Due to broad‐scale habitat loss, European Rollers (Coracias garrulus) have been decreasing in numbers rapidly during the 20th century in parts of their European distribution range. In Austria, as of 2017, only a completely isolated relict population of two breeding pairs and a few non‐breeders remained in Styria compared to about 270 pairs in the 1950s. In 2018, no breeders have been recorded. Since 2002, all nestlings and adult birds in Austria have been ringed. Given the small census size, combined with lack of immigration from other populations, genetic depletion seems likely. In the present study, mitochondrial control region sequence and microsatellite data based on blood samples of nestlings from recent years were collected and compared with museum samples from historical times (when Rollers were more common and widespread in Austria) and with birds across the distribution range to arrive at a first preliminary phylogeographic dataset for the species. The mitochondrial DNA showed a decrease in variation over time in Austria, eventually reaching monomorphism, while genetic diversity of 10 microsatellite loci was higher than expected and a change in genetic structuring through time was observed. These results indicate drift effects in this relict European Roller population caused by the fast population breakdown and small population size. Our phylogeographic analysis indicates a division into a European and an Asian group, roughly (but not exactly) in accordance with the two subspecies C. garrulus garrulus (Europe) and Coracias garrulus semenowi (Asia). The lack of substructuring in the European group along with the results from nuclear DNA markers show the Austrian Rollers to be part of a formerly continuous population and opens the way to restocking the present relict population with birds from Eastern Europe (“genetic rescue”).  相似文献

Costimulatory molecules like ICOS are crucial in mediating T cell differentiation and function after allergen contact and thereby strongly affect the immunologic decision between tolerance or allergy development. In this study, we show in two independent approaches that interruption of the ICOS signaling pathway by application of a blocking anti-ICOSL mAb in wild-type (WT) mice and in ICOS(-/-) mice inhibited respiratory tolerance development leading to eosinophilic airway inflammation, mucus hypersecretion, and Th2 cytokine production in response to OVA sensitization. Respiratory Ag application almost doubled the number of CD4(+)Foxp3(+) regulatory T cells (Tregs) in the lung of WT mice with 77% of lung-derived Tregs expressing ICOS. In contrast, in ICOS(-/-) mice the number of CD4(+)CD25(+)Foxp3(+) Tregs did not increase after respiratory Ag application, and ICOS(-/-) Tregs produced significantly lower amounts of IL-10 than those of WT Tregs. Most importantly, in contrast to WT Tregs, ICOS(-/-) Tregs did not convey protection when transferred to "asthmatic" recipients demonstrating a strongly impaired Treg function in the absence of ICOS signaling. Our findings demonstrate a crucial role of ICOS for the generation and suppressive function of Tregs conveying respiratory tolerance and support the importance of ICOS as a target for primary prevention strategies.  相似文献

A crepuscular or nocturnal lifestyle has evolved in bees severaltimes independently, probably to explore rewarding pollen sourceswithout competition and to minimize predation and nest parasites.Despite these obvious advantages, only few bee species are nocturnal.Here we show that the sensitivity of the bee apposition eyeis a major factor limiting the ability to forage in dim light.We present data on eye size, foraging times, and light levelsfor Megalopta genalis (Augochlorini, Halictidae) in Panama,and Lasioglossum (Sphecodogastra) sp. (Halictini, Halictidae)in Utah, USA. M. genalis females forage exclusively during twilight,but as a result of dim light levels in the rain forest, theyare adapted to extremely low intensities. The likely factorlimiting their foraging activity is finding their nest entranceon return from a foraging trip. The lowest light intensity atwhich they can do this, both in the morning and the evening,is 0.0001 cd m–2. Therefore, they leave the nest at dimmerlight levels in the morning than in the evening. Lasioglossum(Sphecodogastra) foraging is limited by light intensity in theevening, but probably by temperature in the morning in the temperateclimate of Utah. We propose that the evolution of nocturnalityin bees was favored by the large variance in the size of females.  相似文献

In Experiment 1, development of zona pellucida-intact (ZPI) morulae was measured every 24 hours for 120 hours after encapsulation in 2% sodium alginate (ALG) or 0.1% poly-L-lysine (PLL). Encapsulation significantly reduced development to hatched blastocysts at 48 and 72 hours. Developmental stages and diameters of ZPI and zona pellucida-free (ZPF) unencapsulated and encapsulated morulae were measured every 24 hours for 72 hours in Exeriment 2. At 72 hours, the percentage of ZPI embryos developing to expanded blastocysts, their diameters and their nuclear counts were not different from each other or from ZPF embryos. In Experiment 3, ZPI morulae encapsulated in ALG or PLL were transferred into recipients. Five of six recipients that received unencapsulated embryos (n=71) delivered 16 live pups. None of the recipients of encapsulated embryos delivered offspring; therefore, a final experiment was performed to examine fetal development on Day 10 of gestation. The percentage of pregnant recipients was similar for all 3 treatments: unencapsulated (71.4%), ALG (87.5%) and PLL (87.5%). However, the presence of viable fetuses was higher for unencapsulated embryos (42.1%) than for ALG (17%) and PLL (14.6%) embryos. These results suggest that encapsulation did not detrimentally affect embryonic size or cellular development in vitro; however, mortality occurred in vivo due to an asynchronous condition between the uterine environment and the embryos.  相似文献

Variation in apolipoprotein E (APOE) has been shown to have the strongest genetic effect on human longevity. The aim of this study was to unravel the evolutionary history of the three major APOE alleles in Europe by analysing ancient samples up to 12,000 years old. We detected significant allele frequency shifts between populations and over time. Our analyses indicated that selection led to large frequency differences between the earliest European populations (i.e., hunter-gatherers vs. first farmers), possibly due to changes in diet/lifestyle. In contrast, the allele distributions in populations from ~4000 BCE onward can mainly be explained by admixture, suggesting that it also played an important role in shaping current APOE variation. In any case, the resulting allele frequencies strongly influence the predisposition for longevity today, likely as a consequence of past adaptations and demographic processes.  相似文献

The interaction of an antimicrobial peptide, MSI-78, with phospholipid bilayers has been investigated using atomic force microscopy, circular dichroism, and nuclear magnetic resonance (NMR). Binding of amphipathic peptide helices with their helical axis parallel to the membrane surface leads to membrane thinning. Atomic force microscopy of supported 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) bilayers in the presence of MSI-78 provides images of the membrane thinning process at a high spatial resolution. This data reveals that the membrane thickness is not reduced uniformly over the entire bilayer area. Instead, peptide binding leads to the formation of distinct domains where the bilayer thickness is reduced by 1.1 +/- 0.2 nm. The data is interpreted using a previously published geometric model for the structure of the peptide-lipid domains. In this model, the peptides reside at the hydrophilic-hydrophobic boundary in the lipid headgroup region, which leads to an increased distance between lipid headgroups. This picture is consistent with concentration-dependent 31P and 2H NMR spectra of MSI-78 in mechanically aligned DMPC bilayers. Furthermore, 2H NMR experiments on DMPC-d54 multilamellar vesicles indicate that the acyl chains of DMPC are highly disordered in the presence of the peptide as is to be expected for the proposed structure of the peptide-lipid assembly.  相似文献

The objective of this study was to compare the conception rate for fixed-timed artificial insemination (FTAI) and observed heat artificial insemination (HAI) prior to the scheduled FTAI in Ovsynch and Heatsynch synchronization protocols. In Experiment 1, lactating dairy cows (n=535) received two set-up injections of 25mg prostaglandin F(2alpha) (PGF(2alpha)) i.m., 14 days apart starting at 36+/-3 days in milk (DIM). Cows were blocked by parity and were randomly allocated to either Ovsynch or Heatsynch groups. All cows received 100 microg of GnRH i.m. 14 days after the second set-up injection of PGF(2alpha), followed by a third injection of 25mg PGF(2alpha) i.m., 7 days later. In the Ovsynch group, HAI cows (n=29) were bred on standing estrus after the third PGF(2alpha) before the scheduled second GnRH, whereas FTAI cows (n=218) that were not observed in estrus, received a second injection of 100 microg of GnRH i.m., 48 h after the third PGF(2alpha) and received TAI 8 h after the second GnRH. In the Heatsynch group, all cows (n=288) received 0.5 mg of estradiol cypionate (ECP) 24 h after third PGF(2alpha) and HAI cows (n=172) were bred on standing estrus and FTAI cows (n=116) that were not observed in estrus, received TAI 72 h after the third PGF(2alpha). In Experiment 2, repeat breeder cows (n=186) were randomly assigned to either Ovsynch or Heatsynch groups. The FTAI and HAI cows were inseminated similar to Experiment 1. All cows were observed for estrus three times daily. The associations with the conception rate were modeled with logistic regression separately for Experiments 1 and 2. Of all the variables included in the model in Experiment 1, type of AI (HAI versus FTAI, P=0.0003) and parity (primiparous versus multiparous, P=0.05) influenced the first service conception rate. Over-all conception rate and first service conception rate for HAI cows were higher compared to FTAI cows (33.8% versus 21.3%, and 35.3% versus 21.0%; P=0.001). In the Heatsynch group, cows that received HAI had significantly higher over-all conception rate and first service conception rate compared to FTAI (35.2% versus 17.3% and 36.0% versus 15.5%; P=0.0001). The conception rates in repeat breeder cows for HAI and FTAI (30.1% versus 22.3%) were not different (P>0.1). In conclusion, it was recommended to include AI at observed estrus and fixed-time AI for cows not observed in estrus in order to improve the conception rate in synchronization protocols.  相似文献

The diurnal hummingbird hawkmoth Macroglossum stellatarum can learn the achromatic (intensity-related) and the chromatic (wavelength-related) aspect of a spectral colour. Free-flying moths learn to discriminate two colours differing in the chromatic aspect of colour fast and with high precision. In contrast, they learn the discrimination of two stimuli differing in the achromatic aspect more slowly and less reliably. When trained to use the chromatic aspect, they disregard the achromatic aspect, and when trained to use the achromatic aspect, they disregard the chromatic aspect, at least to some degree. In a conflicting situation, hummingbird hawkmoths clearly rely on the chromatic aspect of colour. Generally, the moths pay attention to the most reliable cue that allows them to discriminate colours in the learning situation. This is usually the chromatic aspect of the colour but they can learn to attend to the achromatic aspect instead. There is no evidence for relative colour learning, i.e. moths do not learn to choose the longer or shorter of two wavelengths, but it is possible that they learn to choose the darker or brighter shade of a colour, and thereby its relative intensities.  相似文献
