Thyrotropin releasing hormone (TRH) administered both intraventricularly and parenterally suppressed stress induced eating (using the mild tail pinch model) in a dose related manner. This suppression was partially reversed by intraventricular administration of the long acting synthetic enkephalin analog, D-Ala-Met-Enkephalin (p < 0.01). Preliminary data suggests that the naturally occurring metabolic breakdown product of TRH, histidyl-proline-diketopiperazine, may be the active substance with TRH acting as a pro-hormone. The TRH effect was present in hypophysectomized animals showing that the TRH-induced decrease in food ingestion was not secondary to an increase in thyrotropin or thyroid hormones. TRH did not alter blood glucose levels. Our data is compatible with a possible physiological role for TRH, as a peptidergic mediator of satiety acting as a direct antagonist of the lateral hypothalamic of satiety acting as a direct antagonist of the lateral hypothalamic enkephalin-mediated feeding center.  相似文献

In 1976, Kilian and Bulow described the association of beta-glucuronidase with the genus Escherichia (97% positive) and suggested that a beta-glucuronidase assay would be a useful identification test. Since that report, papers about the sensitivity and specificity of this enzyme for the identification of Escherichia coli from clinical sources, food, seawater, potable-water supplies, and various environmental sources have appeared. A study was undertaken to determine the efficacy and specificity of the defined-substrate technology beta-glucuronidase (Colilert) assay for the identification of this species from fecal samples. A total of 460 human, 105 cow, and 55 horse E. coli isolates were tested. Results showed 95.5% beta-glucuronidase-positive isolates in 24 h and 99.5% positive after 28 h of incubation. Only one E. coli isolate was negative. There were no significant differences in the percentage of beta-glucuronidase-positive isolates among the human or animal isolates. There were no non-E. coli isolates that were positive. All subjects carried beta-glucuronidase-positive E. coli.  相似文献

Neonatal pigs were used to develop a surgical biopsy procedure to remove bone tissue from the ilium of small animals, with potential application for infants and small children. While the neonatal pig was under general anaesthesia, a scalpel was used to make a punch incision down to the ilium. Then a Craig Biopsy Trephine was used to remove a core sample of the bone. The samples ranged from 5 to 15 mm in length and 2 to 3 mm in diameter, with an average dry weight of 34.4 mg. The samples were adequate for mineral (calcium and zinc) analysis in our laboratory and may be equally suitable for histological or biochemical analyses. Surgical trauma was minimal, which permitted each pig to be biopsied every 7 days for 5 weeks without adverse consequences.  相似文献

Bruton X-linked agammaglobulinemia (XLA) is a phenotypically recessive genetic disorder of B lymphocyte development. Female carriers of XLA, although asymptomatic, have a characteristic B cell lineage-specific skewing of the pattern of X inactivation. Skewing apparently results from defective growth and maturation of B cell precursors bearing a mutant active X chromosome. In this study, carrier status was tested in 58 women from 22 families referred with a history of agammaglobulinemia. Primary carrier analysis to examine patterns of X inactivation in CD19+ peripheral blood cells (B lymphocytes) was conducted using quantitative PCR at the androgen-receptor locus. Obligate carriers of XLA demonstrated > 95% skewing of X inactivation in peripheral blood CD19+ cells but not in CD19- cells. Carrier status for mothers of isolated affected males could be assessed in 10 of 11 families: 7 women showed skewing, and 3 did not. Five carriers were found in six families in which there were no living affected males. Among all those tested, one individual's carrier status was considered to be indeterminate and five women were noninformative for the carrier test. Results obtained by the carrier test were congruent with linkage analysis (where applicable) using the RFLPs DXS178 and DXS94 and two newly developed polymorphic microsatellite markers, DXS178CA and DXS101AAT. Refinements in techniques for primary carrier testing and genetic mapping of XLA now make possible an ordered approach to diagnosis, prenatal diagnosis, and genetic counseling.  相似文献

Two component systems (TCSs) are a primary mechanism of signal sensing and response in bacteria. Systematic characterization of an entire TCS could provide a mechanistic understanding of these important signal transduction systems. Here, genetic selections were employed to dissect the molecular basis of signal transduction by the HitRS system that detects cell envelope stress in the pathogen Bacillus anthracis. Numerous point mutations were isolated within HitRS, 17 of which were in a 50-residue HAMP domain. Mutational analysis revealed the importance of hydrophobic interactions within the HAMP domain and highlighted its essentiality in TCS signaling. In addition, these data defined residues critical for activities intrinsic to HitRS, uncovered specific interactions among individual domains and between the two signaling proteins, and revealed that phosphotransfer is the rate-limiting step for signal transduction. Furthermore, this study establishes the use of unbiased genetic selections to study TCS signaling and provides a comprehensive mechanistic understanding of an entire TCS.  相似文献

An integrated microscopic (light and electron microscopy) and macroscopic investigation of chambered pith development was made of Phytolacca americana L. Terminal internodes have a solid pith cylinder in contrast to the alternating diaphragms and chambers occurring in subjacent pith. Macroscopically, chambers and diaphragms of any one internode are of equal size. Microscopically, diaphragms vary in height within an internode (from 1–6 cells high). Nevertheless, all diaphragms become thicker circumferentially (5–12 cells high) and connect with long files of intact peripheral pith cells. Diaphragm cells have a large centrally positioned vacuole with a thin, parietal layer of cytoplasm; nuclei, mitochondria, endoplasmic reticulum, and unidentified organelles differentiate in the cytoplasm of diaphragm cells. Although schizogenous activity has most often been implicated as the mechanism by which chambered pith develops in vegetative organs of angiosperms, the results of this study show that cavities in pokeweed result from both schizogenous and lysigenous mechanisms. Schizogeny is suggested by the fact that central pith cells of terminal internodes are longer and thinner walled than peripheral pith cells arranged in vertical files, thus indicating elongation of cells as a possible result of internode elongation. The precise developmental pattern and arrangement of chambers and diaphragms also suggest schizogenous processes. Lysigenous or enzymatic activity is indicated by the fact that cavities are bounded by broken cells, and wall fragments and organelles are often found within enlarging cavities. Chamber formation occurs continuously acropetally and centrifugally in the central pith. A comparison of diaphragms is made with Liriodendron tulipifera and Juglans nigra in an attempt to resolve differences in structure and terminology regarding the differentiation of chambered and diaphragmed pith.  相似文献
