RNAs, more than ever before, are increasingly viewed as biomolecules of the future, in the versatility of their functions and intricate three-dimensional folding. To effectively study them by nuclear magnetic resonance (NMR) spectroscopy, structural biologists need to tackle two critical challenges of spectral overcrowding and fast signal decay for large RNAs. Stable-isotope nucleotide labeling is one attractive solution to the overlap problem. Hence, developing effective methods for nucleotide labeling is highly desirable. In this work, we have developed a facile and streamlined source of recombinant enzymes from the pentose phosphate pathway for making such labeled nucleotides. The Escherichia coli (E. coli) genes encoding ribokinase (RK), adenine phosphoribosyltransferase (APRT), xanthine/guanine phosphoribosyltransferase (XGPRT), and uracil phosphoribosyltransferase (UPRT) were sub-cloned into pET15b vectors. All four constructs together with cytidine triphosphate synthetase (CTPS) and human phosphoribosyl pyrophosphate synthetase isoform 1 (PRPPS) were transformed into the E. coli BL21(AI) strain for protein over-expression. The enzyme preparations were purified to >90% homogeneity by a one-step Ni-NTA affinity chromatography, without the need of a further size-exclusion chromatography step. We obtained yields of 1530, 22, 482, 3120, 2120 and 2280 units of activity per liter of culture for RK, PRPPS, APRT, XGPRT, UPRT and CTPS, respectively; the specific activities were found to be 70, 22, 21, 128, 144 and 113 U/mg, respectively. These specific activities of these enzyme constructs are comparable to or higher than those previously reported. In addition, both the growth conditions and purification protocols have been streamlined so that all the recombinant proteins can be expressed, purified and characterized in at most 2 days. The availability and reliability of these constructs should make production of fully and site-specific labeled nucleotides for making labeled RNA accessible and straightforward, to facilitate high-resolution NMR spectroscopic and other biophysical studies.  相似文献

BACKGROUND AIMS. Lymphedema is a common complication with breast cancer treatment that does not have a definite cure. Our objective was to determine the efficacy of autologous stem cells (ASC) in the treatment of lymphedema secondary to mastectomy and axillary lymphadenectomy in comparison with traditional decongestive treatment with compression sleeves. METHODS. A prospective study including 20 women with lymphedema secondary to breast cancer surgery with axillary lymphadenectomy was conducted. Women were assigned at random to one of two groups. One group of 10 women was injected with ASC in the affected arm, whereas the other 10 women comprised the control group and received traditional compression sleeve therapy (CST). The follow-up for both groups was 12 weeks. Pain, sensitivity and mobility were assessed before and after therapy. RESULTS. There was improvement in the volume of lymphedema in both groups, with no significant difference. In the ASC group there was an overall volume reduction during the follow-up, whereas in the CST group lymphedema recurred after the compression sleeve was removed. CONCLUSIONS. Our findings suggest that ASC injection for patients with lymphedema can be an effective treatment. It reduces arm volume and associated co-morbidities of pain and decreased sensitivity. Traditional CST was also effective for lymphedema reduction, but it was dependent on continuous use of the treatment.  相似文献

Human milk is a rich source of bioactive proteins that support the early growth and development of the newborn. Although the major components of the protein fraction in human milk have been studied, the expression and relative abundance of minor components have received limited attention. We examined the expression of low-abundance proteins in the whey fraction of human milk and their dynamic changes over a twelve-month lactation period. The low-abundance proteins were enriched by ProteoMiner beads, and protein identification was performed by liquid chromatography tandem mass spectrometry. One hundred and fifteen proteins were identified, thirty-eight of which have not been previously reported in human colostrum or milk. We also for the first time described differences in protein patterns among the low-abundance proteins during lactation. These results enhance our knowledge about the complexity of the human milk proteome, which constitutes part of the advantages to the breast-fed infant.  相似文献

An often-suggested mechanism of virus induced neuronal damage is oxidative stress. Astrocytes have an important role in controlling oxidative stress of the Central Nervous System (CNS). Astrocytes help maintain a homeostatic environment for neurons as well as protecting neurons from Reactive Oxygen Species (ROS). CM-H2DCFDA is a cell-permeable indicator for the presence of ROS. CM-H(2)DCFDA enters the cell as a non-fluorescent compound, and becomes fluorescent after cellular esterases remove the acetate groups, and the compound is oxidized. The number of cells, measured by flow cytometry, that are found to be green fluorescing is an indication of the number of cells that are in an oxidative state. CM-H(2)DCFDA is susceptible to oxidation by a large number of different ROS. This lack of specificity, regarding which ROS can oxidize CM-H(2)DCFDA, makes this compound a valuable regent for use in the early stages of a pathogenesis investigation, as this assay can be used to screen for an oxidative cellular environment regardless of which oxygen radical or combination of ROS are responsible for the cellular conditions. Once it has been established that ROS are present by oxidation of CM-H(2)DCFDA, then additional experiments can be performed to determine which ROS or combination of ROSs are involved in the particular pathogenesis process. The results of this study demonstrate that with the addition of hydrogen peroxide an increase in CM-H(2)DCFDA fluorescence was detected relative to the saline controls, indicating that this assay is a valuable test for detecting an oxidative environment within G355-5 cells, a feline astrocyte cell line.  相似文献

Phytoremediation is an environmental biotechnology that seeks to remediate pollution caused by bioaccumulative toxins like copper (Cu). Symbiotic mycorrhizal associations can increase the uptake and delivery of low mobility nutrients and micronutrients to the host plant because they solubilize these substances and increase their catchment area. To analyze the effect of mycorrhizae on the phytoaccumulation of Cu, we studied their ability to solubilize Cu(II) and enhance its absorption by the plant Tagetes erecta L. colonized with the arbuscular mycorrhizal fungus Glomus intraradices. Plants were grown for nine weeks in a growth chamber under controlled conditions of temperature, relative humidity and photoperiod. Cu was added in the insoluble form of CuO to simulate the insoluble Cu-O affixed species in soil. The biotic and abiotic parameters of colonization, foliar area, biomass and the pH of leachates were determined as functions of the Cu concentration that was measured in the roots, shoots and leachates by AAS. The results of Cu absorption showed that the colonized plants accumulated more Cu in the roots as well as the whole plant and that both the colonized and non-colonized plants displayed the typical behavior of Cu excluders. Mycorrhizal colonization of the roots resulted in a proliferation of vesicles and this was observed to scale with root tissue Cu concentrations. Also, the G. intraradices-T. erecta system displayed a higher resistance to the toxicity induced by Cu while nonetheless improving the indices of phytoaccumulative yields. These results suggest that G. intraradices possibly accumulates Cu in its vesicles thereby enhancing the Cu tolerance of T. erecta even while increasing root Cu accumulation. The parameters of bioconcentration factor and translocation factor measured in this work suggest that the system T. erecta-G. intraradices can potentially phytostabilize Cu in contaminated soils.  相似文献

Artisan fresh Mexican-style cheeses are commonly made from raw milk that provides not only rich flavors, but also a diversity of associated lactic acid bacteria (LAB) strains. Enterococcus faecium UQ31 was isolated from panela cheese and produced bacteriocin-like inhibitory substances (BLIS) with a strong anti-Listeria activity. A modified pH–mediated adsorption-desorption purification process resulted in (after SDS-PAGE) two bands showing antimicrobial activities, where most of the activity corresponded to the band with an estimated molecular weight of 7.5 kDa. The BLIS produced by E. faecium UQ31 were heat resistant, stable at ambient storage conditions, and active in the pH range 5–9. The BLIS antimicrobial activities were detected during logarithmic growth phase and remained constant until the end of incubation time (19 h). These BLIS showed a wide anti-Listeria monocytogenes spectra. The E. faecium UQ31 strain or their BLIS represent a promising potential as antimicrobial food preservatives.  相似文献

Comparative phylogeography has revealed remarkable patterns of concordance in the maternal phylogenies of many species. The phylogeography and historical demography of the mitochondrial control region I for 607 Atlantic bluefin tuna (Thunnus thynnus) and 275 swordfish (Xiphias gladius) were analyzed to clarify the complex phylogenetic signals in the North Atlantic-Mediterranean region where they are sympatric. Atlantic bluefin tuna mtDNA is polyphyletic, and includes rare sequences sister to Pacific bluefin tuna (Thunnus orientalis) and introgressed albacore (Thunnus alalunga) sequences. There is no geographic partitioning between Atlantic and Mediterranean samples of Atlantic bluefin tuna (Phi(ST)=0.002). In contrast, Atlantic and Mediterranean swordfish are differentiated (Phi(ST)=0.091) due to the combined effects of vicariance, secondary contact, and dissimilar regional demographic histories. Mediterranean swordfish has substantially less variation, and a more recent history (tau=2.42) than that of Atlantic swordfish (tau=7.02). In spite of the discordant phylogenetic and phylogeographic signals, the demographic history of Atlantic swordfish and Atlantic bluefin tuna (tau=7.51) suggests concordance in the timeline of population expansion. Possible scenarios of cladogenesis, expansion, and contraction, influenced by glacial cycles during the Pleistocene, are formulated.  相似文献

Metamorphosis is broadly defined as a more or less radical morphologicalchange between 2 multicellular life stages within an organism'slife phase, often marking the transition from pre-reproductiveto reproductive stages. It involves structural reorganizationand major physiological changes, generally under the controlof endogenous and exogenous factors and often resulting in changesin habitat use. This concept has been applied to the crustose-to-erectthallus transition of some red algae and the present study evaluatesthe validity of such hypotheses. Available literature suggeststhat the crustose-to-erect thallus transition involves significantmorphological and habitat changes, separating pre-reproductiveand reproductive stages. The onset of the morphological changes(for example axis differentiation) appears regulated by endogenoussignals (growth factors) and growth is modified by environmentalfactors. The algae do not exhibit structural reorganization,however, probably due to their simple morphological structureand the lack of several cell and tissue mechanisms involvingcell motility. The presence of cell walls in the algae impairscell motility and maintains the cell in a fixed position withinthe plant. These are important differences restricting the extensionof the definition of metamorphosis to the crust-to-erect thallustransition. The above restrictions also seem to apply to othermacroalgae, fungi, and terrestrial plants.  相似文献

Danaea tuomistoana A. Rojas (Marattiaceae) is described and illustrated as a new species endemic to Costa Rica; it differs from D. crispa Endres et Richb. by a truncate blade, and by the pinnae, which are 2/5-1/2 incised, cuneated at the base and with a flat margin. The range of D. erecta H. Tuomisto & R.C. Moran in Costa Rica is expanded with a new locality in Puntarenas (8 degrees 57'15" N-82 degrees 50'1" W, 1,500-1,580 masl).  相似文献
