The neutral noncyclic imide and ether containing ionophore (AS701), a selective carrier for Li+ among alkali cations, was found to be capable of mediating the transport of NH4+ and of biogenic amines (catechols and indoles) across lipid bilayer membranes also. Ionophore-mediated electrical properties of planar lipid bilayers were studied under experimental conditions where the positively-charged amine species was dominant. The ionophore was found to act as a selective carrier of the biogenic amines, mediating their electrogenic transport across the membrane, forming 2:1 carrier-amine permeant complexes, carrying a net-charge of +1. Selectively among the amines corresponding to the following sequence: tryptamine (35) > Li+ (1) > serotonin (0.60) > dopamine (0.19) > norepinephrine (0.13) > epinephrine (0.05) > NH4+ (0.05). The molecular factors involved in determining these selectivities are assessed.  相似文献

Summary The neutral noncyclic, lithium-selective ionophore ETH1644, which is structurally different from previously available ionophores of this type, is a selective carrier of Li in lipid bilayer membranes of various lipid composition. The ionophore forms a 21 carrier/cation complex, and the rate-limiting step in the overall transport process is the diffusion of the carrier/ion complex across the membrane.The selectivity sequence for lithiumvs. other ions normally found in biological systems is: Li+ (1)>Na+ (0.017)K+ (0.017) >Cl (0.001), Ca2+ and Mg2+ are impermeant. At neutral pH protons do not interfere with the Li+-carrying ability of this ionophore. On the basis of structural differences and supported by conductance data, it is argued that the improved selectivity of Li+ over the other alkali cations is due more to a decrease in the affinities of the ionophore for the latter cations that to an increase of its affinity to Li+. This ionophore can also act as a carrier of biogenic amines (catecholes, indoles and derivatives), with the structure of the permeant species and mechanism of permeation similar to that observed with the alkali cations. The selectivity sequence is: tryptamine (18.1)>phenylethylamine (11.6)> tyramine (2.4)>Li+(1)>serotonin (0.34)>epinephrine (0.09) >dopamine (0.05)>norepinephrine (0.02), showing the ionophore to be more selective to Li+ than to any of the neurotransmitters studies.  相似文献

Myocardial Na+,K+-ATPase was studied in patients with aortic valve disease, and myocardial Na+,K+- and Ca2+-ATPase were assessed in spontaneously hypertensive rats (SHR) and hereditary cardiomyopathic hamsters using methods ensuring high enzyme recovery. Na+,K+-ATPase was quantified by [3H]ouabain binding to intact myocardial biopsies from patients with aortic valve disease. Aortic stenosis, regurgitation and a combination hereof were compared with normal human heart and were associated with reductions of left ventricular [3H]ouabain binding site concentration (pmol/g wet weight) of 56, 46 and 60%, respectively (p < 0.01). Na+,K+ and Ca2+-ATPases were quantified by K+- and Ca2+-dependent p-nitrophenyl phosphatase (pNPPase) activity determinations in crude myocardial homogenates from SHR and hereditary cardiomyopathic hamsters. When SHR were compared to age-matched Wistar Kyoto (WKY) rats an increase in heart-body weight ratio of 75% (p < 0.001) was associated with reductions of K+- and Ca2+-dependent pNPPase activities (mol/min/g wet weight) of 42 (p < 0.01) and 27% (p < 0.05), respectively. When hereditary cardiomyopathic hamsters were compared to age-matched Syrian hamsters an increase in heart-body weight ratio of 69% (p < 0.001) was found to be associated with reductions in K+- and Ca2+-dependent pNPPase activities of 50 (p < 0.001) and 26% (p = 0.05), respectively. The reductions in Na+,K+- and Ca2+-ATPases were selective in relation to overall protein content and were not merely the outcome of increased myocardial mass relative to Na+,K+- and Ca2+-pumps. In conclusion, myocardial hypertrophy is in patients associated with reduced Na+,K+-ATPase concentration and in rodents with reduced Na+,K+- and Ca2+-ATPase concentrations. This may be of importance for development of heart f in hypertrophic heart disease.  相似文献

Summary The neutral, noncyclic Li-selective ionophore ETH1810, which is a di-imide, differs structurally from previous similar ionophores by removal of the intramolecular symmetry of the N-imide substituents. Properties of this ionophore, as a potential carrier of lithium, were probed through studies of ionophore-induced changes in electrical properties of lipid bilayer membranes. ETH1810 was found capable of transporting lithium and other monovalent cations, across lipid bilayer membranes, forming 21 ionophore: ion membrane-permeating species. It was found to be 10-fold more potent than ETH1644, which was the previous best ionophore of this type. The selectivity sequence among alkali cations was found to be: Li+(1)>Na+ (0,009)>K+ (0.004)>Cs+(0.0035). Among the physiological alkali cations, it constitutes a 40 (vs. Na+) to 160% (vs. K+) improvement over ETH1644. ETH1810 was also found to be capable of acting as a carrier of biogenic amines and related molecules, with the following selectivity sequence: tryptamine (20)>phenylethylamine (7.8)>tyramine (4.3)>serotonin (2.5)>Li+ (1)>NH 4 + (0.013)>dopamine (0.012). It was found that protons, at physiological concentrations, do not interfere with the lithium transport mediated by ETH1810. The relationship between the improvements in ionic selectivity and potencyvs. the differences in structural features is discussed.  相似文献

Pre-emergence soil application of the herbicide diphenamid in concentrations exceeding the normal field rate increased the resistance of tomato plants towards infection by the wilt fungus Fusarium oxysporum f.sp. lycopersici. This was detected as significant increases in the percentage emergence of seedlings although growth parameters of the raised seedlings were reduced. Treated plants exhibited no wilt symptoms, although the pathogen maintained its population at detectable levels in the rhizosphere of tomato plants. However, the growth inhibition caused by diphenamid alone was much less than that reported for the combined application of pathogen and herbicide. Growth activities of F. oxysporum f.sp. lycopersici were inhibited by high concentrations of diphenamid in vitro. It is possible that the biodegradation of this herbicide by species such as Aspergillus candidus (present in substantial counts in treated rhizospheres) was one of the causes of increased tolerence of the pathogen to the herbicide in situ.  相似文献

Familial Mediterranean Fever (FMF) is an autosomal recessive inflammatory disorder predominantly affecting people living in or originating from areas around the Mediterranean Sea, mainly Jews, Armenians, Turks, and Arabs. It is characterized by recurrent attacks of inflammation of serosal membranes and fever resulting in acute abdominal, chest, or joint pain. Over 50 MEditerranean FeVer (MEFV) mutations and polymorphisms have been identified in FMF patients. The objective of this study was to analyze the distribution and frequencies of 12 MEFV mutations in 266 referred Lebanese patients using a reverse-hybridization assay. Of the 266 patients, 129 (48.5%) were positive for at least one mutation and 137 (51.5%) had no mutations detected. Of the 129 patients with mutations, 35 were homozygous, 41 were compound heterozygous and 53 were heterozygous. The five most common mutations M694V, E148Q, V726A, M694I and M680I (G/C) accounted for 26.1, 22.2, 21.3, 9.6 and 7.7%, respectively. The A744S, F479L, R761H and I692del were encountered in 2.9% of patients; P369S and M680I (G/A) were found in 1.2% of patients while K695R was absent. The spectrum of the MEFV mutations among our sampled Lebanese FMF patients shows the high heterogeneity at the allelic level when compared to Arab and non-Arab populations. The most important feature was the relatively high frequency of the E148Q in our study group that allows us to question it as a mutation rather than a polymorphism. Further studies should be conducted to evaluate the role of the E148Q allele.  相似文献

Fibrinogen is a plasma protein that has been reported to be associated with an increased risk of atherothrombotic diseases and venous thrombosis. The most common polymorphism that has been studied so far in different populations is the G-455-->A polymorphism in the promoter region of the beta-fibrinogen gene. We studied 160 healthy unrelated Lebanese individuals for the prevalence of -455G/G, -455G/A and -455A/A genotypes of the beta-fibrinogen gene and the frequency of G and A alleles using a reverse hybridization PCR assay. The prevalence of the G/G, G/A, and A/A genotypes were found to be 60.6, 31.9 and 7.5%, respectively. The frequency of the G and A alleles were found to be 0.77 and 0.23, respectively. As compared to other ethnic groups, the Lebanese individuals were found to have a relatively high prevalence of the A allele which may predispose them to develop cardiovascular diseases as well as thrombotic events. This study provides additional unique genetic information pertaining to the Lebanese population.  相似文献

Sesame (Sesamum indicum L.), the “Queen of oil seeds” is being infected with pathogens, i.e., fungi, bacteria, virus and nematodes. Fusarium oxysporum sp. sesami (Zap.), is one of the fiercest pathogens causing severe economic losses on sesame. This work aimed to evaluate the impact of the cultivation of some preceding crops and seed inoculation with antagonistic predominant rhizospheric bacteria and actinomycetes on the incidence and development of Fusarium damping-off and wilt disease. Results showed that the lowest pre and/or post-emergence damping-off and wilt of sesame were recorded after onion and garlic, followed by wheat compared to clover in both the 2019 and 2020 seasons. In vitro, soil extracts from plots where onion and garlic have been cultivated slightly decreased the conidia germination and mycelium radial growth of F. oxysporum. The numbers of sesame rhizospheric F. oxysporum and fungi were lower after the cultivation of onion and garlic than those after wheat and clover. However, the numbers of actinomycetes and bacteria were higher in the onion, garlic, and clover rhizosphere than wheat. Among all isolated bacteria and actinomycetes associated with sesame roots cultivated after preceding plants, the Tricoderma viride and Bacillus subtilis (isolate No.3) profoundly reduce F. oxysporum mycelial growth in vitro. When sesame seeds were inoculated with Tricoderma viride, Bacillus subtilis, Streptomyces rochei and Pseudomonas fluorescens, the disease incidence of damping-off and wilt significantly decreased in the greenhouse and field trials conducted in both tested growing seasons, also had highly significant on plant health and growth parameters. Therefore, the current study suggested that using the preceding onion and garlic plants could be used for eco-friendly reduction of damping-off and wilt disease of sesame.  相似文献
