional changes and hence for restoring native grasslands or shrublands. 相似文献

The Sundarbans tiger inhabits a unique mangrove habitat and are morphologically distinct from the recognized tiger subspecies in terms of skull morphometrics and body size. Thus, there is an urgent need to assess their ecological and genetic distinctiveness and determine if Sundarbans tigers should be defined and managed as separate conservation unit. We utilized nine microsatellites and 3 kb from four mitochondrial DNA (mtDNA) genes to estimate genetic variability, population structure, demographic parameters and visualize historic and contemporary connectivity among tiger populations from Sundarbans and mainland India. We also evaluated the traits that determine exchangeability or adaptive differences among tiger populations. Data from both markers suggest that Sundarbans tiger is not a separate tiger subspecies and should be regarded as Bengal tiger (P. t. tigris) subspecies. Maximum likelihood phylogenetic analyses of the mtDNA data revealed reciprocal monophyly. Genetic differentiation was found stronger for mtDNA than nuclear DNA. Microsatellite markers indicated low genetic variation in Sundarbans tigers (He= 0.58) as compared to other mainland populations, such as northern and Peninsular (Hebetween 0.67- 0.70). Molecular data supports migration between mainland and Sundarbans populations until very recent times. We attribute this reduction in gene flow to accelerated fragmentation and habitat alteration in the landscape over the past few centuries. Demographic analyses suggest that Sundarbans tigers have diverged recently from peninsular tiger population within last 2000 years. Sundarbans tigers are the most divergent group of Bengal tigers, and ecologically non-exchangeable with other tiger populations, and thus should be managed as a separate “evolutionarily significant unit” (ESU) following the adaptive evolutionary conservation (AEC) concept. 相似文献
