Lymphotoxin-α (LTA) is a pro-inflammatory cytokine that plays an important role in the immune system and local inflammatory response. LTA is expressed in atherosclerotic plaques and has been implicated in the pathogenesis of atherosclerosis and coronary heart disease (CHD). Polymorphisms in the gene encoding lymphotoxin-α (LTA) on Chromosome 6p21 have been associated with susceptibility to CHD, but results in different studies appear to be conflicting. We examined the association of seven single nucleotide polymorphisms (SNPs) across the LTA gene, and their related haplotypes, with risk of myocardial infarction (MI) in the International Study of Infarct Survival (ISIS) case-control study involving 6,928 non-fatal MI cases and 2,712 unrelated controls. The seven SNPs (including the rs909253 and rs1041981 SNPs previously implicated in the risk of CHD) were in strong linkage disequilibrium with each other and contributed to six common haplotypes. Some of the haplotypes for LTA were associated with higher plasma concentrations of C-reactive protein (p = 0.004) and lower concentrations of albumin (p = 0.023). However, none of the SNPs or related haplotypes were significantly associated with risk of MI. The results of the ISIS study were considered in the context of six previously published studies that had assessed this association, and this meta-analysis found no significant association with CHD risk using a recessive model and only a modest association using a dominant model (with narrow confidence intervals around these risk estimates). Overall, these studies provide reliable evidence that these common polymorphisms for the LTA gene are not strongly associated with susceptibility to coronary disease. 相似文献

Polymer networks are based on molecules which are covalently or physically connected in a three-dimensional network. In presence of an appropriate solvent these networks swell by solvent absorption to form gels. These gels, which are called hydrogels in case of water absorption, are able to change their volume by more than a hundred-fold. During the swelling or shrinking process the hydrogels perform a mechanical work. Their volume standardized working capacity can be ten-times larger than that of an electromagnet. Due to their simple design, miniaturisation properties, and their ability to realize many automatic sensor and actuator functions, smart hydrogels offer new solutions in biomedical technology. 相似文献

Summary The Australian irrigation industry diverts significant volumes of water from our rivers, and as such, may also divert and entrain riverine fish. Although it is widely acknowledged that our native fish fauna have been greatly affected by a variety of anthropogenic changes, little is known about the extent or significance of fish entrainment in irrigation systems. This paper presents results from a preliminary investigation into the diversion of fish into irrigation channels undertaken in the Goulburn-Murray Irrigation Network, Victoria, Australia. The case study and our knowledge of the life-history strategies of the Basin's fish fauna suggest that the loss of native fish into irrigation networks may be a substantial problem, which, up until recently, has been largely ignored. We strongly suggest that the impact of diversions on native fish populations requires urgent further investigation and quantification, through the cooperation of water management authorities and fish biologists. 相似文献

Exposure to air pollutants such as ozone (O(3)) induces airway hyperresponsiveness (AHR) and airway inflammation. Toll-like receptors (TLR) are first-line effector molecules in innate immunity to infections and signal via adapter proteins, including myeloid differentiation factor-88 (MyD88). We investigated the sensing of ozone by TLR2, TLR4, and MyD88. Ozone induced AHR in wild-type (WT) C57BL/6 mice, but AHR was absent in TLR2(-/-), TLR4(-/-), and MyD88(-/-) mice. Bronchoalveolar lavage neutrophilia induced by ozone was inhibited at 3 h but not at 24 h in TLR2(-/-) and TLR4(-/-) mice, while in MyD88(-/-) mice, this was inhibited at 24 h. We investigated the expression of inflammatory cytokines and TLR2, TLR4, and MyD88 in these mice. Ozone induced time-dependent increases in inflammatory gene expression of keratinocyte chemoattractant (KC) and IL-6 and of TLR2, TLR4, and MyD88 in WT mice. IL-6 and KC expression induced by ozone was inhibited in TLR2(-/-), TLR4(-/-), and MyD88(-/-) mice. Expression of MyD88 was increased in TLR2(-/-) and TLR4(-/-) mice, while induction of TLR2 or TLR4 was reduced in TLR2(-/-) and TLR4(-/-) mice, respectively. TLR2 and TLR4 mediate AHR induced by oxidative stress such as ozone, while the adapter protein MyD88, but not TLR2 or TLR4, is important in mediating ozone-induced neutrophilia. TLR2 and TLR4 may also be important in regulating the speed of neutrophilic response. Therefore, ozone may induce murine AHR and neutrophilic inflammation through the activation of the Toll-like receptor pathway that may sense noninfectious stimuli such as oxidative stress. 相似文献

Hurricanes frequently affect the forests of South and Central America; however, few studies have quantified their effects to forest structure, especially when concentrating on the food supply of an animal population. Hurricane Iris made landfall in Southern Belize on 8 October 2001, severely damaging a 52 hectare site where the behavioral ecology of a population of Central American Black Howlers (Alouatta pigra) had been under study for 2.5 yr. The hurricane resulted in a mortality rate of 35 percent for major food trees, which was primarily attributed to uprooting, snapping, and major delimbing. This damage accounted for 97 percent of the food tree loss between the two sample periods. Tree species differences were found in both the percentage loss and category of damage to food trees. Trees of different heights also experienced different percentage loss and levels of damage; subcanopy and emergent trees experienced higher loss than canopy trees, and subcanopy trees were frequently uprooted. This was partially attributed to a lack of buttressing on these subcanopy trees. Buttressing was found to decrease the frequency of uprooting. Tree size was the only factor that did not influence either damage or death. Trees from which fruit were eaten by black howlers died more than twice as often as did trees eaten for leaves. 相似文献

We previouslyshowed that increased macrophage andPGE2 production with age is due toenhanced cyclooxygenase (COX) activity and COX-2 expression. This studydetermined the effect of vitamin E supplementation on macrophagePGE2 synthesis in young and old mice and its underlying mechanism. Mice were fed 30 or 500 parts permillion vitamin E for 30 days. Lipopolysaccharide (LPS)-stimulated macrophages from old mice produced significantly morePGE2 than those from young mice.Vitamin E supplementation reversed the increasedPGE2 production in old mice buthad no effect on macrophage PGE2production in young mice. In both LPS-stimulated and unstimulated macrophages, COX activity was significantly higher in old than in youngmice at all intervals. Vitamin E supplementation completely reversedthe increased COX activity in old mice to levels comparable to those ofyoung mice but had no effect on macrophage COX activity of young miceor on COX-1 and COX-2 protein or COX-2 mRNA expression in young or oldmice. Thus vitamin E reverses the age-associated increase in macrophagePGE2 production and COX activity.Vitamin E exerts its effect posttranslationally, by inhibiting COXactivity. 相似文献
