For many commercial potato cultivars, tuber yield is optimal at average daytime temperatures in the range of 14–22 °C. Further rises in ambient temperature can reduce or completely inhibit potato tuber production, with damaging consequences for both producer and consumer. The aim of this study was to use a genetic screen based on a model tuberization assay to identify quantitative trait loci (QTL) associated with enhanced tuber yield. A candidate gene encoding HSc70 was identified within one of the three QTL intervals associated with elevated yield in a Phureja–Tuberosum hybrid diploid potato population (06H1). A particular HSc70 allelic variant was linked to elevated yield in the 06H1 progeny. Expression of this allelic variant was much higher than other alleles, particularly on exposure to moderately elevated temperature. Transient expression of this allele in Nicotiana benthamiana resulted in significantly enhanced tolerance to elevated temperature. An TA repeat element was present in the promoter of this allele, but not in other HSc70 alleles identified in the population. Expression of the HSc70 allelic variant under its native promoter in the potato cultivar Desiree resulted in enhanced HSc70 expression at elevated temperature. This was reflected in greater tolerance to heat stress as determined by improved yield under moderately elevated temperature in a model nodal cutting tuberization system and in plants grown from stem cuttings. Our results identify HSc70 expression level as a significant factor influencing yield stability under moderately elevated temperature and identify specific allelic variants of HSc70 for the induction of thermotolerance via conventional introgression or molecular breeding approaches. 相似文献

Cannabinoid CB1 receptors peripherally modulate energy metabolism. Here, we investigated the role of CB1 receptors in the expression of glucose/pyruvate/tricarboxylic acid (TCA) metabolism in rat abdominal muscle. Dihydrolipoamide dehydrogenase (DLD), a flavoprotein component (E3) of α-ketoacid dehydrogenase complexes with diaphorase activity in mitochondria, was specifically analyzed. After assessing the effectiveness of the CB1 receptor antagonist AM251 (3 mg kg-1, 14 days) on food intake and body weight, we could identified seven key enzymes from either glycolytic pathway or TCA cycle—regulated by both diet and CB1 receptor activity—through comprehensive proteomic approaches involving two-dimensional electrophoresis and MALDI-TOF/LC-ESI trap mass spectrometry. These enzymes were glucose 6-phosphate isomerase (GPI), triosephosphate isomerase (TPI), enolase (Eno3), lactate dehydrogenase (LDHa), glyoxalase-1 (Glo1) and the mitochondrial DLD, whose expressions were modified by AM251 in hypercaloric diet-induced obesity. Specifically, AM251 blocked high-carbohydrate diet (HCD)-induced expression of GPI, TPI, Eno3 and LDHa, suggesting a down-regulation of glucose/pyruvate/lactate pathways under glucose availability. AM251 reversed the HCD-inhibited expression of Glo1 and DLD in the muscle, and the DLD and CB1 receptor expression in the mitochondrial fraction. Objective
To establish the role of liver fibrosis as a predictive tool of response to pegylated interferon alpha (Peg-IFN) and ribavirin (RBV) treatment in human immunodeficiency (HIV)/hepatitis C virus (HCV) coinfected patients, in addition to recognized predictive factors (HCV load, HCV genotype, IL-28B polymorphism).Patients and Methods
A sample of 267 HIV/HCV coinfected patients was treated with Peg-IFN and RBV. Predictive factors of rapid (RVR) and sustained (SVR) virological response were analyzed. Independent variables were age, sex, IL28B, −238 TNF-α and −592 IL-10 polymorphisms, HCV genotype, HCV-RNA levels, significant fibrosis or cirrhosis and CD4+ T cell count.Results
Patients infected by HCV genotype 1 (n = 187) showed RVR and SVR in 12% and 39% of cases, respectively. The parameters associated with RVR were IL28B genotype CC and plasma HCV-RNA levels <600000 IU/ml. Advanced liver fibrosis was negatively associated with SVR in patients without RVR. A SVR was obtained in 42% of subjects with HCV genotype 4, and the independent factors associated with SVR were IL28B genotype CC and an HCV-RNA <600000 IU/ml. A SVR was obtained in 66% of patients with HCV genotypes 2/3; in this case, the independent parameter associated with SVR was the absence of significant liver fibrosis. TNF-α and IL-10 polymorphisms were not associated with SVR, although a significantly higher percentage of −238 TNF-α genotype GG was detected in patients with significant liver fibrosis.Conclusions
In HIV/HCV coinfected patients with HCV genotypes 1 or 4, RVR, mainly influenced by genotype IL28B and HCV-RNA levels, reliably predicted SVR after 4 weeks of therapy with Peg-IFN plus RBV. In patients infected by HCV genotype 3, an elevated relapse rate compromised the influence of RVR on SVR. Relapses were related to the presence of advanced liver fibrosis. The influence of operational conditions (pH, temperature and oxygen transfer rate) on the initial reaction rates of the four reactions involved in the 4S biodesulfurization route of dibenzothiophenes (DBT) has been studied. The bioprocess was carried out using a genetically modified organism, Pseudomonas putida CECT 5279. The rates of the four reactions were calculated from the rates of production of different compounds involved in the 4S pathway, by matrix manipulation. The initial (zero time) reaction rates showed a slight dependence on oxygen transfer rate. Temperature and pH were optimal at 30°C and 9, respectively, temperature being the most important variable. We studied the precipitation of carbonates in 17 strains of moderately halophilic, Gram-positive cocci belonging to two species:
Marinococcus halophilus and Marinococcus albus, isolated from the Salar de Atacama (Chile). They were cultivated in solid and liquid laboratory media for 42 days at salt
concentrations (wt/vol) of 3%, 7.5%, 15%, and 20%. The bioliths precipitated were studied by X-ray diffraction and scanning
electron microscopy. M. halophilus formed crystals at each of the salt concentrations, with a maximum number of strains capable of precipitating carbonates
at 7.5% and 15% salt concentrations. M. albus did not precipitate at 20% and showed a maximum at 7.5%. This behavior is similar to that of other Gram-positive bacteria
and differs from that found in Gram-negative bacteria. The bioliths precipitated were spherical, generally isolated, with
a size of 10–100 μm, varying with salinity. They were of magnesium calcite (CO3 Ca1-x Mgx) with Mg content increasing with increasing salinity and Mg/Ca molar ratio of the culture medium. These results demonstrate
the active role played by M. halophilus and M. albus in the precipitation of carbonates.

Precipitation of minerals was shown by 22 species of moderately halophilic bacteria in both solid and liquid artificial marine salts media at different concentration and different Mg2+-to-Ca2+ ratio. Precipitation of minerals was observed for all the bacteria used. When salt concentration increased, the quantity and the size of bioliths decreased, the time required for precipitation being increased. The precipitated minerals were calcite, magnesian calcite, aragonite, dolomite, monohydrocalcite, hydromagnesite and struvite in variable proportions, depending on the bacterial species, the salinity and the physical state of the medium; the Mg content of the magnesian calcite also varied according to the same parameters. The precipitated minerals do not correspond exactly to those which could be precipitated inorganically according to the saturation indices. Scanning electron microscopy showed that the formation of the bioliths is initiated by grouping of calcified cells and that the dominant final morphologies were spherulitic with fibrous radiated interiors. It was demonstrated that moderately halophilic bacteria play an active role in the precipitation of carbonates and we hypothesize about this process of biomineralization. 相似文献

Colonization and succession over time by bacterivorous protistson laboratory-made marine snow were analysed in five assaysduring 1994. Marine snow was made hom natural seawater usingrolling tanks. In all experiments, the macroaggregates werestable in size and consistency after the fourth day, and thecolonization and succession processes were similar. Newly formedmacre aggregates became colonized by heterotrophic nanoflagellateson the fourth day, mmt of them kinete plastids (Bodo designisand Rhynchomonns nasuta) and bicosoecids (Pseudobodo tremulansand Bicosoeca sp.). Sarcodines and ciliates appeared 1 day later.Among the former, the most abundant genus was Vannella sp.,while scuticociliates (Uronema marinum) and hypotrichs (Euplotesvannus and Aspidisca steini) were the most abundant ciliates.Most of the species observed in the study were more common tobenthic habitats than to pelagic ones. The planktonic existenceof the genera Bodo, Rhynchomonas, Bicosoeca, Euplotes and Aspidiscadepends on the presence of surfaces because they are poor swimmersor immotile, and Pseudobodo and Vannella need attachment forfeeding. The only pelagic protist observed was Uronema, probablybecause its opportunistic behaviour leads it to exploit enrichedenvironments such as marine snow. Flagellate and ciliate abundancesin laboratory-made macroaggregates were much higher than insurrounding water, which indicates that marine snow representsan enhanced habitat for protist growth. 相似文献
