Cnidarians - corals and their relatives - dominate the shallow, illuminated sea-floor in tropical nutrient-poor seas, mainly because their food sources are both heterotrophic, by predation, and autotrophic, from their intracellular symbiotic microalgae, the zooxanthellae. This trophic flexibility is rare in the animal kingdom, and the cellular usage and biological lifetime of the organic food molecules derived from both sources have never been investigated. Here we show that autotrophically-derived carbon is stored mostly in fatty tissue and is quickly consumed by the organism, while predation-derived carbon is incorporated throughout the animal’s body and remains there for a much longer time. We hypothesize that the simple photosynthate molecules are used primarily for immediate energetic processes like respiration, whereas the more complex heterotrophic organic molecules are utilized to build cellular structures such as proteins and membranes. Our study reveals, for the first time, the different fates and roles of autotrophy and heterothrophy in zooxanthellate coelenterates.  相似文献

Autism is a neurodevelopmental disorder of complex etiology in which genetic factors play a major role. We have implicated the neurexin 1 (NRXN1) gene in two independent subjects who display an autism spectrum disorder (ASD) in association with a balanced chromosomal abnormality involving 2p16.3. In the first, with karyotype 46,XX,ins(16;2)(q22.1;p16.1p16.3)pat, NRXN1 is directly disrupted within intron 5. Importantly, the father possesses the same chromosomal abnormality in the absence of ASD, indicating that the interruption of α-NRXN1 is not fully penetrant and must interact with other factors to produce ASD. The breakpoint in the second subject, with 46,XY,t(1;2)(q31.3;p16.3)dn, occurs ~750 kb 5′ to NRXN1 within a 2.6 Mb genomic segment that harbors no currently annotated genes. A scan of the NRXN1 coding sequence in a cohort of ASD subjects, relative to non-ASD controls, revealed that amino acid alterations in neurexin 1 are not present at high frequency in ASD. However, a number of rare sequence variants in the coding region, including two missense changes in conserved residues of the α-neurexin 1 leader sequence and of an epidermal growth factor (EGF)-like domain, respectively, suggest that even subtle changes in NRXN1 might contribute to susceptibility to ASD.  相似文献

Matrix metalloproteinase 13 (MMP13) is a key enzyme implicated in the degradation of the extracellular matrix in osteoarthritis. Clinical administration of broad spectrum MMP inhibitors such as marimastat has been implicated in severe musculo-skeletal side effects. Consequently, research has been focused on designing inhibitors that selectively inhibit MMP13, thereby circumventing musculo-skeletal toxicities. A series of pyrimidine dicarboxamides were recently shown to be highly selective inhibitors of MMP13 with a novel binding mode. We have applied a molecular ruler to this exosite by dual inhibition studies involving a potent dicarboxamide in the presence of two metal chelators of different sizes. A larger hydroxamate mimic overlaps and antagonizes binding of the dicarboxamide to the exosite whereas the much smaller acetohydroxamate synergizes with the dicarboxamide. These studies elucidate the steric requirement for compounds that fit exclusively into the active site, a mandate for generating highly selective MMP13 inhibitors.  相似文献

Enterovirus 71 (EV71) is a causative agent of hand, foot, and mouth disease and is also associated with serious neurological disorders. An attenuated EV71 strain [EV71(S1-3′)] has been established in the cynomolgus monkey infection model; this strain contains the attenuation determinants derived from the type 1 poliovirus vaccine strain, Sabin 1 [PV1(Sabin)], in the 5′ nontranslated region (NTR), 3D polymerase, and 3′ NTR. In this study, we analyzed the effect of the attenuation determinants of PV1(Sabin) on EV71 infection in a NOD/SCID mouse infection model. We isolated a mouse-adapted EV71 strain [EV71(NOD/SCID)] that causes paralysis of the hind limbs in 3- to 4-week-old NOD/SCID mice by adaptation of the virulent EV71(Nagoya) strain in the brains of NOD/SCID mice. A single mutation at nucleotide 2876 that caused an amino acid change in capsid protein VP1 (change of the glycine at position 145 to glutamic acid) was essential for the mouse-adapted phenotype in NOD/SCID mice. Next, we introduced attenuation determinants derived from PV1(Sabin) along with the mouse adaptation mutation into the EV71(Nagoya) genome. In 4-week-old mice, the determinants in the 3D polymerase and 3′ NTR, which are the major temperature-sensitive determinants, had a strong effect on attenuation. In contrast, the effect of individual determinants was weak in 3-week-old NOD/SCID mice, and all the determinants were required for substantial attenuation. These results suggest that a cooperative effect of the attenuation determinants of PV1(Sabin) is essential for attenuated neurovirulence of EV71.  相似文献

We isolated a temperature-sensitive mutant, hrd4-1, deficient in ER-associated degradation (ERAD). The HRD4 gene was identical to NPL4, a gene previously implicated in nuclear transport. Using a diverse set of substrates and direct ubiquitination assays, our analysis revealed that HRD4/NPL4 is required for a poorly characterized step in ERAD after ubiquitination of target proteins but before their recognition by the 26S proteasome. Our data indicate that this lack of proteasomal processing of ubiquitinated proteins constitutes the primary defect in hrd4/npl4 mutant cells and explains the diverse set of hrd4/npl4 phenotypes. We also found that each member of the Cdc48p-Ufd1p-Npl4p complex is individually required for ERAD.  相似文献

The lipid profile of seven species of unicellular eukaryotic microalgae grown under controlled conditions was studied with emphasis on the hydrocarbons and the fatty acids as part of a search for oil-producing algae. Green, slow-growing colonies of Botryococcus braunii Kutz contained the highest lipid content of 45% based on the organic weight, with an increase to 55% under nitrogen deficiency and with no effect of sodium chloride stress. Ankistrodesmus sp. Thomas, Dunaliella spp., Isochrysis sp., Nannochloris sp. Thomas, and Nitzschia sp. Chapman contained an average of 25% lipids under nitrogen sufficient conditions. Nitrogen deficiency resulted in significant increase in the lipid content in all species but Dunaliella spp., which produced a higher content of carbohydrates. Significant low amounts of acyclic hydrocarbons were detected only in Botryococcus braunii Kutz and not in the other algae. The major hydrocarbon fractions in nitrogen deficient Botryococcus braunii Kutz, Dunaliella salina Thomas, Isochrysis spp. and Nannochloris sp. Thomas were cyclic and branched polyunsaturated components which were identified as various isoprenoid derivatives. The polar lipid composition of glycolipids and phospholipids of all species investigated was fairly typical of photosynthetic eukaryotic algae. Fatty acid composition was species specific, with changes occurring in the relative amounts of individual acids of cells cultivated under different conditions and growth phases. All species synthesized C14:0, C16.0, C18:1, C18:2 and C18:3 fatty acids; C 16:4 in Ankistrodesmus sp. Thomas; C18:4 and C 22.6 in Isochrysis sp.; C16:2, C16:3 and C20:5 in Nannochloris sp. Thomas; C16:2, C16:3 and C20:5 in Nitzschia sp. Chapman. Nitrogen deficiency and salt stress induced accumulation of C18:1 in all treated species and to a lesser extent in Botryococcus braunii Kutz. The low production of hydrocarbons under optimal growth conditions and the high production of hydrocarbons under limited growth conditions cannot support the notion that microalgae can be utilized as biosolar energy converters for the production of liquid fuel, but point to the availability of a variety of neutral and polar lipid products.  相似文献
