Grapevine fanleaf virus (GFLV) is the major causal agent of the grapevine degeneration disease. To characterize the genomic RNA2 segment from Iranian isolates of GFLV, leaf samples were collected from infected vineyards in different locations with a long history of vine cultivation. Four isolates were selected for cloning and sequencing on the basis of the restriction profiles of RT‐PCR products. The sequencing data revealed that the RNA2 of the Iranian GFLV isolates were the shortest compared with that of all previously described GFLV isolates. The sizes were 3730 nucleotides (nt) for Shir‐Amin and Urmia isolates and 3749 nt for Takestan and Bonab isolates (excluding the poly (A) tail), due to deletion events in both 5′ and 3′ non‐coding regions. In the phylogenetic tree based on the full‐length nucleotide sequences of GFLV RNA2, all the GFLV isolates clustered into two groups with the exception of the Hungarian isolate (GHu). The Iranian isolates grouped as a distinct cluster. Recombination analyses showed that GFLV‐NW (Germany), GFLV‐F13 (reference isolate), GFLV isolate Shir‐Amin (Iran) and Arabis mosaic virus isolate Lv were recombinant isolates and one of their parents belonged to the same lineage as the Iranian isolates. These findings suggest that these isolates originated from a common ancestor.  相似文献

Bacterial resistance is a serious threat to human health. The production of β-lactamase, which inactivates β-lactams is most common cause of resistance to the β-lactam antibiotics. The Class A enzymes are most frequently encountered among the four β-lactamases in the clinic isolates. Mutations in class A β-lactamases play a crucial role in substrate and inhibitor specificity. SHV and TEM type are known to be most common class A β-lactamases. In the present study, we have analyzed the effect of inhibitor resistant S130G point mutation of SHV type Class-A β-lactamase using molecular dynamics and other in silico approaches. Our study involved the use of different in silico methods to investigate the affect of S130G point mutation on the major physico-chemical properties of SHV type class A β-lactamase. We have used molecular dynamics approach to compare the dynamic behaviour of native and S130G mutant form of SHV β-lactamase by analyzing different properties like root mean square deviation (RMSD), H-bond, Radius of gyration (Rg) and RMS fluctuation of mutation. The results clearly suggest notable loss in the stability of S130G mutant that may further lead to decrease in substrate specificity of SHV. Molecular docking further indicates that S130G mutation decreases the binding affinity of all the three inhibitors in clinical practice.  相似文献

An efficient hairy root induction system for an important endangered medicinal plant, Dracocephalum kotschyi, was developed through Agrobacterium rhizogenes-mediated transformation by modifying the co-cultivation medium using five bacterial strains, A4, ATCC15834, LBA9402, MSU440, and A13 (MAFF-02-10266). A drastic increase in transformation frequency was observed when a Murashige and Skoog medium lacking NH4NO3 KH2PO4, KNO3 and CaCl2 was used, resulting in hairy root induction frequencies of 52.3 %, 69.6 %, 48.6 %, 89.0 %, and 80.0 % by A4, A13, LBA9402, MSU440, and ATCC15834 strains, respectively. For shoot induction, hairy roots and unorganized tumors induced by strain ATCC15834 were placed on an MS media supplemented with 0.1, 0.25, 0.5, and 1 mg/l BA plus 0.1 mg/l NAA. The high frequency of shoot regeneration and number of shoot were obtained in the medium containing 0.25 mg/l BA and 0.1 mg/l NAA. Root induction occurred from the base of regenerated shoots on the MS medium supplemented with 0.5 mg/l IBA after 10 days.  相似文献

Pedalium murex (Pedaliaceae), commonly called Large Caltrops, is known for its pharmacological uses in traditional medicine system. It is reported to have excellent medicinal properties that helps cure reproductive disorders, mainly impotency in men, nocturnal emissions, gonorrhea as well as leucorrhoea in women. Apart from that, it also contains remedy for urinary and gastrointestinal tract disorders. Pharmacologically, the plant has been investigated for antiulcerogenic, nephroprotective, hypolipidemic, aphrodisiac, anti-inflammatory, antidermatophytic, antioxidant, antimicrobial and insecticidal activities. The present review is a bundle of information collected from the published research articles and highlights the phytochemical and pharmacological aspects of P. murex. The information will be helpful in developing the new formulation with therapeutic and economical value in the future.  相似文献

New host records for bats are recorded for the first time: N. schmidlii from Rh. clivosus and R. cystops; N. latreillii from Rh. blasii and P. gaisleri; R. huberi from Rh. clivosus; C. pipistrelli from E. isabellinus and H. savii; C. vicinus from E. isabellinus; S. myoti from P. gaisleri; E. euryalis from P. gaisleri and Rh. blasii; A. vespertilionis from P. gaisleri; I. ricinus from T. teniotis and Rh. hipposideros and H. dromedarii from P. kuhlii. The expression of insecticidal genes must be induced at appropriate time and in sufficient amount to confer protection against targeted pests. However, the increased scientific reports of resistance development in insect pest against insecticidal delta-endotoxins, produced by Bacillus thuringiensis, provide impetus for the development of alternative insect management strategies. The present study was conducted to investigate the importance of targeted expression of a hybrid insecticidal gene (SN19) in potatoes. For this purpose, two plant expression vectors were constructed by cloning hybrid SN19 gene (cry1Ba-domain I–III and cry1Ia-domain II) under the control of a wound-inducible promoter isolated from Asparagus officinalis (AoPR1) and CaMV 35S promoter, and were transferred to Agrobacterium tumefaciens strain EHA 105. Four potato genotypes (Marabel, Innovator, Tokat 10/1 and Tokat 6/24) were transformed with EHA 105 strain harboring pTF101.1 35S–SN19 and pTF101.1 AoPR1–SN19 constructs. Phosphinothricin (PPT) was used at concentration of 1 mg/l for selection of primary transformants. PCR results showed the presence of both introduced SN19 and bar genes in 43 plants out of total 154 putative transgenics. Expression of SN19 protein in primary transformants was confirmed by Western blot assays. The mechanical wounding of transgenic plants exhibited more accumulated levels of SN19 proteins during post wounding period. Leaf biotoxicity assays with Colorado potato beetle (Coleoptera) and tomato leafminer (Lepidoptera) exhibited 100% mortality of the pests in primary transformants. Based on our mortality results with both constructs, we concluded that the potato transgenic lines exhibited targeted expression of insecticidal gene under the control of AoPR1 promoter upon insect wounding with eliminated toxicity of Cry protein and hence can be further used effectively in potato breeding programme.  相似文献

The prevalence of methicillin-resistant Staphylococcus aureus and vanomycin intermediate S. aureus infections is on the rise, globally. This poses a huge challenge due to limited therapeutic options and the limited number of bacterial-specific drug targets available for due conservation with the human host. A serine/threonine phosphatase/kinase stp1/stk1 phospho-signalling system in S. aureus, which is just beginning to be understood, has been shown to be of importance in virulence and susceptibility to glycopeptide antibiotics. In this study, 3D structure of stp1 (clinical strain of S. aureus N315) was predicted using a homology modelling tool MODELLER. The validation of the predicted model was done using various tools such as PROCHECK, ERRAT, VERIFY-3D and ProSA. Molecular dynamics (MD) study was carried out using GROMACS to refine the least energy model generated from MODELLER9v11 and it was compared with the template. The template used was the crystal structure of serine/threonine phosphatase stp1 in Streptoccocus agalactiae (Protein Data Bank ID: 2PK0) with 38% identity with the query. Various validation tools showed the quality of the model generated using MODELLER. PROCHECK predicted 100% residues in the allowed region, ERRAT with overall quality factor of 76.47, VERIFY-3D with average score of >0.2 in 81.78% of residues, WHATIF with packaging quality score of > ? 5 for all residues and ProSA with Z-score of ? 7.02. MD simulation of the protein showed some fluctuations in the aqueous environment and changes in the ligand binding residues after simulation. The availability of the 3D-structural information of a viable drug target in S. aureus stp1 is expected to facilitate structure–activity relationship and interactions with proteins.  相似文献

The phonon and thermal properties of different single- ((n,0) (n = 7,8,9,10,14,15)) and double-walled carbon nanotubes ((7, 0) @ (14, 0), (8, 0) @ (14, 0), (9, 0) @ (15, 0) and (10, 0) @ (15, 0),) were calculated using the combination of density functional theory and non-equilibrium Green’s function methods. It was found that the Seebeck and Peltier coefficients for some of the single- and multi-walled carbon nanotubes have negative values. Moreover, in sharp contrast to low ?T, the higher thermoelectric figure of merit is anticipated at the higher temperature. The effect of the atoms number per unit cell on the phonons energies outweighs the effect of the vacuum and the size of the tubes for DWCNTs. All in all, the electron–phonon coupling generates the roughly plethora of thermoelectric coefficients and thermal conductance.  相似文献

The freshwater ciliate Cyrtohymena (Cyrtohymenides) shii (Shi et al., 1997) Shao et al., 2012 (Hypotricha, Oxytrichidae), isolated from Barsey Rhododendron Sanctuary of The Eastern Himalayas, is slightly flexible, measures about 150 μm × 50 μm in life and possesses citrine cortical granules randomly distributed singly and in small clusters. Cells of our Indian population have five or six dorsal kineties arising from multiple fragmentation of the third dorsal anlage. The subgenus Cyrtohymenides includes species with multiple dorsal kinety fragmentation namely C. (C.) aspoecki (type species), C. (C.) australis, and the present species. Ventral morphogenesis of the genus Cyrtohymena has been reported only for the type species C. muscorum. Notable features of the Indian population include formation of frontal anlagen from four parental cirri, two more parental cirri possibly contribute to these anlagen later, and the formation of primary primordia which later split transversely to form two sets, one for each daughter cell. 18S rDNA sequence of the Indian population matches with those of two populations of C. citrina; it also clusters with Afrokeronopsis aurea, a neokeronopsid, with which it interestingly shares some morphological features, supporting the CEUU hypothesis.  相似文献
