Dividing stem cells can give rise to two types of daughter cells; self-renewing cells that have virtually the same properties as the parent cell, and differentiating cells that will eventually form part of a tissue. The Caenorhabditis elegans germ line serves as a model to study how the balance between these two types of daughter cells is maintained. A mutation in teg-4 causes over-proliferation of the stem cells, thereby disrupting the balance between proliferation and differentiation. We have cloned teg-4 and found it to encode a protein homologous to the highly conserved splicing factor subunit 3 of SF3b. Our allele of teg-4 partially reduces TEG-4 function. In an effort to determine how teg-4 functions in controlling stem cell proliferation, we have performed genetic epistasis analysis with known factors controlling stem cell proliferation. We found that teg-4 is synthetic tumorous with genes in both major redundant genetic pathways that function downstream of GLP-1/Notch signaling to control the balance between proliferation and differentiation. Therefore, teg-4 is unlikely to function specifically in either of these two genetic pathways. Further, the synthetic tumorous phenotype seen with one of the genes from these pathways is epistatic to glp-1, indicating that teg-4 functions downstream of glp-1, likely as a positive regulator of meiotic entry. We propose that a reduction in teg-4 activity reduces the splicing efficiency of targets involved in controlling the balance between proliferation and differentiation. This results in a shift in the balance towards proliferation, eventually forming a germline tumor.  相似文献

We have designed and synthesized a novel series of pyrrolidinones as progesterone receptor partial agonists. Compounds from this series had improved AR selectivity, rat pharmacokinetic properties, and in vivo potency compared to the lead compound. With the rapid pace of the nutrition transition worldwide, understanding influences of child feeding practices within a context characterized by the co-existence of overweight and undernutrition in the same population has increasing importance. This qualitative study describes Brazilian mothers' child feeding practices and their perceptions of their association with child weight status and explores the role of socioeconomic, cultural and organizational factors on these relationships. Forty-one women enrolled in the Family Health/Community Health Workers Programme were selected from rural, urban, coastal and indigenous areas in Ceara State, north-east Brazil, to participate in four focus group discussions. Content analysis identified fourteen emergent themes showing mothers' child feeding practices in this setting were influenced by economic resources, mothers' immediate social support networks (e.g. neighbours and family members) and participation in nutrition assistance programmes. Child malnutrition was the most common nutritional concern; nevertheless, mothers were aware of the negative health consequences of obesity but misunderstood its causes (e.g. foods filled with fat would make a person fat; others thought that birth control pills and stimulants given to children were causes of obesity); several reported their own struggles with overweight. Food assistance programmes emerged as an important influence on children's dietary adequacy, especially among mothers describing dire economic situations. The findings have implications for targeting food assistance as well as health and nutrition education strategies in low-income families undergoing the nutrition transition in north-east Brazil.  相似文献

The composition of the methanogenic archaeal community in the foregut contents of Tammar wallabies (Macropus eugenii) was studied using 16S rRNA and methyl coenzyme reductase subunit A (mcrA) gene clone libraries. Methanogens belonging to the Methanobacteriales and a well-supported cluster of uncultivated archaeon sequences previously observed in the ovine and bovine rumens were found. Methanogen densities ranged from 7.0 × 105 and 3.9 × 106 cells per gram of wet weight.Kangaroos and wallabies belong to the marsupial family Macropodidae and are native to Australia. Because of their geographical isolation, macropod marsupials have evolved separately from other herbivorous animals, such as ruminants, but like ruminants, macropods have a complex gut microbiome that includes fungi, archaea, bacteria, and protozoa to coordinate plant biomass breakdown (11). The macropod foregut is functionally analogous to the rumen, yet for reasons unknown, macropod species produce relatively low levels of methane compared to ruminants (5, 13, 30).New species of bacteria (21) and protozoa (2-4) have been indentified in the macropod foregut, and the presence of fungi has also been reported (5). Preliminary studies have shown that methanogens are present in the kangaroo foregut (1) but can be absent or at levels below detection limits (22). This study represents the first attempt to describe the diversity of methanogens residing in the macropod foregut by using 16S rRNA and methyl coenzyme reductase A (mcrA) clone libraries in combination with quantitative real-time PCR.  相似文献

The objective was to examine BMI of working‐age Canadian adults in relation to occupational prestige, adjusting for other aspects of social class including household income and respondent's education. We analyzed data from 49,252 adults (age 25–64) from Cycle 2.1 of the Canadian Community Health Survey, a cross‐sectional self‐report survey conducted in 2003. Multiple linear regression was used to examine the relation between BMI and occupational prestige, adjusting for other sociodemographic variables. For women, higher ranking occupations showed lower average BMI relative to the lowest ranking occupations, but this effect was largely eliminated when adjusting for education. For men, occupation effects endured in adjusted models and we detected some evidence of a pattern whereby men in occupations characterized by management/supervisory responsibilities were heavier than those in the lowest ranking occupations (i.e., elemental sales and service). Results are interpreted in light of the symbolic value of body size in western culture, which differs for men and women. Men in positions of management/supervision may benefit from the physical dominance conveyed by a larger body size, and thus occupational prestige rankings may help us to understand the gender differences in the patterning of BMI by different indicators of social class.  相似文献

Although CD4(+) cells represent the major target for HIV infection in blood, claims of complement-independent binding of HIV-1 to erythrocytes and the possible role of Duffy blood group antigen, have generated controversy. To examine the question of binding to erythrocytes, HIV-1 was incubated in vitro with erythrocytes from 30 healthy leukapheresis donors, and binding was determined by p24 analysis and adsorption of HIV-1 with reduction of infectivity for CD4(+) target cells. All of the cells, regardless of blood group type, bound HIV-1 p24. A typical preparation of erythrocytes bound <2.4% of the added p24, but erythrocytes selectively removed essentially all of the viral infectivity as determined by decreased infection of CD4(+) target cells; however, cell-associated HIV-1 was approximately 100-fold more efficient, via trans infection, than unadsorbed virus for infection of CD4(+) cells. All of the bound HIV-1 p24 was released by treatment of the cells with EDTA, and binding was optimized by adding Ca2+ and Mg2+ during the washing of erythrocytes containing bound HIV-1. Although the small number of contaminating leukocytes in the erythrocyte preparation also bound HIV-1 p24, there was no significant binding to CD4, and it thus appears that the binding occurred on leukocytes at non-CD4 sites. Furthermore, binding occurred to erythrocyte ghosts from which contaminating leukocytes had been previously removed. The results demonstrate that erythrocytes incubated in vitro with HIV-1 differentially adsorb all of the infectious HIV-1 virions (as opposed to non-infectious or degraded virions) in the absence of complement and independent of blood group, and binding is dependent on divalent cations. By analogy with HIV-1 bound to DC-SIGN on dendritic cells, erythrocyte-bound HIV-1 might comprise an important surface reservoir for trans infection of permissive cells.  相似文献

Background

If the amount of resources allocated to reproduction (K) is fixed, then an increase in seed mass (S) can only be achieved by a decrease in seed number (n = K/S). Thus, log(n) = log(K)−log(S) producing a slope of −1 when seed mass and number are plotted on log-log axes. However, in comparative studies, empirical support for a slope of −1 is limited and contentious, leading some to question the utility of this concept.

Methodology/Principal Findings

First, we show that the expected slope depends on whether genotypes and species producing seeds of different mass are expected to reach the same adult size and that this in turn depends partly on the nature of growth. Second, we present experimental results using a population of recombinant inbred lines (RILs) of Arabidopsis thaliana. When these RILs are grown in large pots with plentiful nutrients, they exhibit a trade-off between seed size and number with a slope of −1.68 (±0.18) on log-log axes. This occurs because of genetic correlations between seed mass and adult size so that, under the right growth conditions, lines producing lighter seeds have the genetic potential to produce larger rosettes and hence a greater total mass of seeds. We re-grew lines in small pots (10 and 40 mm diameter) in a nutrient-poor substrate so that final adult size was heavily restricted by pot size.

Conclusions/Significance

Under our growth conditions, small-seeded lines were unable to produce a greater total mass of seeds. Hence a trade-off emerged between seed mass and seed number with a slope of −1.166±0.319 on log-log axes in 40-mm diameter pots (close to the expected value of −1), although the slope was 0.132±0.263 in 10-mm diameter pots, demonstrating that the nature of the trade-off is sensitive to the growth conditions.  相似文献
