Graded methemoglobinemia (MetHb) was produced in unanesthetized fetal sheep to determine the effects on brain oxygenation. MetHb was induced by infusing methemoglobin-containing erythrocytes in exchange for fetal blood. During the hour after MetHb was established, fetal methemoglobin concentrations averaged 1.23 +/- 0.12 (mild MetHb), 1.71 +/- 0.13 (moderate MetHb), and 2.27 +/- 0.17 g/dl (severe MetHb). MetHb reduced mean arterial O2 content by approximately 19 (mild MetHb), 29 (moderate MetHb), and 39% (severe MetHb). The average preductal arterial PO2 fell by 1.6 (-7%), 2.8 (-11%), and 4.0 Torr (-16%) for mild, moderate, and severe MetHb, respectively. Fetal heart rate increased significantly during mild and moderate MetHb, and mean arterial pressure fell slightly during moderate and severe MetHb. The incidences of fetal breathing and eye movements were reduced in a dose-dependent manner when the calculated brain end-capillary PO2 was less than 14 Torr. We conclude that: 1) the effective capillary PO2 in the fetal brain can be significantly reduced by increasing the distance between non-methemoglobin-laden erythrocytes in capillaries and 2) hypoxic inhibition of fetal breathing probably arises from discrete areas of the brain having a PO2 less than 3 Torr.  相似文献

The disappearance of glycerol from plasma was studied after a single intravenous injection to estimate its volume of distribution (Vdist), plasma clearance rate, and rate constant for irreversible loss (kd). Studies were repeated before and after birth of the lamb to test whether loss of the placenta could account for rapidly increasing plasma concentrations in the newborn. The disappearance of glycerol was closely described by a double-exponential model in each instance. In fetal sheep Vdist averaged 0.41 +/- 0.15 (SD) 1/kg fetal wt (n = 15). This volume decreased to 0.33 +/- 0.11 l/kg (n = 8) soon after functionally removing the placenta (by snaring the umbilical cord and maintaining the fetus with intrauterine ventilation), but the change was not significant. In newborn lambs 1-3 days of age, Vdist averaged 0.45 +/- 0.11 l/kg (n = 5, NS). Plasma clearance rate also did not change significantly, averaging 7.9 +/- 2.9, 7.9 +/- 3.8, and 9.0 +/- 5.9 ml.min-1.kg-1 in the fetus, after simulated birth, and in the newborn lamb, respectively, kd also was not altered measurably and averaged 0.020 +/- 0.006, 0.024 +/- 0.007, and 0.019 +/- 0.007 min-1 during the same time periods. Similar results were obtained by using three widely different amounts of infused glycerol. The results indicate that removal of glycerol does not depend on placental function to an appreciable extent. It is concluded that plasma glycerol concentration reflects principally glycerol turnover and, hence, lipolysis before and after birth.  相似文献

The influence of various culture parameters on the attachment of a recombinant baculovirus to suspended insect cells was examined under normal culture conditions. These parameters included cell density, multiplicity of infection, and composition of the cell growth medium. It was found that the fractional rate of virus attachment was independent of the multiplicity of infection but dependent on the cell density. A first order mathematical model was used to simulate the adsorption kinetics and predict the efficiency of virus attachment under the various culture conditions. This calculated efficiency of virus attachment was observed to decrease at high cell densities, which was attributed to cell clumping. It was also observed that virus attachment was more efficient in Sf900II serum free medium than it was in IPL-41 serum-supplemented medium. This effect was attributed to the protein in serum which may coat the cells and so inhibit adsorption. A general discussion relating the observations made in-these experiments to the kinetics of recombinant baculovirus adsorption to suspended insect cells is presented.  相似文献

The randomly amplified polymorphic DNA (RAPD) assay was used to generate DNA fingerprints from clinical isolates of Staphylococcus haemolyticus isolated from patients treated with continuous ambulatory peritoneal dialysis and previously subjected to a combination of typing methods. The RAPD profiles generated with one of six randomly designed 10-mer primers allowed visual discrimination of strains. Good correlation with the original typing scheme was achieved but RAPD typing allowed discrimination of strains previously indistinguishable.  相似文献

Leaf and stem explants of passionfruit (Passiflora eadulis fv flavicarpa) were co-cultivated with a disarmed strain of Agrobacterium tunefaciens harbouring the co-integrate vector pMON200. Four plants of passionfruit were regenerated from leaf explants on agar-solidified Murashige and Skoog (1962) based medium containing 4.43 M 6-benzyl-aminopurine and supplemented with 86 M kanamycin sulphate. The four plants were rooted by transfer to MS based medium with 14.7 M 3-indolebutyric acid and 2.68 M -naphthyleneacetic acid for 7 d, followed by MS based medium lacking growth regulators. Both media used for rooting contained 172 M kanamycin sulphate. Rooted plants were potted and grown to maturity. Risk is by no means a simple concept. Natural variability and definitional problems with the concept of probability complicate the measurement and use of risk as an analytical tool. Variability requires that risk assessment methods separate natural from total risk when attempting to estimate anthropogenic risk. Failure to do so results in the over estimation of anthropogenic risk and the eventual loss of credibility for risk assessment methodologies. The common frequentist approach to probability is not consistent with anything but a modelling approach to risk assessment. When combined with its ability to account for natural variability, incorporate laboratory-assay data and offer complete statistical and experimental control, modelling is a promising approach to risk assessment. Modelling, however, is not without its drawbacks. Initialization bias can result in the over, or under, estimation of both natural and anthropogenic risk. Furthermore, model estimates are time dependent. The convergence of natural and anthropogenic risk poses problems for modelling-based risk assessment and requires clear statements as to the importance of the time dimension in risk assessment. When combined, the drawbacks to modelling-based risk assessment argue that risk should never be stated as a scalar quantity. Instead, modelling-based risk assessment should provide estimates of the complete range of risk measures (total, natural, and anthropogenic) as well as indications of convergence time. Only then can the modelling-based approach be viewed as the most appropriate means of carrying out scientifically credible risk assessment.  相似文献

We have used electron-microscopic studies, bromodeoxyuridine (BrdU) incorporation and antibody labeling to characterize the development of the Drosophila larval photoreceptor (or Bolwig's) organ and the optic lobe, and have investigated the role of Notch in the development of both. The optic lobe and Bolwig's organ develop by invagination from the posterior procephalic region. After cells in this region undergo four postblastoderm divisions, a total of approximately 85 cells invaginate. The optic lobe invagination loses contact with the outer surface of the embryo and forms an epithelial vesicle attached to the brain. Bolwig's organ arises from the ventralmost portion of the optic lobe invagination, but does not become incorporated in the optic lobe; instead, its 12 cells remain in the head epidermis until late in embryogenesis when they move in conjunction with head involution to reach their final position alongside the pharynx. Early, before head involution, the cells of Bolwig's organ form a superficial group of 7 cells arranged in a rosette pattern and a deep group of 5 cells. Later, all neurons move out of the surface epithelium. Unlike adult photoreceptors, they do not form rhabdomeres; instead, they produce multiple, branched processes, which presumably carry the photopigment. Notch is essential for two aspects of the early development of the visual system. First, it delimits the number of cells incorporated into Bolwig's organ. Second, it is required for the maintenance of the epithelial character of the optic lobe cells during and after its invagination.  相似文献

Bedrock erosional features in a small stream (Little Schultz Creek, Bibb County, Alabama) created a variety of habitats for epilithic growth. One suck habitat was illustrated by the occurrence of small falls (<0.3 m) in the main channel of the stream and blue-green algal mats associated with them. The cohesive, laminar algal mats were found at 15 such sites along a 250-m reach of the stream. The primary mat matrix consisted of the blue-green alga Oscillatoria submembranacea Ardissone and Strafforella. The uppermost portion of each mat consisted of a thin (<1 mm thick) green layer of biologically active filaments. The lower layers were thicker (up to 2 cm thick) and consisted of brown laminae of Oscillatoria filaments, and associated sediments. In addition, numerous diatoms mere associated with the mat surface. Some were loosely attached (e.g. Achnanthes); others (Cymbella tumida (Bréb.) V. H.) were stalked. These mats were present throughout the year and showed a bimodal annual distribution with maxima hi February and July. In February, total mat coverage was higher than in July. This winter maximum may have been related to a mode of growth dependent upon sedimentation from storm events and subsequent upward growth of the alga. Mat primary productivity on an areal basis (432 mg C · m?2· d?1 in March and 907 mg C · M?2· d?1 in April) was 2–12 times the maxima measured on epizoic and cobbles surfaces and other bedrock surfaces in the same stream. The limited areal coverage of the mats, when compared to other surfaces available for algal colonization, made them less important than other epilithic and epizoic surfaces in terms of total primary production in this stream reach. However, we propose that the combination of their unique structure and high primary productivity may make these algal mats sites of high algal and bacterial metabolic activity, which may include anaerobic processes in midchannel, where such activity would not be expected to occur.  相似文献

The city of Perth contains a number of sites that have been contaminated with hydrocarbons due to leakage from petroleum underground storage tanks. Microbial biomass in groundwater and sediment cores from above and below the water table, and from within and outside a plume of hydrocarbon contamination, was examined using phospholipid fatty acid methyl ester analysis. Microbial numbers, calculated from the phospholipid content, ranged from 0·9 × 106 to 7·8 × 106' Escherichia coli equivalent cells' g−1 dry wt of sediment. Over 96% of the microbial biomass was attached to the sediment and the proportion of attached cells did not decrease within the plume of contaminants. The amount of biomass within aquifer samples seemed to be related more to the proximity of the rhizosphere to the shallow aquifer, and other unknown urban inputs, rather than to the effects of the plume of contaminants. Fatty acids common to many bacterial groups dominated within the plume, and as such the analyses gave limited insight into microbial community structure. For site assessment of intrinsic remediation of shallow aquifers in urban areas, estimates of microbial biomass may not provide information that is readily applicable to plume management.  相似文献
