This first article of a two‐article series describes a framework and life cycle–based model for typical almond orchard production systems for California, where more than 80% of commercial almonds on the world market are produced. The comprehensive, multiyear, life cycle–based model includes orchard establishment and removal; field operations and inputs; emissions from orchard soils; and transport and utilization of co‐products. These processes are analyzed to yield a life cycle inventory of energy use, greenhouse gas (GHG) emissions, criteria air pollutants, and direct water use from field to factory gate. Results show that 1 kilogram (kg) of raw almonds and associated co‐products of hulls, shells, and woody biomass require 35 megajoules (MJ) of energy and result in 1.6 kg carbon dioxide equivalent (CO2‐eq) of GHG emissions. Nitrogen fertilizer and irrigation water are the dominant causes of both energy use and GHG emissions. Co‐product credits play an important role in estimating the life cycle environmental impacts attributable to almonds alone; using displacement methods results in net energy and emissions of 29 MJ and 0.9 kg CO2‐eq/kg. The largest sources of credits are from orchard biomass and shells used in electricity generation, which are modeled as displacing average California electricity. Using economic allocation methods produces significantly different results; 1 kg of almonds is responsible for 33 MJ of energy and 1.5 kg CO2‐eq emissions. An array of emerging technologies, from electric vehicles to renewable energy systems, relies on large‐format lithium ion batteries (LIBs). LIBs are a critical enabler of clean energy technologies commonly associated with air pollution and greenhouse gas mitigation strategies. However, LIBs require lithium, and expanding the supply of lithium requires new lithium production capacity, which, in turn, changes the environmental impacts associated with lithium production since different resource types and ore qualities will be exploited. A question of interest is whether this will lead to significant changes in the environmental impacts of primary lithium over time. Part one of this two‐part article series describes the development of a novel resource production model that predicts future lithium demand and production characteristics (e.g., timing, location, and ore type). In this article, part two, the forecast is coupled with anticipatory life‐cycle assessment (LCA) modeling to estimate the environmental impacts of producing battery‐grade lithium carbonate equivalent (LCE) each year between 2018 and 2100. The result is a normalized life‐cycle impact intensity for LCE that reflects the changing resource type, quantity, and region of production. Sustained growth in lithium demands through 2100 necessitates extraction of lower grade resources and mineral deposits, especially after 2050. Despite the reliance on lower grade resources and differences in impact intensity for LCE production from each deposit, the LCA results show only small to modest increases in impact, for example, carbon intensity increases from 3.2 kg CO2e/kg LCE in 2020 to 3.3 kg CO2e/kg LCE in 2100. 相似文献

Background
Evidence-based priority setting is increasingly important for rationally distributing scarce health resources and for guiding future health research. We sought to quantify the contribution of a wide range of infectious diseases to the overall infectious disease burden in a high-income setting.Methodology/Principal Findings
We used health-adjusted life years (HALYs), a composite measure comprising premature mortality and reduced functioning due to disease, to estimate the burden of 51 infectious diseases and associated syndromes in Ontario using 2005–2007 data. Deaths were estimated from vital statistics data and disease incidence was estimated from reportable disease, healthcare utilization, and cancer registry data, supplemented by local modeling studies and national and international epidemiologic studies. The 51 infectious agents and associated syndromes accounted for 729 lost HALYs, 44.2 deaths, and 58,987 incident cases per 100,000 population annually. The most burdensome infectious agents were: hepatitis C virus, Streptococcus pneumoniae, Escherichia coli, human papillomavirus, hepatitis B virus, human immunodeficiency virus, Staphylococcus aureus, influenza virus, Clostridium difficile, and rhinovirus. The top five, ten, and 20 pathogens accounted for 46%, 67%, and 75% of the total infectious disease burden, respectively. Marked sex-specific differences in disease burden were observed for some pathogens. The main limitations of this study were the exclusion of certain infectious diseases due to data availability issues, not considering the impact of co-infections and co-morbidity, and the inability to assess the burden of milder infections that do not result in healthcare utilization.Conclusions/Significance
Infectious diseases continue to cause a substantial health burden in high-income settings such as Ontario. Most of this burden is attributable to a relatively small number of infectious agents, for which many effective interventions have been previously identified. Invariant natural killer T (iNKT) cells are activated during infection, but how they limit microbial growth is unknown in most cases. We investigated how iNKT cells suppress intracellular Mycobacterium tuberculosis (Mtb) replication. When co-cultured with infected macrophages, iNKT cell activation, as measured by CD25 upregulation and IFNγ production, was primarily driven by IL-12 and IL-18. In contrast, iNKT cell control of Mtb growth was CD1d-dependent, and did not require IL-12, IL-18, or IFNγ. This demonstrated that conventional activation markers did not correlate with iNKT cell effector function during Mtb infection. iNKT cell control of Mtb replication was also independent of TNF and cell-mediated cytotoxicity. By dissociating cytokine-driven activation and CD1d-restricted effector function, we uncovered a novel mediator of iNKT cell antimicrobial activity: GM-CSF. iNKT cells produced GM-CSF in vitro and in vivo in a CD1d-dependent manner during Mtb infection, and GM-CSF was both necessary and sufficient to control Mtb growth. Here, we have identified GM-CSF production as a novel iNKT cell antimicrobial effector function and uncovered a potential role for GM-CSF in T cell immunity against Mtb. 相似文献

As a result of the genetic selection of broiler (meat-type breeders) chickens for enhanced growth rate and lower feed conversion ratio, it has become necessary to restrict feed intake. When broilers are fed ad libitum, they would become obese and suffer from several health-related problems. A vital adaptation to starvation is autophagy, a self-eating mechanism for recycling cellular constituents. The autophagy pathway has witnessed dramatic growth in the last few years and extensively studied in yeast and mammals however, there is a paucity of information in avian (non-mammalian) species. Here we characterized several genes involved in autophagosome initiation and elongation in Red Jungle fowl (Gallus gallus) and Japanese quail (coturnix coturnix Japonica). Both complexes are ubiquitously expressed in chicken and quail tissues (liver, leg and breast muscle, brain, gizzard, intestine, heart, lung, kidney, adipose tissue, ovary and testis). Alignment analysis showed high similarity (50.7 to 91.5%) between chicken autophagy-related genes and their mammalian orthologs. Phylogenetic analysis demonstrated that the evolutionary relationship between autophagy genes is consistent with the consensus view of vertebrate evolution. Interestingly, the expression of autophagy-related genes is tissue- and gender- dependent. Furthermore, using two experimental male quail lines divergently selected over 40 generations for low (resistant, R) or high (sensitive, S) stress response, we found that the expression of most studied genes are higher in R compared to S line. Together our results indicate that the autophagy pathway is a key molecular signature exhibited gender specific differences and likely plays an important role in response to stress in avian species. 相似文献

Abstract. Ultrastructural characteristics of the epidermis of 5 deep-sea vestimentiferan tube worms and an orbiniid worm were similar to those described from other organisms inhabiting sulfidic environments. The integument was composed of a simple or pseudostratified layer of columnar or cuboidal cells, covered by a cuticle of varying thickness, and rested on an extracellular matrix. Unusual mitochondrial morphology was observed in the peripheral region of the epidermis, characterized by an electron-dense matrix and granules. The abnormal size, shape, and reduced array of cristae in these mitochondria may have resulted from environmental stress. Mitochondria in the inner epidermal layers, where they are more protected from sulfide exposure, displayed normal morphology. The seep vestimentiferans ( Seepiophila jonesi and Lamellibrachia cf. luymesi ) exhibited numerous electron-dense organelles, similar in morphology to cytolysosomes and previously described as sulfide oxidizing bodies, while in the 3 vent vestimentiferans ( Riftia pachyptila, Tevnia jerichonana , and Oasisia alvinae ) and the seep orbiniid Methanoaricia dendrobranchiata , fewer of these organelles were observed. Energy dispersive x-ray microanalysis of these electron-dense organelles indicated the presence of sulfur, iron, zinc, and copper, as well as lesser amounts of other elements. We propose that these epidermal cytolysosomes are morphologically similar to previously described sulfide oxidizing bodies that have been hypothesized to play a role in maintaining aerobically poised animal metabolism in sulfide-enriched habitats. 相似文献

BACKGROUND: Although thyroid disorders are present in approximately 3% of pregnant women, little is known about the association between maternal thyroid disease and birth defects. METHODS: We assessed the association between maternal thyroid disease, thyroid medication use, and 38 types of birth defects among 14,067 cases and 5875 controls in the National Birth Defects Prevention Study, a multisite, population‐based, case‐control study. Infants in this study were born between October 1997 and December 2004. Information on exposures including maternal diseases and use of medications was collected by telephone interview. RESULTS: We found statistically significant associations between maternal thyroid disease and left ventricular outflow tract obstruction heart defects (1.5; 95% CI, 1.0–2.3), hydrocephaly (2.9; 95% CI, 1.6–5.2), hypospadias (1.6; 95% CI, 1.0–2.5), and isolated anorectal atresia (2.4; 95% CI, 1.2–4.6). Estimates for the association between periconceptional use of thyroxine and specific types of birth defects were similar to estimates for any thyroid disease. Given that antithyroid medication use was rare, we could not adequately assess risks for their use for most case groups. CONCLUSIONS: Our results are consistent with the positive associations between maternal thyroid disease or thyroid medication use and both hydrocephaly and hypospadias observed in some previous studies.
