We studied the role of oxidative stress and the effect of vinpocetine (1.5, 3 or 6 mg/kg) and piracetam (150 or 300 mg/kg)
in acute demyelination of the rat brain following intracerebral injection of ethidium bromide (10 μl of 0.1%). Results: ethidium
bromide caused (1) increased malondialdehyde (MDA) in cortex, hippocampus and striatum; (2) decreased total antioxidant capacity
(TAC) in cortex, hippocampus and striatum; (3) decreased reduced glutathione (GSH) in cortex and hippocampus (4); increased
serum nitric oxide and (5) increased striatal (but not cortical or hippocampal) acetylcholinesterase (AChE) activity. MDA
decreased in striatum and cortex by the lower doses of vinpocetine or piracetam but increased in cortex and hippocampus and
in cortex, hypothalamus and striatum by the higher dose of vinpocetine or piracetam, respectively along with decreased TAC.
GSH increased by the higher dose of piracetam and by vinpocetine which also decreased serum nitric oxide. Vinpocetine and
piracetam displayed variable effects on regional AChE activity. 相似文献

Environmental pollution is one of the ever surging problems receiving careful attention in Egypt, as well as in the world. It has been evidenced that environmental pollutants play an important role in the manifestations of severe reproductive-associated disorders. Among them, an environmental pollutant, Cd, has contributed to several deleterious effects. Maternal exposure to Cd is associated with low birthweight and an increase in spontaneous abortion. Spontaneous abortion is highly frustrating for both patients and physicians. The data from the present study reveal that there was a significant increase in blood level of Cd in the aborter groups as compared to the control group of healthy women. Also there was a significant decrease in serum ferritin, which correlated positively with abortion. Also, plasma levels of malondialdehyde were significantly elevated in aborter groups. There was also a highly significant increase in plasma 8-hydroxyl-2-deoxyguanosine in the aborter groups as compared to the control group, accompanied by a significant decrease in plasma carnosine in aborter groups as compared with the control group. In the same manner there was a reduction in the progesterone hormone in aborter groups. Our results suggest that exposure to Cd is associated with an increased risk of recurrent spontaneous abortion in Egyptian women. 相似文献

Pilosocereus robinii is a rare species which is experiencing sudden population collapse. Identifying and developing effective conservation and management strategies to halt the forestall extinction of this species is crucial. The present study was conducted to assess the best conditions for in vitro propagation of this plant in regard to its morphogenic, genetic as well as the chemical potentials. A successful in vitro propagation system of P. robinii has been developed. MS hormone-free medium induced the best root morphogenic potential. The plants were acclimatized in the greenhouse at 100% survival rate. Besides, the somaclonal variations between the in vitro raised plants were analyzed using PCR-ISSR markers and SDS–PAGE protein, where the regenerated explants on MS medium supplemented with TDZ were the highest in inducing new specific marker bands. Sh6 ISSR primer showed the highest polymorphism value, 81.8% with 33 total amplified fragments, while Sh3 ISSR primer showed the lowest value with polymorphic percentage of 14.3%. Furthermore, SDS–PAGE protein analysis showed no variation in protein pattern of the studied treatments. On the other side, HPLC analysis of the in vitro plantlets extracts has shown that 2iP based treatments were the highest in organic acids accumulation, while the phenolic constituents' accumulation was found to reach its peak in the BA based treatments. 相似文献

Tamoxifen (TAM) is used for the treatment and prevention of estrogen receptor positive breast cancer. However, the limited activity, toxicity and the development of resistance raised the current need for new potent nontoxic antiestrogen. Six novel TAM analogues 5a–f were synthesized using McMurry olefination reaction. Replacement of the dimethylamino group in TAM by piperidino, piperazino or N-methyl piperazino, substituting the phenyl ring with florine atom at p-position and changing the ethyl group by methyl, afforded compounds showing comparable activity to TAM (1). Compounds 5c and 5e showed significant increase in antiproliferative activity in two breast cancer cell lines (MCF-7 and MDA-MB-231) compared to tamoxifen, while other compounds showed similar activity. The increased anticancer activity of compounds 5c and 5e was attributed to their ability to induce ER-independent cell death. 相似文献

Although CD8(+) T lymphocytes targeting lytic infection proteins dominate the immune response to acute and persistent EBV infection, their role in immune control of EBV replication is not known. Rhesus lymphocryptovirus (rhLCV) is a gamma-herpesvirus closely related to EBV, which establishes persistent infection in rhesus macaques. In this study, we investigated cellular immune responses to the rhLCV BZLF1 (rhBZLF1) homolog in a cohort of rhLCV-seropositive rhesus macaques. rhBZLF1-specific IFN-gamma ELISPOT responses ranging between 56 and 3070 spot-forming cells/10(6) PBMC were detected in 36 of 57 (63%) rhesus macaques and were largely mediated by CD8(+) T lymphocytes. The prevalence and magnitude of ELISPOT responses were greater in adult (5-15 years of age) rather than juvenile macaques (<5 years of age), suggesting that rhBZLF1-specific CTL increase over time following early primary infection. A highly immunogenic region in the carboxyl terminus of the rhBZLF1 protein containing overlapping CTL epitopes restricted by Mamu-A*01 and other as yet unidentified MHC class I alleles was identified. The presence of a robust CD8(+) T lymphocyte response targeting this lytic infection protein in both rhesus macaques and humans suggests that these CTL may be important for immune control of EBV-related gamma-herpesvirus infection. These data underscore the utility of the rhLCV-macaque model for studies of EBV pathogenesis. 相似文献

Hydrogel matrices based on starch and dimethylaminoethyl methacrylate (Starch/DMAEMA) were synthesized including γ-irradiation as a clean initiator. The prepared hydrogels were characterized in terms of their gel fraction yield, degree of equilibrium swelling. The prepared hydrogels were examined as carriers for immobilization of Bacillus subtilis that has the ability to secrete an extracellular fibrinolytic enzyme that degrades fibrin. Scanning electron microscope (SEM) analysis showed proliferation of the bacterial cells entrapped inside the polymeric matrix. The immobilization process increases the production time of fibrinolytic enzyme up to 120 h instead of 96 h for the free cells. The optimum temperature of activity broadened and a significant shift in the pH optima was observed upon immobilization. The reusability of immobilized cells under repeated batch fermentation conditions was also investigated. At the optimum production conditions, immobilization of B. subtilis cells onto Starch/DMAEMA resulted in a four fold increase in enzyme activity. 相似文献

Nineteen new compounds containing tetrazole and/or cyanamide moiety have been designed and synthesised. Their structures were confirmed using spectroscopic methods and elemental analyses. Anti-inflammatory activity for all the synthesised compounds was evaluated in vivo. The most active compounds 4c, 5a, 5d–f, 8a and b and 9a and b were further investigated for their ulcerogenic liability and analgesic activity. Pyrazoline derivatives 9b and 8b bearing trimethoxyphenyl part and SO2NH2 or SO2Me pharmacophore showed equal or nearly the same ulcerogenic liability (UI: 0.5, 0.75, respectively), to celecoxib (UI: 0.50). Most of tested compounds showed potent central and/or peripheral analgesic activities. Histopathological investigations were done to evaluate test compounds effect on rat's gastric tissue. The obtained results were in consistent with the in vitro data on COX evaluation. Docking study was also done for all the target compounds inside COX-2-active site. 相似文献

The concentrations of major (Al, Fe, Mg, Ti, K and Na) and minor (Ba, Sr, V, Cu, Mn, Cr, Ni, Zn, Pb and Mo) elements as well as carbonate, organic carbon and total nitrogen have been determined in surface sediments collected at 12 stations from the Bardawil lagoon. The aim of the study was to characterize the geochemistry of the sediments in three different environments, the lagoon, the salt pans and the inlet between the lagoon and the Mediterranean Sea. Higher CaCO3 percentage (53.5–70.5%) were found in the salt pans where biogenic calcareous components and carbonate rock fragments were found in sufficient quantities in the sediment fractions. Based on high C/N ratios, the organic carbon fraction of the Bardawil lagoon surface sediments is clearly dominated by terrigenous material. The distribution of Al, Fe, Mg and Ti are essentially controlled by the mineralogy of the sediments. The ratios of Ba, Sr, Cu, Mn, Pb and Mo to Al are all high in the salt pans and reflect changes in mineralogy and sediment texture. 相似文献

The numbers of T lymphocytes and T cell subsets (CD2(+), CD3(+), CD4(+), CD8(+)), activated T cells (CD26(+)), B cells (CD19(+)), granulocytes (CD15(+)) and natural killer cells (CD16/56) were monitored by flow cytometry in 79 kidney transplant recipients, 35 of whom had cytomegalovirus infection. The percentages of these cells were correlated with viral load, as determined by cytomegalovirus antigenemia. Development of cytomegaloviral infection coincided with a significant reduction in the percentages of CD4(+) (P < 0.005) and CD3(+) (P < 0.05) cells. Monitoring of lymphocyte subsets may provide useful information on immunological events during cytomegaloviral infection. 相似文献
