We report significant and reproducible growth acceleration of human progenitor cells when exposed to rotational flow when compared with stationary conditions. Nonenriched CD34+ umbilical cord derived human hematopoietic progenitor cells were cultured in Petri dishes located at different radial distances with respect to the central axis of a rotating platform. Growth dynamics under 3 or 5 rpm agitation was compared against that observed under typical stationary conditions. Cells cultured at 3 or 5 rpm exhibited (a) the absence of a latency phase, (b) an increase in final cell concentrations by 54–58.5%, and (c) reduced doubling time in their exponential phase by 12–16% in comparison with stationary culture. Cells grown under rotational agitation were confirmed to remain CD34+ by PCR. These results document a significant positive effect of exposure to laminar flow fields on the growth of human hematopoietic progenitor cells. © 2010 American Institute of Chemical Engineers Biotechnol. One of the most striking components of the modern assemblage of South American mammals is the semiaquatic capybara (Caviidae, Hydrochoerinae), the biggest rodent in the world. The large hydrochoerines are recorded from the middle Miocene to the present, mainly in high latitudes of South America. Although less known, they are also recorded in low latitudes of South America, and in Central and North America. We report the first record of capybaras from the late Pliocene of Colombia, found in deposits of the Ware Formation, Guajira Peninsula in northeastern Colombia. We analyze the phylogenetic position within Caviidae, the possible environmental changes in the Guajira Peninsula, and the implications of this finding for the understanding of the Great American Biotic Interchange. The morphological and phylogenetic analyses indicate that the hydrochoerine of the Guajira Peninsula is a new species, ?Hydrochoeropsis wayuu, and this genus is most closely related to Phugatherium. According to the latest phylogenetic results, this clade is the sister group of the lineage of the recent capybaras (Neochoerus and Hydrochoerus). ?Hydrochoeropsis wayuu is the northernmost South American Pliocene hydrochoerine record and the nearest to the Panamanian bridge. The presence of this hydrochoerine, together with the fluvio-deltaic environment of the Ware Formation, suggests that during the late Pliocene, the environment that dominated the Guajira Peninsula was more humid and with permanent water bodies, in contrast with its modern desert habitats. 相似文献

Warming and elevated atmospheric CO2 (eCO2) can elicit contrasting responses on different SOM pools, thus to understand the effects of combined factors it is necessary to evaluate individual pools. Over two years, we assessed responses to eCO2 and warming of SOM pools, their susceptibility to decomposition, and whether these responses were mediated by plant inputs in a semi-arid grassland at the PHACE (Prairie Heating and CO2 Enrichment) experiment. We used long-term soil incubations and assessed relationships between plant inputs and the responses of the labile and resistant pools. We found strong and contrasting effects of eCO2 and warming on the labile C pool. In 2008 labile C was increased by eCO2 and was positively related to plant biomass. In contrast, in 2007 eCO2 and warming had interactive effects on the labile C, and the pool size was not related to plant biomass. Effects of warming and eCO2 in this year were consistent withtreatment effects on soil moisture and temperature and their effects on labile C decomposition. The decomposition rate of the resistant C was positively related to indicators of plant C inputs. Our approach demonstrated that SOM pools in this grassland can have early and contrasting responses to climate change factors. The labile C pool in the mixed-grass prairie was highly responsive to eCO2 and warming but the factors behind such responses were highly dynamic across years. Results suggest that in this grassland the resistant C pool could be negatively affected by increases in plant-production driven available soil C. 相似文献

The levels of expression of Gsα(short and long), Gα(olf) and Gβ(common) subunits, and calcium-sensitive adenylyl cyclases isoforms (AC1, 5/6, and 8) in human brain cortical and caudate membranes were quantified by western blot analysis in order to establish their contribution to the patterns of AC functioning. Both areas expressed Gsα(long) (52 kDa) with values ranging from about 1400 ng/mg of membrane protein in cerebral cortex to close to 600 ng/mg of membrane protein in caudate nucleus. In contrast, Gsα(short) and Gsα(olf) were expressed separately, Gsα(short) in cortical membranes with values around 500 ng/mg of membrane protein and Gα(olf) in caudate membranes with values around 1300 ng/mg of membrane protein. Quantitative measurements of Gβ, revealed a similar expression level in cortical and caudate membranes (5444±732 versus 5511±394 ng/mg protein; p=0.966). The B(max) values of GTPγS-dependent [(3)H]-forskolin binding show the following descending order: rat striatal membranes>rat cortical membranes=human caudate membranes>human cortical membranes. Therefore, as measured immunochemically and by [(3)H]-forskolin binding, there seems to be a vast excess of Gsα subunits over catalytic units of AC. The highest levels of AC5/6 expression were detected in caudate membranes. AC8 was little expressed, and there were no significant differences in the relative values between both human brain regions. Finally, the levels of the AC1 isoform were significantly lower in caudate than in cortical membranes. It is concluded that these stoichiometric data contribute nonetheless to explain the significant differences observed in signalling capacities through the AC system in both human brain regions. 相似文献

Individual differences in cognitive performance are partly dependent, on genetic polymporhisms. One of the single‐nucleotide polymorphisms (SNP) of the CNR1 gene, which codes for cannabinoid receptor 1 (CB1R), is the rs2180619, located in a regulatory region of this gene (6q14–q15). The alleles of the rs2180619 are A > G; the G allele has been associated with addiction and high levels of anxiety (when the G allele interacts with the SS genotype of the 5‐HTTLPR gene). However, GG genotype is observed also in healthy subjects. Considering G allele as risk for ‘psychopathological conditions’, it is possible that GG healthy subjects do not be addicted or anxious, but would have reduced performance, compared to AA subjects, in attentional control and working memory processing. One hundred and sixty‐four healthy young Mexican‐Mestizo subjects (100 women and 64, men; mean age: 22.86 years, SD=2.72) participated in this study, solving a task where attentional control and working memory were required. GG subjects, compared to AA subjects showed: (1) a general lower performance in the task (P = 0.02); (2) lower performance only when a high load of information was held in working memory (P = 0.02); and (3) a higher vulnerability to distractors (P = 0.03). Our results suggest that, although the performance of GG subjects was at normal levels, a lower efficiency of the endocannabinoid system, probably due to a lowered expression of CB1R, produced a reduction in the performance of these subjects when attentional control and working memory processing is challenged . 相似文献

There is increasing evidence that geographic and climatic clines drive the patterns of plant defence allocation and defensive strategies. We quantified early growth rate and both constitutive and inducible chemical defences of 18 Pinaceae species in a common greenhouse environment and assessed their defensive allocation with respect to each species' range across climatic gradients spanning 31o latitude and 2300 m elevation. Constitutive defences traded‐off with induced defences, and these defensive strategies were associated with growth rate such that slow‐growing species invested more in constitutive defence, whereas fast‐growing species invested more in inducible defence. The position of each pine species along this trade‐off axis was in turn associated with geography; moving poleward and to higher elevations, growth rate and inducible defences decreased, while constitutive defence increased. These geographic patterns in plant defence were most strongly associated with variation in temperature. Climatic and geographical clines thus act as drivers of defence profiles by mediating the constraints imposed by trade‐offs, and this dynamic underlays global patterns of defence allocation. 相似文献
