Background

The purpose of this study was to identify incidence and survival patterns in conformational neurodegenerative disorders (CNDDs).

Methods

We identified 2563 reports on the incidence of eight conditions representing sporadic, acquired and genetic, protein-associated, i.e., conformational, NDD groups and age-related macular degeneration (AMD). We selected 245 papers for full-text examination and application of quality criteria. Additionally, data-collection was completed with detailed information from British, Swedish, and Spanish registries on Creutzfeldt-Jakob disease (CJD) forms, amyotrophic lateral sclerosis (ALS), and sporadic rapidly progressing neurodegenerative dementia (sRPNDd). For each condition, age-specific incidence curves, age-adjusted figures, and reported or calculated median survival were plotted and examined.

Findings

Based on 51 valid reported and seven new incidence data sets, nine out of eleven conditions shared specific features. Age-adjusted incidence per million person-years increased from ≤1.5 for sRPNDd, different CJD forms and Huntington''s disease (HD), to 1589 and 2589 for AMD and Alzheimer''s disease (AD) respectively. Age-specific profiles varied from (a) symmetrical, inverted V-shaped curves for low incidences to (b) those increasing with age for late-life sporadic CNDDs and for sRPNDd, with (c) a suggested, intermediate, non-symmetrical inverted V-shape for fronto-temporal dementia and Parkinson''s disease. Frequently, peak age-specific incidences from 20–24 to ≥90 years increased with age at onset and survival. Distinct patterns were seen: for HD, with a low incidence, levelling off at middle age, and long median survival, 20 years; and for sRPNDd which displayed the lowest incidence, increasing with age, and a short median disease duration.

Interpretation

These results call for a unified population view of NDDs, with an age-at-onset-related pattern for acquired and sporadic CNDDs. The pattern linking age at onset to incidence magnitude and survival might be explained by differential pathophysiological mechanisms associated with specific misfolded protein deposits.  相似文献

综述了植物光合作用与温度响应模型研究的进展,围绕光合作用生化模型的4个主要参数:胞间CO2浓度、RuBP最大碳同化速率(Vc max)的活化能、RuBP最大再生速率(Jmax)的活化能和Jmax/Vc max,讨论了影响光合作用 温度响应曲线的内在机理.随着生长温度的升高,所有物种的Vc max活化能均呈增加趋势,而其他参数的变化因物种不同而存在明显差异,说明Vc max的活化能可能是决定光合作用温度依存性的首要参数.最后分析了研究中存在的问题并提出研究展望,认为应整合叶片与群落水平的光合作用模型,从叶面积、太阳辐射、冠层结构、冠层小气候和光合能力等方面研究植物群落对全球变化的响应机理.这对于人们理解和准确估算植物生长、群落碳收支和生态系统初级生产力具有重要意义.  相似文献

1,2,3-triazoles are pharmaceutically significant compounds that have attracted recent interest from medicinal chemists because of their important biological activities. Addressed herein, some 1,2,3-triazoles were synthesized to investigate the inhibitory activities against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes, antioxidant capacity, and antimicrobial effect. The antioxidant profile of 1,2,3-triazoles determined by varied bioanalytical antioxidant methods, including 2,2′-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS.+), 1,1-diphenyl-2-picrylhydrazil (DPPH·), cupric ion (Cu2+) and ferric ion (Fe3+) ascorbic acid, butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT) were used as the standard compounds. In addition, the antibacterial and antifungal activities of these compounds were investigated against seven bacteria and three fungal species using the hollow agar method. As a result of these studies, it was determined that compound 4 showed the best antimicrobial activity and antioxidant activity close to the standards. Inhibitory effects and kinetic studies of these molecules on cholinesterase enzymes were performed. According to the results obtained, compound 4 showed stronger AChE inhibition and compound 3 stronger BChE inhibition compared to other compounds. In kinetic studies, it was found that AChE showed noncompetitive inhibition by compound 4, and BChE showed competitive inhibition by compound 3.  相似文献

Background  

The cattle pathogen, Anaplasma marginale, undergoes a developmental cycle in ticks that begins in gut cells. Transmission to cattle occurs from salivary glands during a second tick feeding. At each site of development two forms of A. marginale (reticulated and dense) occur within a parasitophorous vacuole in the host cell cytoplasm. However, the role of tick genes in pathogen development is unknown. Four genes, found in previous studies to be differentially expressed in Dermacentor variabilis ticks in response to infection with A. marginale, were silenced by RNA interference (RNAi) to determine the effect of silencing on the A. marginale developmental cycle. These four genes encoded for putative glutathione S-transferase (GST), salivary selenoprotein M (SelM), H+ transporting lysosomal vacuolar proton pump (vATPase) and subolesin.  相似文献

Sexes can differ in features associated with differential reproduction, which can be used during courtship or aggressive encounters. Some traits tend to evolve independently between sexes and emerge as sexually dimorphic within the organismal phenotype. We characterize such a relationship by estimating the phenotypic integration of the head morphology and modularity of the crest in the casque‐headed lizards (Corytophanidae). In this clade, some species show extreme sexual dimorphism (e.g., head crests in the genus Basiliscus) while in others, both sexes are monomorphic. To characterize these patterns, we define phenotypic integration at the interspecific level as a pattern or network of traits evidenced by phylogenetically adjusted correlations that persist among species. At this level, modularity is an increased connectedness (e.g., higher correlation) among sections of these networks that persist in a lineage during the evolution of complex phenotypes. To test both concepts, we used phylogenetic geomorphometrics to characterize the head structure of corytophanid lizards, based on a time‐calibrated phylogeny that includes candidate fossil ancestors. We found evidence of an older diversification of corytophanids than previously reported (~67 vs. ~23.5 MYA) and show that this clade includes two morphological head architectures: (1) Sexually dimorphic crests present in males that are evolving independently from the rest of the head structure, and (2) full integration of the head morphology in monomorphic species. We propose that both architectures are optimal evolutionary trajectories of the parietal crest bones in the head of these lizards. In sexually dimorphic species, these bones are elongated and thinner, and gave rise to the extended crest used in male courtship displays. In monomorphic species, the parietal crest grew thicker in both sexes to allow for a better insertion of muscles associated with a stronger bite.  相似文献

Species richness and similarity in metazoan parasite communities of fishes can be influenced by several biotic (age, body size, vagility, feeding and social behavior, among others), and local abiotic (temperature, salinity, etc.) factors. The parasite communities of three species of Oligoplites, marine fishes from the Pacific coast of Mexico, were quantified and analyzed. Four hundred sixty‐eight leatherjackets (O. altus, n=94; O. saurus, n=260; and Orefulgens, n=114) were collected from February 2016 to June 2017 from five locations. Twenty‐eight species of metazoan parasites were recovered and identified: four species of Monogenea (adults), nine of Digenea (seven adults and two metacercariae); two of Cestoda (larvae); four of Nematoda (two adults and two larvae); four of Acanthocephala (two adults, one juvenile, and one cystacanth); four of Copepoda; and one Pentastomida (larvae). At the component community level, species richness ranged from 9 in O. saurus to 19 in O. altus. Different species of helminth dominated the component communities of each species of host. Community composition and species richness of parasites differed among the three species of host, locations, and sampling years. Host feeding behavior, body size, and vagility had the most influence on these differences.  相似文献

The coevolution of ticks and the pathogens that they transmit has ensured their mutual survival. In these studies, we used a functional genomics approach to characterize tick genes regulated in response to Anaplasma marginale infection. Differentially regulated genes/proteins were identified by suppression-subtractive hybridization and differential in-gel electrophoresis analyses of cultured IDE8 tick cells infected with A. marginale. Nine of 17 of these genes were confirmed by real-time RT-PCR to be differentially regulated in ticks and/or IDE8 tick cells in response to A. marginale infection. RNA interference was used for functional studies. Six genes, which encode putative selenoprotein W2a, hematopoietic stem/progenitor cells protein-like, proteasome 26S subunit, ferritin, GST, and subolesin control, were found to affect A. marginale infection in IDE8 tick cells. Four genes, which encode putative GST, salivary selenoprotein M, vATPase, and ubiquitin, affected A. marginale infection in different sites of development in ticks. The results of these studies demonstrated that a molecular mechanism occurs by which tick cell gene expression mediates the A. marginale developmental cycle and trafficking through ticks.  相似文献
