 One hundred and thirty nine restriction fragment length polymorphisms (RFLPs) were used to construct a soybean (Glycine max L. Merr.) genetic linkage map and to identify quantitative trait loci (QTLs) associated with resistance to corn earworm (Helicoverpa zea Boddie) in a population of 103 F2-derived lines from a cross of ‘Cobb’ (susceptible) and PI229358 (resistant). The genetic linkage map consisted of 128 markers which converged onto 30 linkage groups covering approximately 1325 cM. There were 11 unlinked markers. The F2-derived lines and the two parents were grown in the field under a plastic mesh cage near Athens, Ga., in 1995. The plants were artificially infested with corn earworm and evaluated for the amount of defoliation. Using interval-mapping analysis for linked markers and single-factor analysis of variance (ANOVA), markers were tested for an association with resistance. One major and two minor QTLs for resistance were identified in this population. The PI229358 allele contributed insect resistance at all three QTLs. The major QTL is linked to the RFLP marker A584 on linkage group (LG) ‘M’ of the USDA/Iowa State University public soybean genetic map. It accounts for 37% of the total variation for resistance in this cross. The minor QTLs are linked to the RFLP markers R249 (LG ‘H’) and Bng047 (LG ‘D1’). These markers explain 16% and 10% of variation, respectively. The heritability (h2) for resistance was estimated as 64% in this population. Supplementary nitrogen (N) side-dressing via the irrigation system is needed in organic cropping. The aim here was to improve N-extraction efficiency, by testing five extraction protocols with guano, layer and broiler manures. The manure-N released by the different methods and manures was mainly in the form of ammonia and ranged from 50% to 85% with no differences among extraction methods. Volatilised ammonia from the extract solution was trapped. At the end of the extraction period, the pH of the extract solution was raised and the rest of the volatilised ammonia was trapped. In the case of guano, about 89% of the manure-N that was mineralised to the extract solution volatilised (after a pH increase), whereas in the layer and broiler manures, 59% and 54% were volatilised, respectively. Extraction of ammonia, its volatilisation and entrapment could provide a significantly more efficient N source than using the extract solution as currently recommended.  相似文献

The goal of this study was to investigate the entrapment of 3 different model proteins (tetanus toxoid, lysozyme, and insulin) into poly(D,L-lactic acid) and poly(D,L-lactic-co-glycolic acid) nanoparticles and to address process-related stability issues. For that purpose, a modified nanoprecipitation method as well as 2 emulsion-based encapsulation techniques (ie, a solid-in oil-in water (s/o/w) and a double emulsion (w1/o/w2) method) were used. The main modification of nanoprecipitation involved the use of a wide range of miscible organic solvents such as dimethylsulfoxide and ethanol instead of the common acetone and water. The results obtained showed that tetanus toxoid and lysozyme were efficiently incorporated by the double emulsion procedure when ethyl acetate was used as solvent (>80% entrapment efficiency), whereas it was necessary to use methylene chloride to achieve high insulin entrapment efficiencies. The use of the s/o/w method or the formation of a more hydrophobic protein-surfactant ion pair did not improve protein loading. The nanoprecipitation method led to a homogenous population of small nanoparticles (with size ranging from ≈130 to 560 nm) and in some cases also improved experimental drug loadings, especially for lysozyme (entrapment efficiency >90%). With respect to drug content determination, a simple and quick matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) method provided results very close to those obtained by reverse phase-high-performance liquid chromatography. With respect to protein stability, the duration and intensity of sonication were not a concern for tetanus toxoid, which retained more than 95% of its antigenicity after treatment for 1 minute. Only a high methylene chloride:water ratio was shown to slightly decrease toxoid antigenicity. Finally, no more than 3.3% of A21 desamido insulin and only traces of covalent insulin dimer were detected in nanoparticles. In conclusion, both the double emulsion and nanoprecipitation methods allowed efficient protein encapsulation. MALDI-TOF MS allowed accurate drug content determination. Responses of the halotolerant yeast Rhodotorula mucilaginosa YRH2 to salt stress was studied. Strain YRH2 was isolated from chemical industry park wastewater evaporation ponds that are characterized by large fluctuations in salinity and pH. Upon shift to high salt medium there is a shutdown of protein synthesis. Radiolabeling and separation of proteins from salt stressed and non-stressed cells identified down-regulated heat shock 70 proteins Ssb1/2p, by N-terminal sequencing and Western blotting. Ssb's role in salt stress in both R. mucilaginosa and S. cerevisiae was examined and we show that its response to salt stress and amino acid limitation is similar. Other proteins such as the heat shock 70 protein Kar2p/BiP and Protein Disulfide Isomerase were strongly induced in response to a shift to high salt in R. mucilaginosa and reacted in a manner similar to the effect of tunicamycin, a known unfolded protein response inducer. Also, assaying carboxypeptidase Y, we showed that high salt medium reduces the specific activity of the enzyme in R. mucilaginosa. It is suggested that the changes in the expression of the heat shock 70 proteins is a part of a mechanism which alleviates the damaging effects of high salt on protein folding in the yeast Rhodotorula mucilaginosa.  相似文献

DNA marker analysis has mapped a quantitative trait locus for soybean, Glycine max (L.) Merr., resistance to the corn earworm, Helicoverpa zea (Boddie) (Lepidoptera: Noctuidae), on the USDA soybean genetic linkage map near the classical gene Pb, which conditions pubescence tip. This study was initiated to determine the effect of pubescence tip on resistance to H. zea larvae and to examine the effect on beet armyworm, Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae), and soybean looper, Pseudoplusia includens (Walker) (Lepidoptera: Noctuidae), larvae. The effect of blunt (pb) and sharp (Pb) pubescence tip was tested in antixenosis and antibiosis bioassays on H. zea, S. exigua, and P. includens larvae with near-isolines and insect-resistant and -susceptible genotypes differing in pubescence tip morphology. Sharp pubescence tip significantly reduced defoliation (antixenosis) from H. zea, S. exigua, and P. includens and weight gain (antibiosis) of H. zea. The weight gain of P. includens was unaffected, and S. exigua weight gain was significant for one pair of near-isolines differing in pubescence tip but not the other. The results indicate that sharp pubescence tip would be beneficial to introgress into elite soybean germplasm due to its association with resistance to H. zea, S. exigua, and P. includens.  相似文献

Effective strategies are needed to manage insect resistance to Bacillus thuringiensis (Bt) proteins expressed in transgenic crops. To evaluate a multiple resistance gene pyramiding strategy, eight soybean (Glycine max) lines possessing factorial combinations of two quantitative trait loci (QTLs) from plant introduction (PI) 229358 and a synthetic Bt cry1Ac gene were developed using marker-assisted selection with simple sequence repeat markers. Field studies were conducted in 2000 and 2001 to evaluate resistance to corn earworm (Helicoverpa zea) and soybean looper (Pseudoplusia includens), and detached leaf bioassays were used to test antibiosis resistance to Bt-resistant and Bt-susceptible strains of tobacco budworm (TBW; Heliothis virescens). Based on defoliation in the field and larval weight gain on detached leaves, lines carrying a combination of cry1Ac and the PI 229358 allele at a QTL on linkage group M were significantly more resistant to the lepidopteran pests, including the Bt-resistant TBW strain, than were the other lines. This is the first report of a complementary additive effect between a Bt transgene and a plant insect resistance QTL with an uncharacterized mode of action that was introgressed using marker-assisted selection.  相似文献

Unlined manure ponds are constructed on clay soil worldwide to manage farm waste. Seepage of ammonia-rich liquor into underlying soil layers contributes to groundwater contamination by nitrate. To identify the possible processes that lead to the production of nitrate from ammonia in this oxygen-limited environment, we studied the diversity and abundance of ammonia-transforming microorganisms under an unlined manure pond. The numbers of ammonia-oxidizing bacteria and anammox bacteria were most abundant in the top of the soil profile and decreased significantly with depth (0.5 m), correlating with soil pore-water ammonia concentrations and soil ammonia concentrations, respectively. On the other hand, the numbers of ammonia-oxidizing archaea were relatively constant throughout the soil profile (10(7) amoA copies per g(soil)). Nitrite-oxidizing bacteria were detected mainly in the top 0.2 m. The results suggest that nitrate accumulation in the vadose zone under the manure pond could be the result of complete aerobic nitrification (ammonia oxidation to nitrate) and could exist as a byproduct of anammox activity. While the majority of the nitrogen was removed within the 0.5-m soil section, possibly by combined anammox and heterotrophic denitrification, a fraction of the produced nitrate leached into the groundwater.  相似文献

Levels of inorganic nitrogen species (ammonia, nitrite, and nitrate), ammonia oxidation potential (AOP), and diversity of ammonia-oxidizing bacteria (AOB) were studied in the sediments of a 50-km-long segment of an ephemeral stream in the Negev desert, receiving untreated wastewater. Water analysis in downstream sampling points showed reductions of 91.7% in biological oxygen demand, 87.7% in chemical oxygen demand, 73.9% in total nitrogen, and 72.8% in total ammonia nitrogen. Significant AOP levels in the sediment were detected mainly in the fall and spring seasons. Denaturing gradient gel electrophoresis of AOB 16S rRNA gene fragments showed that in most sampling points, the streambed was dominated by Nitrosospira cluster 3 strains similar to those dominating the stream bank’s soils and sediments in nearby springs. Nitrosomonas strains introduced by discharged wastewater and others dominated some sections of the stream characterized by high organic carbon levels. The results suggest that climatic conditions in the Negev desert select for AOB belonging to Nitrosospira cluster 3, and these conditions dominate the aquatic environment effect along most of the stream sections. In addition, the nitrification–denitrification processes were not sufficient to reduce nitrogen levels in the sediment and prevent the eutrophication of some sections of the stream ecosystem. Thus, the discharge of high nitrogen wastewater into desert streams should be done carefully as it may endanger the already fragile ecosystem.  相似文献
