Opening of the aziridine ring of 2-deoxy-1,2-epimino-2-epi-fortimicin B (10) has been effected with both chloride and azide. The reactions are both stereo- and regiospecific and give 2-chloro-2-deoxyfortimicin B (2c) and 2-azido-2-deoxy-fortimicin B (2d). The nucleophilic displacements of the methanesulfonate groups of some 1-N-benzyloxycarbonyl-2-O-(methylsulfonyl)fortimicin derivatives with chloride, azide, and cyanide in N,N-dimethylformamide are dependent both on the nature of the nucleophile and the specific 1-N-benzyloxycarbonyl-2-methanesulfonate employed as the substrate. Striking differences in the stereochemistry of the azide displacements with different 2-methanesulfonates are believed to have a conformational basis. 2-Amino-2-deoxyfortimicin A (1c) and both of the 2-epimeric 2-chloro-2-deoxyfortimicins A (1b) and (5) were prepared for antibacterial assay and the in vitro results are reported.  相似文献

The species' germination response evolves based on its population environment; therefore, the responses of each local population evolve independently. We investigated two xeric species from central Mexico, Echeveria gibbiflora and Penstemon campanulatus, the populations of which inhabit two localities (Reserva Ecológica del Pedregal de San Ángel [REPSA] and Parque Ecológico de la Ciudad de México [PECM]) that differ in environmental conditions. For both species and populations, final germination, cardinal temperatures, thermal time and range of temperature for germination (RTG) were determined in seeds that were (a) collected recently, (b) stored in a laboratory for 2 months and (c) reciprocally buried in field conditions for 2 months. The results indicated that for both species, seed population, laboratory storage and temperature were significant for final germination. These responses indicated differences in germination based primarily on the site in P. campanulatus (PECM seeds germinated at higher percentages than REPSA seeds) and the burial site in the REPSA seeds of E. gibbiflora. Cluster and discriminant analyses were conducted for both species, identifying the following significant variables for group treatments: base temperature between the stored and buried seeds of E. gibbiflora and the ceiling temperature between the recently collected, stored and buried seeds of P. campanulatus. The results suggest that instead of seed population, burial (in both species) and laboratory storage (in P. campanulatus) narrowed the RTG. These responses could indicate plasticity in both germination and dormancy in response to environmental conditions experienced in the different habitats, which are crucial for understanding species' adaptive capacity.  相似文献

Migration inhibitory factor (MIF) was produced by guinea pig lymph node cells stimulated with concanavalin A in the absence and presence of the glycosylation inhibitor tunicamycin. The active supernatants were purified on Sephadex G-100 and fractionated into pH 3-MIF and pH 5-MIF using isoelectrofocusing. When produced in the absence of tunicamycin pH 3-MIF shows extensive charge heterogeneity with activity focusing from pH 3.0 to 4.5; it elutes from Sephadex G-75 with molecules of an apparent MW of 70,000. In contrast, pH 3-MIF produced in the presence of tunicamycin (TM-pH 3-MIF) focuses as a sharp homogeneous peak with a pI of 3.6 to 4.0 and elutes from Sephadex G-75 with molecules of an apparent MW of 25,000–35,000. TM-pH 3-MIF is trypsin sensitive and displays a buoyant density similar to that of proteins which contain little or no carbohydrate (?25 1.26?1.34). Tunicamycin caused no detectable change in the characteristics of pH 5-MIF. This study indicates that lymphocytes stimulated in the presence of tunicamycin produce a novel species of pH 3-MIF with characteristics distinct from classical pH 3-MIF.  相似文献

The correlation between shape and concentration of silver nanoparticles (AgNPs), their cytotoxicity and formation of reactive oxygen species (ROS) in the presence of electromagnetic fields (EMFs) has been investigated. In addition, the bio-effects caused by the combination of EMFs and graphene nanoparticles (GrNPs) have been also assessed. The AgNPs of three shapes (triangular, spherical and colloidal) and GrNPs were added in high concentrations to the culture of human fibroblasts and exposed to EMF of three different frequencies: 900, 2400 and 7500 MHz. The results demonstrated the dependence of the EMF-induced cytotoxicity on the shape and concentration of AgNPs. The maximal cell killing effect was observed at 900 MHz frequency for NPs of all shapes and concentrations. The highest temperature elevation was observed for GrNPs solution irradiated by EMF of 900 MHz frequency. The exposure to EMF led to significant increase of ROS formation in triangular and colloidal AgNPs solutions. However, no impact of EMF on ROS production was detected for spherical AgNPs. GrNPs demonstrated ROS-protective activity that was dependent on their concentration. Our findings indicate the feasibility to control cytotoxicity of AgNPs by means of EMFs. The effect EMF on the biological activity of AgNPs and GrNPs is reported here for the first time.  相似文献

Microbial activities that affect global oceanographic and atmospheric processes happen throughout the water column, yet the long-term ecological dynamics of microbes have been studied largely in the euphotic zone and adjacent seasonally mixed depths. We investigated temporal patterns in the community structure of free-living bacteria, by sampling approximately monthly from 5 m, the deep chlorophyll maximum (∼15–40 m), 150, 500 and 890 m, in San Pedro Channel (maximum depth 900 m, hypoxic below ∼500 m), off the coast of Southern California. Community structure and biodiversity (inverse Simpson index) showed seasonal patterns near the surface and bottom of the water column, but not at intermediate depths. Inverse Simpson''s index was highest in the winter in surface waters and in the spring at 890 m, and varied interannually at all depths. Biodiversity appeared to be driven partially by exchange of microbes between depths and was highest when communities were changing slowly over time. Meanwhile, communities from the surface through 500 m varied interannually. After accounting for seasonality, several environmental parameters co-varied with community structure at the surface and 890 m, but not at the intermediate depths. Abundant and seasonally variable groups included, at 890 m, Nitrospina, Flavobacteria and Marine Group A. Seasonality at 890 m is likely driven by variability in sinking particles, which originate in surface waters, pass transiently through the middle water column and accumulate on the seafloor where they alter the chemical environment. Seasonal subeuphotic groups are likely those whose ecology is strongly influenced by these particles. This surface-to-bottom, decade-long, study identifies seasonality and interannual variability not only of overall community structure, but also of numerous taxonomic groups and near-species level operational taxonomic units.  相似文献

Abstract Inflammatory responses that developed in the footpad during the first 48 h after inoculation of Candida albicans were compared in six genetically defined inbred strains of mice. Tissue responses consisted predominantly of accumulations of polymorphonuclear leucocytes, the magnitude of which was significantly less in mice lacking the fifth component of complement (C5). Despite this, there was no difference between C5-sufficient and C5-deficient mice in the total infectious burden, nor did depletion of complement by treatment with cobra venom factor cause any detectable reduction in the numbers of inflammatory cells in the area of the lesion. Ablation of granulocytes had no significant effect on the fungal burden over the period of the experiment. Immunisation provided some protection against tissue damage, but did not reduce the number of yeasts at the site of infection.  相似文献
