Aggregation of protein into insoluble intracellular complexes and inclusion bodies underlies the pathogenesis of human neurodegenerative diseases. Importance of cytochrome c (cyt c) arises from its involvement in apoptosis, sequence homology and for studying molecular evolution. A systemic investigation of polyethylene glycol (PEG) and trifluoroethanol (TFE) on the conformational stability of cyt c as a model hemeprotein was made using multi-methodological approach. Cyt c exists as molten globule (MG) at 60 % PEG-400 and 40 % TFE as confirmed by far-UV CD, attenuated total reflection Fourier transform infrared spectroscopy, Trp environment, 8-anilino-1-naphthalene-sulfonic acid (ANS) binding and blue shift in the soret band. Q-band splitting in MG states specifies conformational changes in the hydrophobic heme-binding pocket. Aggregates were detected at 90 % PEG-400 and 50 % TFE as confirmed by increase thioflavin T and ANS fluorescence and shift in Congo red absorbance. Detection of prefibrils and protofibrils at 90 % PEG-400 and 50 % TFE was possible after 72-h incubation. Single cell gel electrophoresis of prefibrils and protofibrils showed DNA damage confirming their toxicity and potential health hazards. Nanoemulsions are increasingly being investigated for their fascinating capability of loading both hydrophobic and hydrophilic molecules while their stability is still an issue, being affected by various factors. In this study, to evaluate the dominant factors affecting the stability of nanoemulsions, artificial neural networks (ANNs) were implemented. Nanoemulsions of almond oil in water containing oleic acid-coated superparamagnetic iron oxide nanoparticles were prepared using a mixture of Tween 80 and Span 80 as surfactant system and ethanol as a co-surfactant. The ratio of transparency of the samples at 30 min and 7 days after preparation was taken as an indication of the stability of samples. Four independent variables, namely, concentration of nanoparticle, surfactant, oil, and alcohol were investigated to find their relations with the dependent variable (i.e., transparency ratio). Using ANNs modeling, it was concluded that the stability is affected by all variables, with all variables showing reverse effect on the stability beyond an optimum amount.Key words: artificial neural networks, nanoemulsion, optimization, stability, superparamagnetic iron oxide 相似文献

Artificial neural networks (ANNs) were used in this study to determine factors that control the polydispersity index (PDI) in an acetaminophen nanosuspension which was prepared using nanoprecipitation in microfluidic devices. The PDI of prepared formulations was measured by dynamic light scattering. Afterwards, the ANNs were applied to model the data. Four independent variables, namely, surfactant concentration, solvent temperature, and flow rate of solvent and antisolvent were considered as input variables, and the PDI of acetaminophen nanosuspension was taken as the output variable. The response surfaces, generated as 3D graphs after modeling, were used to survey the interactions happening between the input variables and the output variable. Comparison of the response surfaces indicated that the antisolvent flow rate and the solvent temperature have reverse effect on the PDI, whereas solvent flow rate has direct relation with PDI. Also, the effect of the concentration of the surfactant on the PDI was found to be indirect and less influential. Overall, it was found that minimum PDI may be obtained at high values of antisolvent flow rate and solvent temperature, while the solvent flow rate should be kept to a minimum. 相似文献

This study investigates the effect of pentose sugars (ribose and arabinose) on the structural and chemical modifications in glucose oxidase (GOD) as well as genotoxic potential of this modified form. An intermediate state of GOD was observed on day 12 of incubation having CD minima peaks at 222 and 208?nm, characteristic of α-helix and a few tertiary contacts with altered tryptophan environment and high ANS binding. All these features indicate the existence of molten globule state of the GOD with ribose and arabinose on day 12. GOD on day 15 of incubation forms β structures as revealed by CD and FTIR which may be due to its aggregation. Furthermore, GOD on day 15 showed a remarkable increase in Thioflavin T fluorescence at 485?nm. Comet assay of lymphocytes and plasmid nicking assay in presence of glycated GOD show DNA damage which confirmed the genotoxicity of advance glycated end products. Hence, our study suggests that glycated GOD results in the formation of aggregates and the advanced glycated end products, which are genotoxic in nature. 相似文献

Two platinum(IV) complexes, [Pt(4bt)Cl4] (4) and [Pt(dpyam)Cl4]·DMF (5) (where 4bt is 4,4′-bithiazole and dpyam is 2,2′-dipyridylamine) were prepared from the reaction of H2PtCl6·6H2O with 4,4′-bithiazole and 2,2′-dipyridylamine, respectively, in methanol. Both complexes were fully characterized and their structures were determined by the X-ray diffraction method. These complexes have a bidentate nitrogenous ligand with four chloride anions attached to a Pt(IV) metal in a distorted octahedral environment. These complexes along with three previously reported analogous complexes were used for in vitro cytotoxicity evaluation against four cultures, NIH-3T3, Caco-2, HT-29 and T47D by MTT assay. The methyl group position in the ligand plays an important role in the cytotoxicity of relevant compounds in different cultures. Interestingly, in some cases, the IC50 values of the new complexes were higher for normal cells but lower against cancer cells in comparison with cisplatin, especially in T47D (breast ductal carcinoma). 相似文献

Parkinson’s disease (PD) is a slowly progressive neurodegenerative movement disorder. Apoptosis, neuroinflammation, and oxidative stress are the current hypothesized mechanisms for PD pathogenesis. Tetramethylpyrazine (TMP), the major bioactive component of Ligusticum wallichii Franchat (ChuanXiong), Family Apiaceae, reportedly has anti-apoptotic, anti-inflammatory and antioxidant effects. This study investigated the role of ‘TMP’ in preventing rotenone-induced neurobiological and behavioral sequelae. A preliminary dose–response study was conducted where rats received TMP (10, 20, and 40 mg/kg, i.p.) concomitantly with rotenone (2 mg/kg, s.c.) for 4 weeks. Catalepsy, locomotor activity, striatal dopamine content, and tyrosine hydroxylase “TH” and α-synuclein immunoreactivity were evaluated. The selected TMP dose (20 mg/kg) was used for western blot analysis of Bax, Bcl2, and DJ-1, immunohistochemical detection of nuclear factor kappa B (NF-кB), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX2), and glial fibrillary acidic protein (GFAP) expression, in addition to biochemical analysis of caspase-3 activity, nuclear factor erythroid 2-related factor 2 (Nrf2), and heme oxygenase-1 (HO-1) levels. Results showed that TMP (20 mg/kg) significantly improved midbrain and striatal TH expression and striatal dopamine content as well as the motor deficits, compared to rotenone-treated group. These results were correlated with reduction in caspase-3 activity and α-synuclein expression, along with improvement of midbrain and striatal Bax/Bcl2 ratio compared to rotenone-treated group. TMP also attenuated rotenone-induced upregulation of Nrf2/HO-1 pathway. Furthermore, TMP downregulated rotenone-induced neuroinflammation markers: NF-кB, iNOS, COX2, and GFAP expression in both the midbrain and striatum. Taken together, the current study suggests that TMP is entitled to, at least partially, preventing PD neurobiological and behavioral deficits by virtue of its anti-apoptotic, anti-inflammatory, and antioxidant actions. 相似文献
