Novel ω-N-amino analogs of B13 (Class E) were designed, synthesized and tested as inhibitors of acid ceramidase (ACDase) and potential anticancer agents deprived of unwanted lysosomal destabilization and ACDase proteolytic degradation properties of LCL204 [Szulc, Z. M.; Mayroo, N.; Bai, A.; Bielawski, J.; Liu, X.; Norris, J. S.; Hannun, Y. A.; Bielawska, A. Bioorg. Med. Chem. 2008, 16, 1015].Representative analog LCL464, (1R,2R)-2-N-(12′-N,N-dimethylaminododecanoyl amino)-1-(4″-nitrophenyl)-1,3-propandiol, inhibited ACDase activity in vitro, with a similar potency as B13 but higher than LCL204. LCL464 caused an early inhibition of this enzyme at a cellular level corresponding to decrease of sphingosine and specific increase of C14- and C16-ceramide. LCL464 did not induce lysosomal destabilization nor degradation of ACDase, showed increased cell death demonstrating inherent anticancer activity in a wide range of different cancer cell lines, and induction of apoptosis via executioner caspases activation. LCL464 represents a novel structural lead as chemotherapeutic agent acting via the inhibition of ACDase.  相似文献

In the course of our studies on compartment-specific lipid-mediated cell regulation, we identified an intimate connection between ceramides (Cers) and the mitochondria-dependent death-signaling pathways. Here, we report on a new class of cationic Cer mimics, dubbed ceramidoids, designed to act as organelle-targeted sphingolipids (SPLs), based on conjugates of Cer and dihydroceramide (dhCer) with pyridinium salts (CCPS and dhCCPS, respectively). Ceramidoids having the pyridinium salt unit (PSU) placed internally (alpha and gamma- CCPS) or as a tether (omega-CCPS) in the N-acyl moiety were prepared by N-acylation of sphingoid bases with different omega-bromo acids or pyridine carboxylic acid chlorides following capping with respective pyridines or alkyl bromides. Consistent with their design, these analogs, showed a significantly improved solubility in water, well-resolved NMR spectra in D(2)O, broadly modified hydrophobicity, fast cellular uptake, and higher anticancer activities in cells in comparison to uncharged counterparts. Structure-activity relationship (SAR) studies in MCF-7 breast carcinoma cells revealed that the location of the PSU and its overall chain length affected markedly the cytotoxic effects of these ceramidoids. All omega-CCPSs were more potent (IC(50/48 h): 0.6-8.0 microM) than their alpha/gamma-CCPS (IC(50/48 h): 8-20 microM) or D-erythro-C6-Cer (IC(50/48 h): 15 microM) analogs. omega-DhCCPSs were also moderately potent (IC(50/48 h): 2.5-12.5 microM). Long-chain omega-dhCCPSs were rapidly and efficiently oxidized in cells to the corresponding omega-CCPSs, as established by LC-MS analysis. CCPS analogs also induced acute changes in the levels and composition of endogenous Cers (upregulation of C16-, C14-, and C18-Cers, and downregulation of C24:0- and C24:1-Cers). These novel ceramidoids illustrate the feasibility of compartment-targeted lipids, and they should be useful in cell-based studies as well as potential novel therapeutics.  相似文献

Renal failure is associated with aortic valve calcification. Using our rat model of uremia-induced reversible aortic valve calcification, we assessed the role of apoptosis and survival pathways in that disease. We also explored the effects of raloxifene, an estrogen receptor modulator, on valvular calcification. Gene array analysis was performed in aortic valves obtained from three groups of rats (n = 7 rats/group): calcified valves obtained from rats fed with uremic diet, valves after calcification resolution following diet cessation, and control. In addition, four groups of rats (n = 10 rats/group) were used to evaluate the effect of raloxifene in aortic valve calcification: three groups as mentioned above and a fourth group fed with the uremic diet that also received daily raloxifene. Evaluation included imaging, histology, and antigen expression analysis. Gene array results showed that the majority of the altered expressed genes were in diet group valves. Most apoptosis-related genes were changed in a proapoptotic direction in calcified valves. Apoptosis and decreases in several survival pathways were confirmed in calcified valves. Resolution of aortic valve calcification was accompanied by decreased apoptosis and upregulation of survival pathways. Imaging and histology demonstrated that raloxifene significantly decreased aortic valve calcification. In conclusion, downregulation of several survival pathways and apoptosis are involved in the pathogenesis of aortic valve calcification. The beneficial effect of raloxifene in valve calcification is related to apoptosis modulation. This novel observation is important for developing remedies for aortic valve calcification in patients with renal failure.  相似文献

BackgroundThere is sparse literature on whether training in endobronchial ultrasound (EBUS)-guided transbronchial needle aspiration (TBNA) improves the diagnostic yield of conventional TBNA (cTBNA).ObjectivesThe aim of this study was to evaluate the diagnostic yield of cTBNA before and after the introduction of EBUS.MethodsThis was a retrospective analysis of patients who underwent cTBNA at our center. The study was divided into two periods, before and after the introduction of EBUS at our facility. The diagnostic yield of cTBNA was compared between the study periods. Rapid on-site cytological examination was not available.ResultsA total of 1,050 patients (61.6% men; mean age 45.6 years) underwent cTBNA during the study period (849 before EBUS; 201 after EBUS). Sarcoidosis (n = 527) followed by bronchogenic carcinoma (n = 222) formed the most common indications for performing cTBNA. There was a significant increase in both the success of obtaining a representative sample (from 71% to 85%), and the diagnostic yield (from 33% to 49.5%) of cTBNA, after the introduction of EBUS. The increase in the diagnostic yield of cTBNA after introduction of EBUS remained significant even after adjusting for years of performing cTBNA and the type of anesthesia (topical vs. sedation and topical) on a multivariate analysis.ConclusionThe diagnostic yield of cTBNA at our facility increased after the introduction of EBUS-TBNA. However, given the retrospective nature of the study, prospective studies are required to confirm our findings.  相似文献

InRita rita the upper lip is associated with the rostral cap and the lower lip with a skin fold. The epithelia of the lips, which are modified with respect to the peculiar feeding behaviour of the fish, are mucogenic. The superficial layer of epithelial cells, in addition to the mucous cells, are involved in active secretion of mucopolysaccharides. Mucus, apart from its diverse functions, as in fish epidermis, lubricates the epithelia giving protection against possible mechanical injury during the searching and catching of prey from bottom debris. Furthermore, the epithelia are thick, an adaptation to their mucogenic nature, so as to provide additional protection. The contents of the club cells are proteinaceous, which correlates with the helical filaments reported in similar cells in fish epidermis. Characteristic ridges on the surface of the lips are considered to be an adaptation to assist in manipulation during the procuring of food. Mucus secreted in the grooves between them reduces friction and prevents the deposition of foreign matter. The skin fold facilitates an increase in gape. The taste buds and ampullary organs, which on the lips and the rostral cap are associated with the localization of prey and triggering of a pick-up reflex, are not located on the skin fold, as the latter often remains concealed between the lower lip and the ventral head skin. The importance of histochemical analysis as a supplement to histological studies is emphasised.  相似文献

The aim of the present study was to evaluate the role of arbuscular mycorrhizal (AM) fungi on metal uptake, oxidative effects and antioxidant defence mechanisms under cadmium (Cd) and lead (Pb) stresses in Cajanus cajan (L.) Millsp. (pigeonpea). Treatments consisted of two concentrations each of Cd (25 and 50 mg/kg of soil) and Pb (500 and 800 mg/kg of soil) singly as well as in combination. Both metals induced oxidative damage through increased lipid peroxidation, electrolyte leakage and hydrogen peroxide levels, but Cd was found to be more toxic than Pb. Compared with the effects of Cd or Pb alone, the combination of Cd and Pb acted synergistically; however, Pb immobilisation in soil controlled the uptake of Cd in plants. There was a direct correlation between the type of genotype, heavy metal content and oxidative damage in concentration dependent manner. Superoxide dismutase (SOD), catalase (CAT) and peroxidase (POX) increased under stress. The toxicity symptoms of the metal stress were significantly higher in Sel-141-97 genotype when compared with Sel-85 N. The high ratio of glutathione to its oxidised form, glutathione disulfide (GSH/GSSG), could be restored by means of higher glutathione reductase (GR) activity and increased GSH synthesis in mycorrhizal stressed plants. AM inoculations with Glomus mosseae significantly arrested uptake of Cd and Pb into the root system and further translocation into the above ground parts and led to decreased lipid peroxidation and electrolyte leakage. Increased activities of SOD, CAT, POX as well as GR were observed in all mycorrhizal stressed plants.  相似文献
