Functions of the epididymis differ by region, and this may be reflected in epithelial structure. Therefore, tissues from the initial segment (IS), proximal and central caput (PCap, CCap), and proximal and central corpus (PCor, CCor) epididymidis were examined by light and transmission electron microscopy. The proportion of principal cells in the epithelium was highest (p less than 0.05) in the CCap (74%) and lowest in the CCor (68%), whereas proportions of basal cells (25%), apical cells (1.4%), and white blood cells (2%) were similar in all regions. Volume density (VD) of the nucleus was lower (p less than 0.05) in principal cells in the IS (7%) than in other regions (10%). There was no regional difference in VD of the Golgi complex (14%) or endoplasmic reticulum (19%) in principal cells. The VD of mitochondria averaged 4% in the IS through CCap, but only 2.5% in PCor or CCor (p less than 0.05). The VD of clear vesicles + multivesicular bodies (8%) and dense vesicles (6%) were higher (p less than 0.05) in the CCap than in other regions (1% each), while there were more lipid droplets (12%) in the PCor than in other regions (less than or equal to 2%). Most quantitative differences in VD of organelles within principal cells were small even though significant. Extragonadal reserves totalled 89 X 10(9) spermatozoa for 5--16-year-old sexually rested stallions and 60 X 10(9) for 2--4-year-olds. Regardless of age, the cauda epididymidis contained 62% of the total reserves and the vas deferens, including the ampulla, contained 7% of the total reserves of spermatozoa. The caput plus corpus epididymidis from 5--16-year-old stallions (N = 41) contained 14.9 X 10(9) spermatozoa per side as compared (P less than 0.01) to 8.5 X 10(9) for 2--4-year olds (N = 30). Frequency of ejaculation did not influence the number of spermatozoa found in caput plus corpus epididymidis. Across all ages, the number of spermatozoa potentially available for ejaculation from the cauda epididymidis and vas deferens on each side totalled 54 X 10(9). Collection of 5 successive ejaculates from sexually rested stallions removed 40--60% of the available spermatozoa while ejaculation every 2nd day reduced (P less than 0.05) the number of spermatozoa potentially available for ejaculation by 27%. OSF-1 (also known as pleiotrophin, HB-GAM, HBGF-8 or HBNF) is a heparin-binding, neurotrophic protein. Its tissue-specific expression in rats is developmentally regulated and the protein is highly conserved between species. The protein is believed to be involved in neuronal development. Previous experiments in our laboratory showed that OSF-1 is primarily expressed in brain and bone. The biological function of OSF-1 in bone is unknown. In order to overcome the limited availability of the native protein, we now report on the high-level expression of human OSF-1 in Escherichia coli. The protein is present in the form of inclusion bodies, which were isolated and solubilized. The partially purified protein was refolded and further purified employing heparin sepharose chromatography. N-terminal sequence determination revealed the same amino acid sequence as the natural mature protein. The isolated backfolded recombinant human OSF-1 did promote neurites outgrowth in primary cultures of cortical neurons. 相似文献
