An efficient adventitious shoot regeneration procedure was developed for Gypsophila paniculata L. Using cultivar Arbel, shoot regeneration from the three upper internodes of the stem was monitored on MS media supplemented with different cytokinins (thidiazuron, benzyladenine, kinetin or zeatin) and an auxin (naphthaleneacetic acid). Thidiazuron was found to be the most efficient cytokinin, with up to 100% of the explants forming shoots, at an average of up to 19 shoots per explant being regenerated. The highest percentage of shoot formation was observed in the stem explants originating from the first internode, with all cytokinins tested. The adventitious origin of shoots regenerated from stem explants was confirmed by scanning electron microscopy. The regeneration procedure was found to be applicable to five other gypsophila cultivars (Perfecta, Golan, Gilboa, Flamingo and Tavor). Regenerating plants were successfully transferred to soil, and did not differ in flower color, size or shape from standard vegetatively propagated plants derived from cuttings. Recent findings on differences between the gregarious and solitary phases of locusts are reviewed in relation to flight fuel utilization, adipokinetic responses, and adipokinetic hormones. Laboratory results obtained with Locusta migratoria migratorioides show that the amount of lipid reserves, resting levels of haemolymph lipids, and hyperlipaemic responses to flight and to injection of corpus cardiacum extract or of synthetic adipokinetic hormones, are higher in crowded than in isolated locusts. No major phase-dependent differences seem to exist in flight-related carbohydrate metabolism. The adipokinetic hormone content of the corpora cardiaca is higher in younger isolated locusts than in crowded ones. Adipokinetic hormone precursor-related peptide content of the corpora cardiaca is also higher in isolated than in crowded locusts. Crowded locusts have higher lipid reserves and higher hyperlipaemic responses to flight than isolated locusts also in Schistocerca gregaria and, following injection of synthetic adipokinetic hormone, the formation of low density lipophorin is higher in crowded than in isolated locusts of this species. The laboratory results obtained with isolated and crowded locusts are extrapolated to understand the ecophysiology of the migrations of solitary and gregarious field populations of L.m. migratorioides according to available information on the differences in the migration of the two phases. It is inferred that in this species solitary locusts have a rather coarse adipokinetic strategy focused on a single prereproductive long-distance migratory flight, whereas gregarious locusts possess a fine adipokinetic balance for reiterative, sometimes unpredictably long-distance, migrations in the prereproductive, as well as reproductive, periods. The differences between the adipokinetic strategies of solitary and gregarious S. gregaria seem to be less dramatic, nevertheless, they indicate a better adaptation of the gregarious phase to prolonged flights.  相似文献

The mutations 185delAG, 188del11, and 5382insC in the BRCA1 gene and 6174delT in the BRCA2 gene were analyzed in 199 Ashkenazi and 44 non-Ashkenazi Jewish unrelated patients with breast and/or ovarian cancer. Of the Jewish Ashkenazi women with ovarian cancer, 62% (13/21) had one of the target mutations, as did 30% (13/43) of women with breast cancer alone diagnosed before the age 40 years and 10% (15/141) of those with breast cancer diagnosed after the age 40 years. Age at ovarian cancer diagnosis was not associated with carrier status. Of 99 Ashkenazi patients with no family history of breast and/or ovarian cancer, 10% carried one of the mutations; in two of them the mutation was proved to be paternally transmitted. One non-Ashkenazi Jewish ovarian cancer patient from Iraq carried the 185delAG mutation. Individual mutation frequencies among breast cancer Ashkenazi patients were 6.7% for 185delAG, 2.2% for 5382insC, and 4.5% for 6174delT, among ovarian cancer patients; 185delAG and 6174delT were about equally common (33% and 29%, respectively), but no ovarian cancer patient carried the 5382insC. More mutations responsible for inherited breast and ovarian cancer probably remain to be found in this population, since 79% of high-incidence breast cancer families and 35% of high-incidence breast/ovarian cancer families had none of the three known founder mutations.  相似文献

We studied inheritance of resistance to dicrotophos in greenhouse whitefly,Trialeurodes vaporariorum Westwood (Homoptera, Aleyrodidae). Compared with females from a field-collected susceptible strains (S), females from a resistant strain (R) ofT. vaporariorum derived from heavily treated cotton fields had a 28-fold greater LC50 to dicrotophos in laboratory bioassays. Concentration-mortality lines obtained from female progeny of reciprocal F1 crosses (R XS and S XR) were similar, suggesting that inheritance of dicrotophos resistance was autosomal and not influenced by maternal effects. Responses of F1 female progeny were similar to those of the parental S strain, indicating that the resistance was partially recessive (degree of dominance, D, was −0.61). Mortality observed in female progeny obtained from a backcross (F1♀ XR) corresponded more closely to expectations derived from polygenic models than to expectations from a monogenic model. The estimated number of effective factors (sensu Lande, 1981) contributing equally to resistance was three.  相似文献

Summary 2-Methyl-(4-tert-butyl)cinnamaldehyde (1) was reduced by Saccharomyces cerevisiae (baker's yeast) to S-3-(4-tert-butyl)-phenyl-2-propanol (4) in high chemical and very high optical yield (e.e. 99%). Chlorination of 4 to 5, and alkylation of the corresponding cyclic amines complete this short enantioselective synthesis of S-1-(l'-pyperidino)-2-methyl-3-(4tert-butyl)-phenyl-propane (6) and S-1-(1'-(3,5-cisdimethyl)morpholino)-2-methyl-3-(4-tert-butyl)-phenyl-propane (7), the S-enantiomers of fenpropidine and fenpropimorph, commercialy important systemic fungicides.  相似文献

The rapid development of intensive fed aquaculture (e.g. finfish and shrimp) throughout the world is associated with concerns about the environmental impacts of such often monospecific practices, especially where activities are highly geographically concentrated or located in suboptimal sites whose assimilative capacity is poorly understood and, consequently, prone to being exceeded. One of the main environmental issues is the direct discharge of significant nutrient loads into coastal waters from open-water systems and with the effluents from land-based systems. In its search for best management practices, the aquaculture industry should develop innovative and responsible practices that optimize its efficiency and create diversification, while ensuring the remediation of the consequences of its activities to maintain the health of coastal waters. To avoid pronounced shifts in coastal processes, conversion, not dilution, is a common-sense solution, used for centuries in Asian countries. By integrating fed aquaculture (finfish, shrimp) with inorganic and organic extractive aquaculture (seaweed and shellfish), the wastes of one resource user become a resource (fertilizer or food) for the others. Such a balanced ecosystem approach provides nutrient bioremediation capability, mutual benefits to the cocultured organisms, economic diversification by producing other value-added marine crops, and increased profitability per cultivation unit for the aquaculture industry. Moreover, as guidelines and regulations on aquaculture effluents are forthcoming in several countries, using appropriately selected seaweeds as renewable biological nutrient scrubbers represents a cost-effective means for reaching compliance by reducing the internalization of the total environmental costs. By adopting integrated polytrophic practices, the aquaculture industry should find increasing environmental, economic, and social acceptability and become a full and sustainable partner within the development of integrated coastal management frameworks.  相似文献

The worldwide prevalence of metabolic syndrome, which includes obesity and its associated diseases, is rising rapidly. The human gut microbiome is recognized as an independent environmental modulator of host metabolic health and disease. Research in animal models has demonstrated that the gut microbiome has the functional capacity to induce or relieve metabolic syndrome. One way to modify the human gut microbiome is by transplanting fecal matter, which contains an abundance of live microorganisms, from a healthy individual to a diseased one in the hopes of alleviating illness. Here we review recent evidence suggesting efficacy of fecal microbiota transplant (FMT) in animal models and humans for the treatment of obesity and its associated metabolic disorders.  相似文献

Mycoplasmas are the most important contaminants of cell cultures throughout the world. They are considered as a major problem in biological studies and biopharmaceutical economic issues. In this study, our aim was to find the best standard technique as a rapid method with high sensitivity, specificity and accuracy for the detection of mycoplasma contamination in the cell lines of the National Cell Bank of Iran. Thirty cell lines suspected to mycoplasma contamination were evaluated by five different techniques including microbial culture, indirect DNA DAPI staining, enzymatic mycoalert® assay, conventional PCR and real-time PCR. Five mycoplasma-contaminated cell lines were assigned as positive controls and five mycoplasma-free cell lines as negative controls. The enzymatic method was performed using the mycoalert® mycoplasma detection kit. Real-time PCR technique was conducted by PromoKine diagnostic kits. In the conventional PCR method, mycoplasma genus-specific primers were designed to analyze the sequences based on a fixed and common region on 16S ribosomal RNA with PCR product size of 425 bp. Mycoplasma contamination was observed in 60, 56.66, 53.33, 46.66 and 33.33 % of 30 different cell cultures by real-time PCR, PCR, enzymatic mycoalert®, indirect DNA DAPI staining and microbial culture methods, respectively. The analysis of the results of the different methods showed that the real-time PCR assay was superior the other methods with the sensitivity, specificity, accuracy, predictive value of positive and negative results of 100 %. These values were 94.44, 100, 96.77, 100 and 92.85 % for the conventional PCR method, respectively. Therefore, this study showed that real-time PCR and PCR assays based on the common sequences in the 16S ribosomal RNA are reliable methods with high sensitivity, specificity and accuracy for detection of mycoplasma contamination in cell cultures and other biological products.  相似文献
