OBJECTIVE: The aim of this study was to investigate the cholesterol-lowering mechanisms of corn fiber oil (CFO), ferulate phytostanyl esters (FPEs) and parent compounds of FPE, including sitostanol and ferulic acid, in hamsters. METHOD: Seventy male Golden Syrian hamsters were randomly assigned to six experimental diets for 4 weeks: (1) cornstarch-casein-sucrose-based control diet (control); and (2) control diet plus 0.1% (wt/wt) cholesterol (cholesterol-control). The remaining four groups were given cholesterol-control diet with: (3) 10% (wt/wt) CFO; (4) 0.5% (wt/wt) sitostanol; (5) 0.23% (wt/wt) ferulic acid; and (6) 0.73% (wt/wt) FPE. At the end of dietary intervention, total plasma cholesterol, high-density lipoprotein cholesterol and triglyceride concentrations were determined. Parameters of cholesterol kinetics, including cholesterol absorption and synthesis, as well as mRNA expression of sterol transporters such as Niemann-Pick C1 like 1 (NPC1L1), ATP-binding cassette G5 (ABCG5) and ABCG8, were assessed. RESULTS: Supplementation with CFO decreased (P<.0001) plasma total cholesterol levels by 29% as compared with the cholesterol-control group, while FPE and sitostanol reduced (P<.02) cholesterolemia by 15% and 14%, respectively. CFO and sitostanol decreased (P<.05) cholesterol absorption by 24% compared to the cholesterol-control group. Dietary intervention did not alter the intestinal gene expression of ABCG5, ABCG8 and NPC1L1. CONCLUSION: The present results show that the CFO-induced and sitostanol-induced decrease in cholesterol absorption is independent of intestinal enterocyte sterol transporters such as ABCG5, ABCG8 and NPC1L1 in hamsters.  相似文献

Plasminogen activator inhibitor-1 (PAI-1) is an inhibitor of fibrinolysis. Increased plasma PAI-1 levels play an essential role in the pathogenesis of cardiovascular risk and other diseases associated with thrombosis. The 4G/5G polymorphism of the PAI-1 promoter region has been extensively studied in different populations. We studied 160 healthy unrelated Lebanese individuals using a reverse hybridization PCR assay to detect the 5G/5G, 4G/5G and, 4G/4G genotypes of the PAI-1 gene and the frequencies of the 4G and 5G alleles. We found that 4G/5G genotype was the most prevalent (45.6%) followed by 5G/5G (36.9%) and 4G/4G (17.5%). The frequencies of the 4G and 5G alleles were calculated to be 0.403 and 0.597, respectively. Compared to other ethnic communities, the Lebanese population was found to harbour a relatively high prevalence of the rare 4G allele. This, in turn, may predispose this population to develop cardiovascular diseases and other thrombotic clinical conditions. This study aids to enhance our understanding of the genetic features of the Lebanese population.  相似文献

BACKGROUND: Mutations in the 5,10-methylenetetrahydrofolate reductase (MTHFR) gene could reduce the enzyme activity and lead to hyperhomocysteinemia, a condition that has been associated with several vascular conditions, in particular, coronary artery disease and deep vein thrombosis. AIM: The aim of this study was to assess the prevalence of the two most common polymorphisms, C677T and A1298C, which have not been well studied in the Lebanese population. METHODS: We randomly selected 205 healthy individuals originating from different Lebanese provinces and religious communities. The CVD StripAssay was used to test for MTHFR gene polymorphisms. RESULTS: We found that for C677T, the prevalence of C/C, C/T, and T/T genotypes was 65.3%, 30.8%, and 3.9%, respectively, with an overall carrier rate of 34.6% and allelic frequency of 0.19. However, the A1298C genotypic prevalence of A/C, A/A, and C/C was 50.2%, 25.9%, and 23.9%, respectively, with an overall carrier rate of 74.14% and an allelic frequency of 0.49. CONCLUSIONS: Compared to all other populations reported so far, the Lebanese population harbors the highest prevalence of the MTHFR A1298C polymorphism. This is an important finding to be followed in terms of clinical significance and sheds light on an additional unique genetic feature in this community.  相似文献

Species from the genus Mayetiola are observed in the main cereal cultures of Tunisia. Some researchers have studied M. destructor that attacks wheat and M. hordei that attacks barley. However, a third important species observed in oat, M. avenae, has not been studied and is not well documented in Tunisia. A method to easily separate the species is needed to clarify the occurrences of these gall midge species. This study aimed to first distinguish between the three species of gall midges by molecular characterization and second to reveal the phylogenetic relationships within and between the three species of Mayetiola collected from 5 different regions of northern Tunisia. To achieve these purposes, two regions of the mitochondrial DNA, cytochrome oxidase subunit I gene, and the 16S rRNA gene were amplified by polymerase chain reaction and sequenced. For each marker, a set of 75 individuals were used for DNA analysis. Phylogenetic trees were created using the DNA sequences of all samples from the 3 species. Results showed significant separation of the three different species into dissimilar clades. Each clade contained only specimens from the same species. Differences were observed between DNA sequences of the same species. The differences within the same species were not representative of geographical variations but coexisted within a population Therefore, using the COI and 16S rRNA genes as markers can clearly separate M. avenae, M. destructor and M. hordei.  相似文献

The morphological and molecular studies provide greater taxonomic resolution for the scleractinian coral identification. The Euphylliidae corals are among the scleractinian family for which their corallite and polyp morphologies have been examined for species identification. However, knowledge on the molecular study for coral identification in Malaysia is very limited. Therefore, this study aimed to examine the morphological structures and phylogenetic analyses for six Euphylliidae coral species using the mitochondrial gene of cytochrome oxidase subunit I (COI). The results showed that the Euphylliidae corals are present under both “complex” and “robust” coral clades as supported by many researchers. The result also revealed that the species phylogeny of Euphylliidae corals is in concordances with its morphological structures of corallites. It can be concluded the combination between morphological structures and phylogenetic analyses provide more accurate identification than relying on morphological study alone. Hence, it provides a future direction for the scleractinian research progress in species identification.  相似文献

Local delivery of antibiotic into injured bone is a demand. In this work, different scaffolds of chitosan (C) with or without bioactive glass (G) were prepared using the freeze-drying technique in 2:1, 1:1, and 1:2 weight ratios. Chitosan scaffolds and selected formulas of chitosan to bioglass were loaded with ciprofloxacin in 5%, 10%, and 20% w/w. Scaffold morphology showed an interconnected porous structure, where the glass particles were homogeneously dispersed in the chitosan matrix. The kinetic study confirmed that the scaffold containing 1:2 weight ratio of chitosan to glass (CG12) showed optimal bioactivity with good compromise between Ca and P uptake capacities and Si release rate. Chitosan/bioactive glass scaffolds showed larger t 50 values indicating less burst drug release followed by a sustained drug release profile compared to that of chitosan scaffolds. The cell growth, migration, adhesion, and invasion were enhanced onto CG12 scaffold surfaces. Samples of CG12 scaffolds with or without 5% drug induced vascular endothelial growth factor (VEGF), while those containing 10% drug diminished VEGF level. Only CG12 induced the cell differentiation (alkaline phosphatase activity). In conclusion, CG12 containing 5% drug can be considered a biocompatible carrier which would help in the localized osteomyelitis treatment.  相似文献

Kinetic studies of X exchange on [AuX4] square-planar complexes (where X=Cl and CN) were performed at acidic pH in the case of chloride system and as a function of pH for the cyanide one. Chloride NMR study (330-365 K) gives a second-order rate law on [AuCl4] with the kinetic parameters: (k2Au,Cl)298=0.56±0.03 s−1 mol−1 kg; ΔH2‡ Au,Cl=65.1±1 kJ mol−1; ΔS2‡ Au,Cl=−31.3±3 J mol−1 K−1 and ΔV2 Au,Cl=−14±2 cm3 mol−1. The variable pressure data clearly indicate the operation of an Ia or A mechanism for this exchange pathway. The proton exchange on HCN was determined by 13C NMR as a function of pH and the rate constant of the three reaction pathways involving H2O, OH and CN were determined: k0HCN,H=113±17 s−1, k1HCN,H=(2.9±0.7)×109 s−1 mol−1 kg and k2HCN,H=(0.6±0.2)×106 s−1 mol−1 kg at 298.1 K. The rate law of the cyanide exchange on [Au(CN)4] was found to be second order with the following kinetic parameters: (k2Au,CN)298=6240±85 s−1 mol−1 kg, ΔH2 Au,CN=40.0±0.8 kJ mol−1, ΔS2 Au,CN=−37.8±3 J mol−1 K−1 and ΔV2 Au,CN=+2±1 cm3 mol−1. The rate constant observed varies about nine orders of magnitude depending on the pH and HCN does not act as a nucleophile. The observed rate constant of X exchange on [AuX4] are two or three orders of magnitude faster than the Pt(II) analogue.  相似文献

L6 myoblasts stably transfected with a GLUT4 cDNA harboring an exofacial myc epitope tag (L6-GLUT4myc myoblasts) were used to study the role of protein kinase B alpha (PKBalpha)/Akt1 in the insulin-induced translocation of GLUT4 to the cell surface. Surface GLUT4myc was detected by immunofluorescent labeling of the myc epitope in nonpermeabilized cells. Insulin induced a marked translocation of GLUT4myc to the plasma membrane within 20 min. This was prevented by transient transfection of a dominant inhibitory construct of phosphatidylinositol (PI) 3-kinase (Deltap85alpha). Transiently transfected cells were identified by cotransfection of green fluorescent protein. A constitutively active PKBalpha, created by fusion of a viral Gag protein at its N terminus (GagPKB), increased the cell surface density of GLUT4myc compared to that of neighboring nontransfected cells. A kinase-inactive, phosphorylation-deficient PKBalpha/Akt1 construct with the mutations K179A (substitution of alanine for the lysine at position 179), T308A, and S473A (AAA-PKB) behaved as a dominant-negative inhibitor of insulin-dependent activation of cotransfected wild-type hemagglutinin (HA)-tagged PKB. Furthermore, AAA-PKB markedly inhibited the insulin-induced phosphorylation of cotransfected BAD, demonstrating inhibition of the endogenous PKB/Akt. Under the same conditions, AAA-PKB almost entirely blocked the insulin-dependent increase in surface GLUT4myc. PKBalpha with alanine substitutions T308A and S473A (AA-PKB) or K179A (A-PKB) alone was a less potent inhibitor of insulin-dependent activation of wild-type HA-PKB or GLUT4myc translocation than was AAA-PKB. Cotransfection of AAA-PKB with a fourfold DNA excess of HA-PKB rescued insulin-stimulated GLUT4myc translocation. AAA-PKB did not prevent actin bundling (membrane ruffling), though this response was PI 3-kinase dependent. Therefore, it is unlikely that AAA-PKB acted by inhibiting PI 3-kinase signaling. These results outline an important role for PKBalpha/Akt1 in the stimulation of glucose transport by insulin in muscle cells in culture.  相似文献

The study aims to investigate the effect of foliar spray with three plant growth regulators (PGRs) p-Chlorophenoxyacetic acid (CPA) at 20 and 40 ppm; Gibberellic acid (GA3) at 20 and 30 ppm, 1-Naphthaleneacetic acid (NAA) at 10 and 20 ppm on the response of fruit set, yield, and fruit quality of some hot pepper cultivars (Chillina, Parbirian, Shampion, and Hyffa) grown in sandy soil under plastic tunnels as compared to the control. Spraying Chillina cultivar GA3 at 30 ppm significantly increased the number of fruits/ plant and fruit set (%), yield/plant, and total yield/fad. In addition, the contents of TSS and Vit C, furthermore, maximum capsaicin content were observed in chili fruits in both seasons. However, the interaction between Chillina cultivar and spraying with GA3 at 20 ppm ranked second in yield and quality. The interaction between Parbirian cultivars and spraying with GA3 at 20 or 30 ppm increased the number of flowers/plants in both seasons. On the other hand, the interaction between Shampion cultivar and spraying with tap water (control) gave the lowest values of the number of flowers/ plants, the number of fruits/ plant and fruit set (%), yield, and its components, and fruit quality in both seasons.  相似文献
