Degradation of acrylic copolymers by white-rot fungi   总被引:4,自引:0,他引:4  
Various water-soluble homopolymers and copolymers of acrylamide (AAm) and acrylic acid (AA) which contained phenolic sites, such as guaiacol, lignin sulfonate (LS) and 3,4-dihydroxybenzoic acid (3,4-DHBA), were tested with regard to their degradability by white-rot fungi. Compared with Phanerochaete chrysosporium, Pleurotus ostreatus caused a significantly higher decrease in the average molecular weight (w) of most of the copolymers and the homopolymer under the applied culture conditions. However, the w of poly(guaiacol/AAm) increased significantly during incubation with Pl ostreatus. P. chrysosporium was able to reduce only the w of the poly(LS/AA) to a significant degree and not that of the other polymers. The mineralization rate of AAm and AA copolymers and terpolymers of AAm, AA and phenolics (LS, 3,4-DHBA, guiacol), which were tested with P. ostreatus and Trametes versicolor, turned out to be low (0.8–3.2%). While the rates of mineralization were similar among all polymers, the decrease in radioactivity from the culture media was higher with the terpolymers bearing phenolic sites. UV spectra of the culture media revealed that the phenolic sites in the terpolymers were significantly degraded by both fungi. Obviously, the degradation of phenolics within the polymer chain caused a higher decrease in w but did not significantly increase the mineralization rate.  相似文献

Until now, the predominant use cases of industrial robots have been routine handling tasks in the automotive industry. In biotechnology and tissue engineering, in contrast, only very few tasks have been automated with robots. New developments in robot platform and robot sensor technology, however, make it possible to automate plants that largely depend on human interaction with the production process, e.g., for material and cell culture fluid handling, transportation, operation of equipment, and maintenance. In this paper we present a robot system that lends itself to automating routine tasks in biotechnology but also has the potential to automate other production facilities that are similar in process structure. After motivating the design goals, we describe the system and its operation, illustrate sample runs, and give an assessment of the advantages. We conclude this paper by giving an outlook on possible further developments.  相似文献

Yeast reporter system for rapid determination of estrogenic activity   总被引:5,自引:0,他引:5  
An in vitro test system for the determination of estrogens, xeno- and phytoestrogens, based on the activation of human estrogen receptor-alpha, has been examined for ability in monitoring environmental estrogens. The system consists of an expression plasmid for the human estrogen receptor-alpha and a reporter plasmid containing the lacZ gene under the control of the vitellogenin hormone response element. These plasmids have been transformed into S. cerevisae. Cultivation of yeast in the presence of estrogenic substances leads to activation of the estrogen receptor and induces the expression of the reporter lacZ. beta-Galactosidase activity of the translated gene lacZ is a measure of the estrogenic activity of a compound. First, the selectivity of the system was compared to data available in the literature. Then the sensitivity of the system was checked. The detection limit is 0.1 ng 17-beta estradiol or an equivalent activity per liter, if a sample can be concentrated 1000-fold. The system has been further characterized by selected compounds with known and unknown estrogenic activity.  相似文献

cGMP- and cAMP-dependent protein kinases (cGK I, cGK II, and cAK) are important mediators of many signaling pathways that increase cyclic nucleotide concentrations and ultimately phosphorylation of substrates vital to cellular functions. Here we demonstrate a novel mRNA splice isoform of cGK II arising from alternative 5' splicing within exon 11. The novel splice variant encodes a protein (cGK II Delta(441-469)) lacking 29 amino acids of the cGK II Mg-ATP-binding/catalytic domain, including the conserved glycine-rich loop consensus motif Gly-x-Gly-x-x-Gly-x-Val which interacts with ATP in the protein kinase family of enzymes. cGK II Delta(441-469) has no intrinsic enzymatic activity itself, however, it antagonizes cGK II and cGK I, but not cAK. Thus, the activation and cellular functions of cGK II may be determined not only by intracellular cGMP levels but also by alternative splicing which may regulate the balance of expression of cGK II versus its own inhibitor, cGK II Delta(441-469).  相似文献

Although chromatographic separation of whey proteins has been considered too expensive, whey may serve as an excellent model mixture to investigate and validate the use of simulation tools in the development and optimization of chromatographic separations and the outcome could easily be utilized since the model system has an intrinsic value. Besides, milk from transgenic animals could be an attractive source of pharmaceuticals which must be separated from the other proteins in the milk. Several whey proteins are of interest especially, alpha-lactalbumin, beta-lactoglobulins, immunoglobulins, lactoperoxidase, and lactoferrin. The scope of the project is to develop a consistent set of chromatographic data for whey proteins including isotherms, transport properties and scale-up studies and to develop the appropriate models for the anion exchangers Q-Sepharose XL, Source 30Q, Ceramic Q-HyperD F, and Merck Fractogel EMD TMAE 650 (S). In this work we have determined and correlated gradient and isocratic retention volumes in the linear range of the isotherm for alpha-lactalbumin, beta-lactoglobulin A and B, and bovine serum albumin at a pH from 6 to 9 at various NaCl concentrations.  相似文献

The purpose of the present study was to investigate the application of various sample preparation methods (cell washing before lysis, purification of DNA using phenol extraction method, immunomagnetic separation-IMS) for the final PCR identification of Salmonellacells. The presence of PCR inhibitors in processed food products (milk powder and dried eggs) can be the cause of false-negative results in PCR without IMS of target cells. It was also demonstrated that IMS-PCR was successfully used for identification and quick confirmation of untypical Salmonella strains isolated from human stool samples and rabbit meat. However, IMS cannot eliminate intracellular PCR inhibitors present in immunoseparated Salmonella cells. These inhibitors must be taken into consideration in evaluation of PCR procedure.  相似文献

Microbial processes were investigated in the soil of a declining, more eutrophic (Romberk West) and a healthy looking, less eutrophic (Romberk East) freshwater reed stand. Soil was sampled monthly from June to September 1997. Glucose induced carbon dioxide (CO2) production in oxic and anoxic conditions, methane (CH4) production, nitrification and denitrification activities were measured in laboratory conditions in suspensions prepared from homogenised soil samples. Within a stand the proportion of anaerobic (as opposed to aerobic) microbial activity was greatest in June. Potential methanogenesis was highest in June and decreased later in both stands. Methane production was approximately the same in June at both stands but it was higher at Romberk East than at Romberk West stand in later months. Denitrifying activity was higher in August than July at both stands. Nitrifying activity was undetectable at both stands over the entire study period. Generally Romberk West was more anaerobic than Romberk East, with lower redox potential, higher amounts of oxygen-consuming organic matter and a lower ratio of CO2 production in oxic conditions to CO2 production in anoxic conditions. Microbial activity was apparently restricted at Romberk West stand in comparison to Romberk East. The shift from aerobic to anaerobic microbial metabolism and a coinciding restriction of metabolic activities at Romberk West are thought to be indicative of a strengthened oxygen stress in the soil, associated with accumulation of metabolites toxic to both the microorganisms and the reed. Possible links between eutrophication, microbial characteristics and reed performance are discussed.  相似文献

Wheat plants were grown from sowing to day 18 in 26-dm3 chambers at three different CO2 concentrations: 150 (-CO2), 350 (C, control), 800 (+CO2) mol mol-1. Afterwards, plants of the three variants were grown at the same natural CO2 concentration. Plant characteristics were measured just before the transfer (0 days after CO2 treatment, DAT), and at 5 – 8 DAT on the 1st leaf, and at 12 – 22 DAT on the 4th leaf. Decreased or increased CO2 concentrations caused acclimations which persisted after transplantation to natural CO2 concentration. At 5 – 8 DAT, stomatal density, stomatal conductance (gs), CO2 saturated net photosynthetic rate (PNsat0), radiation saturated net photosynthetic rate (PNsat1), and carboxylation efficiency () were higher in -CO2 plants and lower in +CO2 plants than in C plants. As compared with C plants, the photochemical efficiency () was lower in -CO2 and higher in -CO2 plants, however, chlorophyll (Chl) a, Chl b, Chl a–b and carotenoid contents were lower in both -CO2 and +CO2 plants. On the 4th leaf, which emerged on plant after finishing CO2 treatments, at 12 – 22 DAT, no differences in stomatal density and g, between treatments were observed. In -CO2 plants, pigment content and PNsat0 were higher, was lower, and PNsat1 and were not different from C plants. In contrast, in +CO2 plants, pigment content, PNsat1 and were lower, and PNsat0 and were unchanged. Leaf area, dry mass, and tiller development increased in +CO2 plants and decreased in -CO2 plants. In the interval between 8 and 22 DAT, lower net assimilation rate in +CO2 than in -CO2 plants was observed.  相似文献

Accumulation of the pathogenesis-related (PR) proteins localised in intercellular spaces of barley primary leaves, chlorophyll content, structure of chloroplasts, and photosynthesis were examined during natural and in vitro induced leaf senescence (cultivation of whole plants in the dark or detached leaves under nutrient deficiency). Some of PR proteins accumulated during natural senescence, but their accumulation pattern was different from those of pathogen-induced as well as during in vitro-induced senescence, which indicate different molecular bases of these processes. Photosynthetic rate and chlorophyll content indicate that natural senescence of barley primary leaves began from 15th day after sowing. In 35-d-old first leaves, the chloroplasts showed typical characteristics of senescence as significant decrease of size, greater grana, and prominent plastoglobuli. The chloroplasts contained more grana under in vitro induced senescence and they had reduced length in the dark. Correspondingly, accumulation of PR proteins was detectable on about the 15th day but the content of some PR proteins increased in later stages of senescence. The effect of simulated plant stress and parasitism by Aphelinus abdominalis (Dalman) on the mobility of four species of aphids was investigated. The aphids were placed on water stressed potted plants or on excised leaf segments on dry or moist filter-paper in Petri dishes. Winter wheat (Triticum aestivum) was infested with Metopolophium dirhodum (Walker), Sitobion avenae (F.) and Rhopalosiphum padi (L.) (segments only), aubergine (Solanum melongena) leaf segments with Macrosiphum euphorbiae (Thomas). The aphids that moved off the plants were removed and their development stage determined at 24-h (drought-stressed plants) or 3-h (leaf segments) intervals. On intact plants, aphid larvae were reluctant to move, and only moved after moulting into adults. On cut leaves, young 1st and 2nd instar larvae were more reluctant to move than 3rd and 4th instar larvae and adults. The numbers moving were initially positively correlated with aphid population density, and in M. euphorbiae occurred mainly during the night. Depending on aphid species and parasitoid age, parasitism by A. abdominalis retarded or accelerated movement, but the differences between young and old larvae and adults persisted after parasitism. A. abdominalis lays male eggs in small hosts and female eggs in large hosts. Consequently, its sex ratio was affected by differential movement of the host aphids of the male and female parasitoids. Old larvae and adult aphids readily moved and carried off female parasitoids, while the small aphids were more likely to remain and give rise to male-biased sex ratios.  相似文献
