Developing new methods for modelling infectious diseases outbreaks is important for monitoring transmission and developing policy. In this paper we propose using semi-mechanistic Hawkes Processes for modelling malaria transmission in near-elimination settings. Hawkes Processes are well founded mathematical methods that enable us to combine the benefits of both statistical and mechanistic models to recreate and forecast disease transmission beyond just malaria outbreak scenarios. These methods have been successfully used in numerous applications such as social media and earthquake modelling, but are not yet widespread in epidemiology. By using domain-specific knowledge, we can both recreate transmission curves for malaria in China and Eswatini and disentangle the proportion of cases which are imported from those that are community based.  相似文献

There is little information about habitat use pattern of river lapwing in India as well as in Southeast Asia. In this regard, we employed the line transect method to quantify the density of river lapwing concerning vegetation height, water depth and soil penetrability. Scan and focal sampling methodology was adapted to investigate the activities (foraging and roosting) and breeding events. The results showed that invertebrates were abundant at 4 cm of soil penetrability, particularly in open unvegetated river bank (OURB) and open unvegetated river island (OURI) habitat types. River lapwings were significant in short vegetation. They occurred in high density in the vicinity of river water at 5 cm of water depth and 4 cm of soil penetrability in OURB and OURI habitat types. Human activities also influenced the density of river lapwing in different habitat types. Results of GLM analysis revealed that all interactions strongly influenced the density of river lapwings except two interactions, e.g. vegetation height*water depth and vegetation height*soil penetrability. Moreover, we found that river lapwing foraged more frequently in OURB and OURI habitat types while less in vegetated river bank (VRB) and crop fields (CF). Roosting activities were more frequently observed in VRB and CF habitat types. Different breeding events (nest seen, chick seen, alarm calling and territory defence) were primarily seen in OURB and OURI habitat types. Finally, developing greater understanding of the river lapwing could help us to design effective conservation measures for this species.

TQ (thymoquinone), the bioactive constituent of black seed (Nigella sativa), has been shown to inhibit the growth of various human cancers both in vitro and in vivo. This study reports the radiosensitizing effect of TQ on human breast carcinoma cells (MCF7 and T47D). TQ in combination with single dose of ionizing radiation (2.5 Gy) was found to exert supra-additive cytotoxic effects on both the carcinomas as measured by cell proliferation and colony-formation assays. Annexin V binding and FACS analysis revealed the role of enhanced apoptosis and cell cycle modulation in the mechanism of TQ-mediated radiosensitization, thus supporting TQ as an adjuvant for preclinical testing in cancer chemo-radiotherapy.  相似文献

In the present study we tested the hypothesis whether hyperhomocysteinemia, an elevated homocysteine level, induces venous phenotype in artery. To test our hypothesis, we employed wild type (WT) and cystathionine β-synthase heterozygous (+/-) (CBS+/-) mice treatment with or without folic acid (FA). Aortic blood flow and velocity were significantly lower in CBS+/-mice compared to WT. Aortic lumen diameter was significantly decreased in CBS+/-mice, whereas FA treatment normalized it. Medial thickness and collagen were significantly increased in CBS+/-aorta, whereas elastin/collagen ratio was significantly decreased. Superoxide and gelatinase activity was significantly high in CBS+/-aorta vs WT. Western blot showed significant increase in MMP-2, -9,-12, TIMP-2 and decrease in TIMP-4 in aorta. RT-PCR revealed significant increase of vena cava marker EphB4, MMP-13 and TIMP-3 in aorta. We summarize that chronic HHcy causes vascular remodelling that transduces changes in vascular wall in a way that artery expresses vein phenotype.  相似文献

Macrophages have been shown to kill Mycobacterium tuberculosis through the action of the antimicrobial peptide cathelicidin (CAMP), whose expression was shown to be induced by 1,25-dihydroxyvitamin D3 (1,25D3). Here, we investigated in detail the antimycobacterial effect of murine and human cathelicidin against Mycobacterium smegmatis and M. bovis BCG infections. We have synthesized novel LL-37 peptide variants that exhibited potent in vitro bactericidal activity against M. smegmatis, M. bovis BCG and M. tuberculosis H37Rv, as compared with parental peptide. We show that the exogenous addition of LL-37 or endogenous overexpression of cathelicidin in macrophages significantly reduced the intracellular survival of mycobacteria relative to control cells. An upregulation of cathelicidin mRNA expression was observed that correlated with known M. smegmatis killing phases in J774 macrophages. Moreover, RNAi-based Camp knock-down macrophages and Camp(-/-) bone marrow derived mouse macrophages were significantly impaired in their ability to kill mycobacteria. M. smegmatis killing in Camp(-/-) macrophages was less extensive than in Camp(+/+) cells following activation with FSL-1, an inducer of cathelicidin expression. Finally we show that LL-37 and 1,25D3 treatment results in increase in colocalization of BCG-containing phagosomes with lysosomes. Altogether, these data demonstrate that cathelicidin plays an important role in controlling intracellular survival of mycobacteria.  相似文献

The level of chromium (Cr) contamination in soils and irrigated mine wastewater at South Kaliapani chromite mine region of Orissa, (India) were investigated. Chromium bioaccumulation in rice plants (Oryza sativa L. cv. Khandagiri) irrigated with Cr+6 contaminated mine wastewater was analyzed along with its attenuation from mine wastewater. The levels of Cr+6 in irrigated mine wastewaters in successive rice grown plots were analyzed on 75 days and 100 days after transplantation of seedlings. Total chromium content in different parts of rice plants and soil samples from different plots was analyzed during harvesting stage (125 days after transplantation). Cr accumulation was significantly high in surface soils (0-20 cm) with a mean value of 11,170 mg kg(-1), but it decreased significantly after the crop harvest. About 70% to 90% reduction of Cr+6 levels was observed in irrigated mine wastewater when passed through successive rice plots. High bio-concentration of Cr in leaves with values ranging from 125-498 mg kg(-1) as compared to stem (25-400 mg kg(-1)) and grain (5-23 mg kg(-1)) was noticed. The reduction of Cr+6 levels is related to plant age, high biomass and area of water passage and was attributed to rhizofiltration technique.  相似文献

Raloxifene hydrochloride (R-HCl), a BCS class II drug, remains a mainstay in the prevention and pharmacologic therapy of osteoporosis. Its absolute bioavailability, however, is 2% due to poor solubility and extensive first pass metabolism. The present study describes two simultaneous approaches to improve its bioavailability, complexation of R-HCl with cyclodextrin(s), and formulation of mucoadhesive microspheres of the complex using different proportions of carbopol and HPMC. Microspheres were pale yellow in color, free-flowing, spherical, and porous in outline. The particle size ranged between 3 and 15 μm, and entrapment efficiency was found to be within 81.63% to 87.73%. A significant improvement in the solubility of R-HCl was observed, and it differed with the combination of excipients used. X-ray diffraction and differential scanning calorimetry studies revealed that enhancement in drug solubility was resulted due to a change in its crystallinity within the formulation. Microspheres possessed remarkable mucoadhesion and offered controlled drug release, lasting up to 24 h. They produced a sharp plasma concentration–time profile of R-HCl within 30 min post-administration to Wistar rats. [AUC]0–24 h was found to be 1,722.34 ng h/ml, and it differed significantly to that of pure drug powder (318.28 ng h/ml). More than fivefold increase in AUC and more than twofold increase in MRT were observed. FT-IR studies evidenced no interaction among drug and excipients. The results of this study showed that mucoadhesive microspheres could be a viable approach to improve the pharmacokinetic profile of R-HCl.  相似文献
