Vibrio cholerae is a human pathogen and natural inhabitant of aquatic environments. Serogroups O1/O139 have been associated with epidemic cholera, while non-O1/non-O139 serogroups usually cause human disease other than classical cholera. V. cholerae non-O1/non-O139 from the Neusiedler See, a large Central European lake, have caused ear and wound infections, including one case of fatal septicaemia. Recent investigations demonstrated rapid planktonic growth of V. cholerae non-O1/non-O139 and correlation with zooplankton biomass. The aim of this study was to elucidate the interaction of autochthonous V. cholerae with two dominant crustacean zooplankton species in the lake and investigate the influence of the natural bacterial community on this interaction. An existing data set was evaluated for statistical relationships between zooplankton species and V. cholerae and co-culture experiments were performed in the laboratory. A new fluorescence in situ hybridisation protocol was applied for quantification of V. cholerae non-O1/non-O139 cells, which significantly reduced analysis time. The experiments clearly demonstrated a significant relationship of autochthonous V. cholerae non-O1/non-O139 with cladocerans by promoting growth of V. cholerae non-O1/non-O139 in the water and on the surfaces of the cladocerans. In contrast, copepods had a negative effect on the growth of V. cholerae non-O1/non-O139 via competing bacteria from their surfaces. Thus, beside other known factors, biofilm formation by V. cholerae on crustacean zooplankton appears to be zooplankton taxon specific and may be controlled by the natural bacterial community.  相似文献

Inner Mongolia steppe grasslands are widely used for livestock farming and the regrowth ability of grassland species is therefore strongly influenced not only by water and nutrient availability but also quite heavily by grazing. However, little is known on how grazing, water and nitrogen interactively affect the dominant C3 species (Leymus chinensis, Stipa grandis, Agropyron cristatum) and the C4 species (Cleistogenes squarrosa). Therefore in the 2007 and 2008 growing seasons, a field experiment was carried out to test the hypothesis that under different grazing intensities the dominant species show different short-term regrowth response to simultaneous variation in the availability of water and nitrogen. Single factor and interaction effects of the addition of water (rainfed vs. simulated wet-year) and nitrogen (0 or 25 kg N ha?1) were analysed along a gradient of four grazing intensities (ungrazed, lightly, moderately and heavily grazed) after one month of grazing exclusion. Water and nitrogen addition affected short-term regrowth of all species in a similar way whereas species responded differently to grazing. Simulated wet-year water availability consistently resulted in higher standing biomass, relative growth rate and cellulase digestible organic matter yield. Supplementary nitrogen promoted standing biomass and crude protein concentration. The nutritive value of all species’ standing biomass showed a similar increase with more intensive grazing. However, heavy grazing led to a clear shift in the relative biomass of the species, i.e. mainly a promotion of the C4 grass, C. squarrosa. In contrast to our hypothesis, there were no differences among species in their response to water or nitrogen addition, whereas, heavy grazing induced the expected species-specific response. Our results suggest that heavy grazing rather than nitrogen or water determine short-term shifts in species composition of the investigated steppe ecosystem. Furthermore, differences in the species-specific growth response to grazing may increase the proportion of the C4 grass C. squarrosa in steppe communities, whereas higher availability of nitrogen and water may lead to higher forage biomass and nutritive value of all investigated species but in short-term cannot compensate for the grazing induced changes in species composition.  相似文献

The major aims of this study were, firstly, to analyse the grazing-induced steppe degradation process and, secondly, to identify an efficient and sustainable grazing management system for the widely degraded Inner Mongolian typical steppe ecosystem. From 2005?C2008 a grazing experiment was conducted to compare two grazing management systems, the Mixed System (MS) and the Traditional System (TS), along a gradient of seven grazing intensities, i.e. ungrazed (GI0), very-light (GI1), light (GI2), light-moderate (GI3), moderate (GI4), heavy (GI5), and very-heavy (GI6). Each grazing intensity treatment was considered a production unit comprising two adjacent plots, one for hay-making (single-cut system) and one for grazing. Hay-making and grazing alternated annually in the MS, while in the TS the same plots were used either for hay-making or for grazing. Effects of management system, grazing intensity, and year on end-of-season standing biomass (ESSB), aboveground net primary production (ANPP), relative difference in ANPP between 2005 and 2008 (ANPPDiff), relative growth rate (RGR), and sward characteristics (litter accumulation, soil coverage) were analysed. Litter accumulation of production units was affected by grazing intensity (P?<?0.001) and decreased from GI0 to GI6 by 83%. Correspondingly, soil coverage decreased (P?<?0.001) from GI0 to GI6 by 43%, indicating an increased vulnerability to soil erosion. We found varying compensatory growth responses to grazing intensity among years, probably because of temporal variability in precipitation. The ability of plants to partially compensate for grazing damage was enhanced in years of greater seasonal precipitation. The ANPP of production units was negatively affected by grazing intensity and decreased from GI0 to GI6 by 37, 30, and 55% in 2006 (P?<?0.01), 2007 (P?<?0.05), and 2008 (P?<?0.001), respectively. The effect of management system × grazing intensity interaction on ANPP (P?<?0.05) and ANPPDiff (P?<?0.05) suggested greater grazing resilience of the MS as compared to the TS at GI3 to GI6.  相似文献

To understand carbon partitioning in roots and nodules of Datisca glomerata, activities of sucrose-degrading enzymes and sugar transporter expression patterns were analyzed in both organs, and plasmodesmal connections between nodule cortical cells were examined by transmission electron microscopy. The results indicate that in nodules, the contribution of symplastic transport processes is increased in comparison to roots, specifically in infected cells which develop many secondary plasmodesmata. Invertase activities are dramatically reduced in nodules as compared to roots, indicating that here the main enzyme responsible for the cleavage of sucrose is sucrose synthase. A high-affinity, low-specificity monosaccharide transporter whose expression is induced in infected cells prior to the onset of bacterial nitrogen fixation, and which has an unusually low pH optimum and may be involved in turgor control or hexose retrieval during infection thread growth.  相似文献

Electric field strength values calculated by wave propagation modeling were applied as an exposure metric in a case–control study conducted in Germany to investigate a possible association between radio frequency electromagnetic fields (RF‐EMF) emitted from television and radio broadcast transmitters and the risk of childhood leukemia. To validate this approach it was examined at 850 measurement sites whether calculated RF‐EMF are an improvement to an exposure proxy based on distance from the place of residence to a transmitter. Further, the agreement between measured and calculated RF‐EMF was explored. For dichotomization at the 90% quantiles of the exposure distributions it was found that distance agreed less with measured RF‐EMF (Kappa coefficient: 0.55) than did calculated RF‐EMF (Kappa coefficient: 0.74). Distance was a good exposure proxy for a single transmitter only which uses the frequency bands of amplitude modulated radio, whereas it appeared to be of limited informative value in studies involving several transmitters, particularly if these are operating in different frequency bands. The analysis of the agreement between calculated RF‐EMF and measured RF‐EMF showed a sensitivity of 76.6% and a specificity of 97.4%, leading to an exposure misclassification that still allows one to detect a true odds ratio as low as 1.4 with a statistical power of >80% at a two‐sided significance level of 5% in a study with 2,000 cases and 6,000 controls. Thus, calculated RF‐EMF is confirmed to be an appropriate exposure metric in large‐scale epidemiological studies on broadcast transmitters. Monitoring of lipophilic marine toxins was carried out in three shellfish production sites on the West Coast of Ireland. The toxins were monitored using passive samplers (solid phase adsorption toxin tracking; SPATT) and toxin-free mussels that were replaced weekly in the selected sampling stations. The toxin profiles and concentrations obtained in the SPATT and in the transplanted mussels were compared with those observed in indigenous (native) mussels from each production site as well as with the phytoplankton that was detected in the water. Numerous lipophilic toxins were detected in the SPATT discs by ultra-performance liquid-chromatography–mass spectrometry/mass spectrometry (UPLC–MS/MS) and included okadaic acid (OA), dinophysistoxin-2 (DTX2), pectenotoxin-2 (PTX2), AZA1, AZA2, yessotoxin (YTX) and SPX-13-DesMe-C. The accumulation rate of toxins in the indigenous mussels and in the SPATT discs correlated well. Toxins were detected in all SPATT discs from all locations, even in the absence of toxin-producing phytoplankton, as observed previously by other research groups. It is quite clear from our data that the presence of okadaic acid in the water (even at high concentrations) did not induce toxicity in the transplanted mussels in the absence of phytoplankton. A severe toxic event of azaspiracids (AZAs) occurred in one of the sampling stations. The SPATT discs accumulated predominantly AZA1 and -2 suggesting that both toxins are biosynthesized by the AZA-producing organism. As opposed to the DSP event, the AZA event resulted in the contamination of the transplanted mussels for several consecutive weeks. This is the first study that reports the presence of YTX and SPX-13-DesMe-C in Irish waters. In our study conditions, the SPATT did not enable the forecasting of shellfish contamination as the increase in toxin concentration occurred at the same time in the shellfish and in the SPATT.  相似文献

Dilated cardiomyopathy (DCM) is a common disease in humans and dogs. Large-breed dogs and especially Irish wolfhounds belong to the frequently affected breeds. Male Irish wolfhounds show a significantly higher prevalence of DCM than females. Therefore, we evaluated X chromosome markers for linkage with DCM as well as a human candidate gene on the X chromosome. A set of X chromosomal microsatellites was genotyped in Irish wolfhound families segregating for DCM. In addition, exon and intron sequences of the tafazzin (TAZ) gene were assayed for polymorphisms segregating in these families. Statistical analysis of the microsatellite markers did not reveal linkage to DCM. Furthermore, all Irish wolfhounds included in this study were monomorphic for TAZ, and only 8 sequence differences to the Dog Genome Assembly 2.1 could be found. The results indicate that due to the lack of mutations, TAZ is unlikely to cause DCM in Irish wolfhounds.  相似文献
