A clinical isolate of Pseudomonas aeruginosa, JES, was resistant to extended-spectrum cephalosporins with a marked synergistic effect with clavulanic acid on a routine antibiogram. Preliminary PCR analysis revealed the presence of blaVEB-1, an integron-located gene encoding an extended-spectrum beta-lactamase previously identified in Escherichia coli MG-1. Using class 1 integron primers and blaVEB-1 intragenic primers, the insert region of the blaVEB-1 containing integron along with some flanking sequence from P. aeruginosa JES was amplified and subsequently sequenced. In50 contains within its variable region, in addition to qacE delta 1 and sull genes commonly found in class 1 integrons, two gene cassettes, veb1 and aadB. In50 is peculiar since its attI1 site is interrupted by two novel insertion sequences, IS1999 and IS2000. P. aeruginosa JES and Escherichia coli MG-1 strains were isolated from patients previously hospitalized in south east Asian countries. The finding of blaVEB-1 in these strains and on different integrons underlines the interspecies spread of this integron-located extended-spectrum beta-lactamase gene.  相似文献

Seeds of tomato (Lycopersicon lycopersicum var. Peto 86) were planted in soil inoculated with two vesicular-arbuscular mycorrhizal isolates (Glomus intraradices Schenck & Smith). After 7 weeks, plants were transplanted to the field and sprayed twice with cytokinin and thidiazuron, at 50% flowering and 3 weeks later. These treatments significantly increased the percentage of infected roots (30–43%) and chlorophyll content (8–13%). Mycorrhizal plants had less carbohydrates in the roots than in the leaves. Total protein and phenylalanine contents showed pronounced increases, while proline content decreased. Treatment of the mycorrhizal plants with thidiazuron or cytokinin significantly increased plant dry weight (51–54%), and the tomato yield significantly increasecd after inoculation with both isolates (18–35% and 14–32%).  相似文献

The purified, specific polysaccharide from Vibrio cholera type NAG, NV 384, O-antigen, 2A, 2Bhuman, contains glucose (5.14%), galactose (4.21%), mannose (64.8%), xylose (3.16%), arabinose (1.98%), fucose (1.50%), mannuronic acid (14.3%), phosphate (0.32%), 2-amino-2-deoxy-D-glucose (2.9%), and 2-amino-2-deoxy-D-galactose (1.0%). Various reactions have shown that the material comprises a phosphoric diester-linked polysaccharide containing mainly (1→2)-linked mannopyranose residues that are highly branched with other sugar residues.  相似文献

Tendon matrix integrity is vital to ensure adequate mechanical properties for efficient function. Although historically tendon was considered to be relatively inert, recent studies have shown that tendon matrix turnover is active. During normal physiological activities some tendons are subjected to stress and strains much closer to their failure properties than others. Tendons with low safety margins are those which function as energy stores such as the equine superficial digital flexor tendon (SDFT) and human Achilles tendon (AT). We postulate therefore that energy storing tendons suffer a higher degree of micro-damage and thus have a higher rate of matrix turnover than positional tendons. The hypothesis was tested using tissue from the equine SDFT and common digital extensor tendon (CDET). Matrix turnover was assessed indirectly by a combination of measurements for matrix age, markers of degradation, potential for degradation and protein expression. Results show that despite higher cellularity, the SDFT has lower relative levels of mRNA for collagen types I and III. Non-collagenous proteins, although expressed at different levels per cell, do not appear to differ between tendon types. Relative levels of mRNA for MMP1, MMP13 and both pro-MMP3 and MMP13 protein activity were significantly higher in the CDET. Correspondingly levels of cross-linked carboxyterminal telopeptide of type I collagen (ICTP) were higher in the CDET and tissue fluorescence lower suggesting more rapid turnover of the collagenous component. Reduced or inhibited collagen turnover in the SDFT may account for the high level of degeneration and subsequent injury compared to the CDET.  相似文献

Three strains ofStreptomyces rimosus were grown on four different media. The one suitable for the production of oxytetracycline byStreptomyces rimosus 12907 was modified by black strap molasses, fodder yeast (40% total protein) and rice bran. The volume of the fermentation medium was scaled up in a 1200-litre fermentor aerated with sterile air obtained from a system used in the purification of air. 850 g crude oxytetracyeline was obtained when the fermented medium (700 litres) was extracted with 1-butanol. In some spiders, a discrete portion of the male's copulatory organ (the apical sclerite) breaks off during copulation and remains in the female's reproductive tract. Apical sclerites may prevent insemination by rivals (sperm competition), stimulate females to favourably bias paternity (cryptic choice) or breakage may reflect sexual conflict over copulation duration with little direct effect on paternity. It has been assumed that any benefits of organ breakage are balanced by a large cost (male sterility) in species where males could otherwise mate multiply, but this has never been experimentally tested. We examined these ideas in the Australian redback spider (Latrodectus hasselti Thorell 1870, Araneae: Theridiidae), a species where males are functionally sterile after one normal mating. We experimentally removed sclerites and found males were able to mate, had similar copulation durations and transferred similar numbers of sperm as males with intact sclerites. Benefits of organ breakage were examined by forcing intact, rival males to inseminate the same or opposite reproductive tracts (female have paired, independent tracts in this taxon) and assessing paternity as a function of sclerite location. As predicted, apical sclerites were typically deposited at the entrance to the female's sperm storage organ, where they could physically block insemination by rivals. First male precedence was common when males inseminated the same tract and deposited sclerites at the entrance to the spermatheca, but not when sclerites were found elsewhere in the tract, or when rivals inseminated opposite tracts (where physically blocking rivals was impossible). Our data show that, in redbacks, copulatory organ breakage is not a side‐effect of sexual conflict, is unlikely to be a cue for cryptic female choice, but allows males to avoid sperm competition. Moreover, copulatory organ damage can have minimal reproductive cost for males, so assumptions of sterility after organ breakage are unjustified without supporting data.  相似文献
